Morocco is rich in temporary pools which harbour numerous rare plant species. Long-term conservation of such threatened plant communities should be based on the understanding of their past dynamics. Despite conditions unfavourable to pollen preservation, surface sediments of acidic temporary pools are shown to contain pollen assemblages likely to allow vegetation reconstruction. Knowledge of the modern relationships between pollen and vegetation is, however, necessary for interpreting fossil data in terms of past vegetation. Surface pollen assemblages and floristic surveys of a temporary pool in Benslimane forest, western Morocco, are compared in order to evaluate the pollen record of the local hydrophytic vegetation. Floristic surveys were carried out for 12 years (1996-2008) along two crossing permanent transects. A set of 21 surface-sediment samples, taken along the same transects in 2007, were analysed for pollen. The spatial relationships between vegetation and pollen assemblages are explored by means of multivariate analyses, statistical tests and linear regressions. The calculation of representation indices moreover allows proposing quantitative ways for pollen-based plant-abundance reconstruction.Results reveal that the vegetation structure along the hydrological gradient is well recorded in the pollen assemblages, with: (1) a marginal zone characterised by terrestrial taxa and rare amphibious taxa (Elatine, Pilularia), (2) an intermediate zone of amphibious taxa (Alisma-type, Illecebrum/Paronychia, Isoetes velata-type), and (3) a central zone of aquatics (Myriophyllum alterniflorum, Ranunculus-type). The best correlation between the pollen record and total pool vegetation was found in the centre of the pool, which supports the reliability of the study of a single core from the centre of the pool for the reconstruction of the past dynamics of the local hydrophytic vegetation. Both the qualitative ‘community’ approach (representation indices and indicator pollen taxa) and the quantitative ‘taxa’ approach (correction factors) suggest that reconstructions of past populations can be achieved from a few taxa, namely Isoetes velata-type, Myriophyllum alterniflorum and Ranunculus-type. For these taxa, regression parameters (slope and y-intercept) have been calculated between pollen percentages and plant percentages in present vegetation, and between pollen influxes and plant abundances, respectively. These parameters can be extended to interpret fossil data from other temporary pools within the same region to reconstruct their relative and absolute past plant abundances.  相似文献

Seventy-five V regions encoded by the sequenced genome of one Macaca mulatta specimen have been identified by homology and paired with similar human counterparts. When the human V region of each pair presented no allelic polymorphism, it was directly compared with its homolog. This was the case for 37 pairs and percents of identity ranged between 84–97%. When the human V region presented allelic polymorphism, this polymorphism was found to be significantly smaller (p < 0.0001, p < 0.0001, p = 0.03 for IGHV, IGLV, IGKV regions respectively), 4.2-fold on average, than the differences observed between human and macaque V regions. Similar results were obtained when analyzing framework regions (FRs) only. These results, in agreement with others, demonstrate the existence of differences between human and macaque V regions, confirm the need for the humanization of macaque V regions intended for therapeutic use and call into question the validity of patents relying on the “undistinguishable” character of human and macaque V regions or FRs.Key words: antibody, therapeutic, non-human primate, human, V region, identity, humanization, patent  相似文献

Gold nanorods (GNR) within tumor microregions are characterized by their ability to absorb near IR light and emit heat in what is called photoplasmonic effect. Yet, the efficacy of nanoparticles is limited due to intratumoral tissue distribution reasons. In addition, distribution of GNRs to normal tissue might result in non specific toxicity. In the current study, we are assessing the intratumoral and tissue distribution of PEGylated GNRs on the top of its antitumor characteristics when given intravenously or intratumoral to solid tumor bearing mice and coupled with laser photoplasmonic sessions. PEGylated GNRs with a longitudinal size of less than 100 nm were prepared with aspect ratio of 4.6 showing strong surface plasmon absorption at wavelength 800 nm. Pharmacokinetics of GNR after single I.V. administration (0.1 mg/kg) showed very short systemic circulating time (less than 3 h). On the other hand, tissue distribution of I.V. GNR (0.1 mg/kg) to normal animals showed preferential deposition in spleen tissue. Repeated administration of I.V. GNR resulted in preferential accumulation in both liver and spleen tissues. In addition, I.V. administration of GNR to Ehrlich carcinoma tumor bearing mice resulted in similar tissue distribution; tumor accumulation and anti-tumor effect compared to intratumoral administration. In conclusion, the concentration of GNR achieved within tumors microregions after I.V. administration was comparable to I.T. administration and sufficient to elicit tumoral growth arrest when coupled with laser-aided photoplasmonic treatment.  相似文献

BackgroundBotulinum neurotoxins (BoNTs) are considered to be the most toxic substances known on earth and are responsible for human botulism, a life-threatening disease characterized by flaccid muscle paralysis that occurs naturally by food-poisoning or colonization of the gastrointestinal tract by BoNT-producing clostridia. BoNTs have been classified as category A agent by the Centers of Disease Control and Prevention (CDC) and are listed among the six agents with the highest risk to be used as bioweapons. Neutralizing antibodies are required for the development of effective anti-botulism therapies to deal with the potential risk of exposure.ResultsIn this study, a macaque (Macaca fascicularis) was immunized with recombinant light chain of BoNT/E3 and an immune phage display library was constructed. After a multi-step panning, several antibody fragments (scFv, single chain fragment variable) with nanomolar affinities were isolated, that inhibited the endopeptidase activity of pure BoNT/E3 in vitro by targeting its light chain. Furthermore, three scFv were confirmed to neutralize BoNT/E3 induced paralysis in an ex vivo mouse phrenic nerve-hemidiaphragm assay. The most effective neutralization (20LD50/mL, BoNT/E3) was observed with scFv ELC18, with a minimum neutralizing concentration at 0.3 nM. Furthermore, ELC18 was highly effective in vivo when administered as an scFv-Fc construct. Complete protection of 1LD50 BoNT/E3 was observed with 1.6 ng/dose in the mouse flaccid paralysis assay.ConclusionThese scFv-Fcs antibodies are the first recombinant antibodies neutralizing BoNT/E by targeting its light chain. The human-like nature of the isolated antibodies is predicting a good tolerance for further clinical development.  相似文献

Mowat-Wilson syndrome is a mental retardation-multiple congenital anomaly syndrome characterized by a typical facies, developmental delay, epilepsy, and variable congenital malformations, including Hirschsprung disease, urogenital anomalies, congenital heart disease, and agenesis of the corpus callosum. This disorder is sporadic and is caused by heterozygous mutations or deletions of the ZFHX1B gene located in the 2q22 region. We report here the first Moroccan patient, born to consanguineous parents, with Mowat-Wilson syndrome, due to a de novo, unreported mutation of the ZFHX1B gene.  相似文献

Aldrex T enhanced the mean number of groundnut nodules due to in vitro application at a higher than field concentration (6 g/kg groundnut kernels). Dressing at lower and agricultural applications (1.5 and 3, respectively) not only depressed but also considerably delayed nodule formation. This feature was demonstrated by the great losses in nodule number and weight as well. The latter picked up after 11 weeks to equate that of the controls. Interference of the fungicide in nodulation was highly significant at the sixth week of sowing. An insignificant increase in the number of root nodules was recorded at the tenth week of plant growth thus signifying recovery ofRhizobium from aldrex sensitivity. Dressing at 6 g/kg seeds did not affect the number and weight of nodules particularly after the fourth week of groundnut growth. Subsequently a highly significant increase in nodule weight was noticed.Nitrate and nitrite yield of nodules was favourably affected when kernels received twice the agricultural dose when compared with the non-dressed controls.  相似文献
