The ongoing outbreak of severe acute respiratory syndrome (SARS) coronavirus 2 (SARS‐CoV‐2) demonstrates the continuous threat of emerging coronaviruses (CoVs) to public health. SARS‐CoV‐2 and SARS‐CoV share an otherwise non‐conserved part of non‐structural protein 3 (Nsp3), therefore named as “SARS‐unique domain” (SUD). We previously found a yeast‐2‐hybrid screen interaction of the SARS‐CoV SUD with human poly(A)‐binding protein (PABP)‐interacting protein 1 (Paip1), a stimulator of protein translation. Here, we validate SARS‐CoV SUD:Paip1 interaction by size‐exclusion chromatography, split‐yellow fluorescent protein, and co‐immunoprecipitation assays, and confirm such interaction also between the corresponding domain of SARS‐CoV‐2 and Paip1. The three‐dimensional structure of the N‐terminal domain of SARS‐CoV SUD (“macrodomain II”, Mac2) in complex with the middle domain of Paip1, determined by X‐ray crystallography and small‐angle X‐ray scattering, provides insights into the structural determinants of the complex formation. In cellulo, SUD enhances synthesis of viral but not host proteins via binding to Paip1 in pBAC‐SARS‐CoV replicon‐transfected cells. We propose a possible mechanism for stimulation of viral translation by the SUD of SARS‐CoV and SARS‐CoV‐2.  相似文献

  • Floral nectar is considered the most important floral reward for attracting pollinators. It contains large amounts of carbohydrates besides variable concentrations of amino acids and thus represents an important food source for many pollinators. Its nutrient content and composition can, however, strongly vary within and between plant species. The factors driving this variation in nectar quality are still largely unclear.
  • We investigated factors underlying interspecific variation in macronutrient composition of floral nectar in 34 different grassland plant species. Specifically, we tested for correlations between the phylogenetic relatedness and morphology of plants and the carbohydrate (C) and total amino acid (AA) composition and C:AA ratios of nectar.
  • We found that compositions of carbohydrates and (essential) amino acids as well as C:AA ratios in nectar varied significantly within and between plant species. They showed no clear phylogenetic signal. Moreover, variation in carbohydrate composition was related to family-specific structural characteristics and combinations of morphological traits. Plants with nectar-exposing flowers, bowl- or parabolic-shaped flowers, as often found in the Apiaceae and Asteraceae, had nectar with higher proportions of hexoses, indicating a selective pressure to decelerate evaporation by increasing nectar osmolality.
  • Our study suggests that variation in nectar nutrient composition is, among others, affected by family-specific combinations of morphological traits. However, even within species, variation in nectar quality is high. As nectar quality can strongly affect visitation patterns of pollinators and thus pollination success, this intra- and interspecific variation requires more studies to fully elucidate the underlying causes and the consequences for pollinator behaviour.

Premise

Many flowering plants depend on insects for pollination and thus attract pollinators by offering rewards, mostly nectar and pollen. Bee pollinators rely on pollen as their main nutrient source. Pollen provides all essential micro- and macronutrients including substances that cannot be synthesized by bees themselves, such as sterols, which bees need for processes such as hormone production. Variations in sterol concentrations may consequently affect bee health and reproductive fitness. We therefore hypothesized that (1) these variations in pollen sterols affect longevity and reproduction in bumble bees and (2) can thus be perceived via the bees' antennae before consumption.

Methods

We studied the effect of sterols on longevity and reproduction of Bombus terrestris workers in feeding experiments and investigated sterol perception using chemotactile proboscis extension response (PER) conditioning.

Results

Workers could perceive several sterols (cholesterol, cholestenone, desmosterol, stigmasterol, β-sitosterol) via their antennae but not differentiate between them. However, when sterols were presented in pollen, and not as a single compound, the bees were unable to differentiate between pollen differing in sterol content. Additionally, different sterol concentrations in pollen neither affected pollen consumption nor brood development or worker longevity.

Conclusions

Since we used both natural concentrations and concentrations higher than those found in pollen, our results indicate that bumble bees may not need to pay specific attention to pollen sterol content beyond a specific threshold. Naturally encountered concentrations might fully support their sterol requirements and higher concentrations do not seem to have negative effects.

Zusammenfassung Die Tanycyten am Abgang des Recessus infundibularis des Kaninchen-Gehirns unterscheiden sich von anderen Ependymzellen und von Astrocyten. Für ihre Stoff-transportfunktion sprechen ihre membranreichen Insertionen an stark vergößerten Flächen der perivasculären Basalmembran, ihr Gehalt an vielen großen Mitochondrien bei fehlendem Ergastoplasma sowie ein großer Golgiapparat, der in allen Teilen des Fortsatzes vorkommt.
Summary The tanycytes in the wall of the III. ventriculus near the entrance into the recessus infundibularis can be distinguished from astrocytes and other ependymal cells. The ultrastructure of tanycytes indicates that these cells are able to transport material. There are many large mitochondria, while ergastoplasma is almost absent. A greatly enlarged Golgi-apparatus is found in all parts of the cell processes. The tanycytes reach the perivascular basement membrane with many small processes which increase the contact with the perivascular basement membrane having extensions into the surrounding tissue.

Summary Using light microscopic immunohistochemistry, somatostatinpositive structures were observed in the cortex of the rat. These structures, including cells and fibers, are widely distributed in all cortical laminae and are also found in the basal ganglia. The positive results were obtained exclusively in two groups of animals sacrificed during two different months of two subsequent years. Adipose tissue biopsy and an oral glucose tolerance test (OGTT) (50 g) were performed in 17 non-hyperlipoproteinemic subjects without overt diabetes mellitus. All the persons were weight stable at the time of investigation. A significant correlation between fasting insulin concentration and the mean adipocyte size was observed, whereas no correlation was noted between the ideal body weight index and fasting insulin level. Persons with larger adipocytes had elevated insulin levels as well as higher and longer lasting increments following the glucose challenge. They also exhibited significantly higher mean glucose levels during the OGTT. When these patients were matched for glucose tolerance with the subgroup having smaller mean adipocyte sizes, the difference in insulin levels was still demonstrable. The importance of adipose cell enlargement regulating basal and stimulated insulin output is underlined.  相似文献

Summary With the aid of light- and electron- microscopic immunocytochemistry, somatostatin- and luliberin (LRF)-positive fibers can be demonstrated in the rat subfornical organ (SFO). Each of the neurohormones has a specific location: LRF in the lateral parts of the organ, and somatostatin in the center of the posterior zone. Common to both neurohormone-containing fibers is the pattern in which they reach the organ as well as the fact that their terminals are located in the perivascular spaces of fenestrated vessels, i.e., within the limited neurohemal regions of the organ. Since injection of India ink of different colors demonstrates that the capillary bed of the SFO is connected with the central capillaries of the choroid plexus, the question arises as to whether the neurohormones released in the area of the SFO influence the choroid plexus.Supported by the Deutsche Forschungsgemeinschaft (Grant Nr.
