Two new tetracyclic triterpenoids zafaral [24,25,26,27-tetranorapotirucalla-(apoeupha)-6alpha-methoxy-7alpha-acetoxy-1,14-dien-3,16-dione-21-al] (1) and meliacinanhydride [24,25,26,27-tetranorapotirucalla-(apoeupha)-6alpha-hydroxy,11alpha-methoxy-7alpha,12alpha-diacetoxy,1,14,20(22)-trien-3-one] (2) have been isolated from the methanolic extract of neem leaves along with two known constituents nimocinol and isomeldenin. Their structures and the relative configurations were determined by spectroscopic methods ((1)H and (13)C NMR, IR, and MS) and 2D NMR experiments.  相似文献

The soils of the Pampas are thought to be generally non-contaminated but there is growing evidence of trace element accumulation at some specific sites. The goal of this study was to measure the current levels of the main Potentially Toxic Elements (PTE) in the top horizon and in specific soil profiles so that we would establish the baseline concentrations of these elements. Eighty-eight top soils and three soil profiles were sampled. The samples were acid digested. Arsenic, boron, barium, cadmium, cobalt, chromium, copper, lead, manganese, mercury, molybdenum, nickel, silver, selenium and zinc were determined with inductively coupled argon plasma emission spectrometry (ICPES).

All of the values found are within the normal range for uncontaminated soils as reported from several continents. Elements with high environmental risk potential are lower than the admissible range of the European Union and some of them are orders of magnitude lower than those of the United States Environmental Protection Agency (US-EPA) 501 levels. Potentially Toxic Elements contents increased with depth or showed a maximum concentration at the B2 horizon. This is related to the parent material and the pedogenetic processes but not to recent contamination. Soil profiles showed higher concentrations of PTE in clayey horizons. However, these relationships did not appear in top soil samples in any soil Great Group studied. The shown data establishes a baseline for PTE concentrations for Pampas soils.  相似文献

Glasshouse experiments were conducted to assess the influence of Pseudomonas fluorescens, Azotobacter chroococcum, Azospirillum brasilense and composted organic fertilizers (cow dung, horse dung, goat dung and poultry manure) alone and in combination on the multiplication of Meloidogyne incognita and growth of tomato. P. fluorescens was better at improving tomato growth and reducing galling and nematode multiplication than A. chroococcum or A. brasilense. Among composted organic fertilizers, poultry manure resulted in less galling and nematode multiplication than occurred with goat dung. However, composted goat dung was better in reducing nematode multiplication and improving plant growth than horse dung. Cow dung was the composted organic fertilizer least effective in reducing galling and nematode multiplication. Poultry manure with P. fluorescens was the best combination for the management of M. incognita on tomato but improved management of M. incognita can also be obtained if goat dung is used with P. fluorescens or poultry manure with A. chroococcum.  相似文献

Betulinic acid (1) significantly caused cytotoxicity to endothelial cell line ECV304 (IC(50) 1.26+/-0.44 microg/mL) in a 5-day MTT assay. Novel and more potent derivatives of betulinic acid (2, 4, 6-8) have been synthesized with IC(50) less than 0.4 microg/mL. The endothelial cell specificity against human tumor cell lines DU145, L132, A549, and PA-1 were determined. Further betulinic acid (1) inhibited TLS formation of ECV304 cells on Matrigel(TM) by 5.5% while its derivatives caused an inhibition of 13.1-49.2%.  相似文献

ClpX and related AAA+ ATPases of the Clp/Hsp100 family are able to denature native proteins. Here, we explore the role of protein stability in ClpX denaturation and subsequent ClpP degradation of model substrates bearing ssrA degradation tags at different positions. ClpXP degraded T. thermophilus RNase-H* with a C-terminal ssrA tag very efficiently, despite the very high global stability of this thermophilic protein. In fact, global thermodynamic stability appears to play little role in susceptibility to degradation, as a far less stable RNase-H*-ssrA mutant was degraded more slowly than wild type by ClpXP and a completely unfolded mutant variant was degraded less than twice as fast as the wild-type parent. When ssrA peptide tags were covalently linked to surface cysteines at positions 114 or 140 of RNase-H*, the conjugates were proteolyzed very slowly. This resistance to degradation was not caused by inaccessibility of the ssrA tag or an inability of ClpXP to degrade proteins with side-chain linked ssrA tags. Our results support a model in which ClpX denatures proteins by initially unfolding structural elements attached to the degradation tag, suggest an important role for the position of the degradation tag and direction of force application, and correlate well with the mapping of local protein stability within RNase-H* by native-state hydrogen exchange.  相似文献

The 3Dee database of domain definitions was developed as a comprehensive collection of domain definitions for all three-dimensional structures in the Protein Data Bank (PDB). The database includes definitions for complex, multiple-segment and multiple-chain domains as well as simple sequential domains, organized in a structural hierarchy. Two different snapshots of the 3Dee database were analyzed at September 1996 and November 1999. For the November 1999 release, 7,995 PDB entries contained 13,767 protein chains and gave rise to 18,896 domains. The domain sequences clustered into 1,715 domain sequence families, which were further clustered into a conservative 1,199 domain structure families (families with similar folds). The proportion of different domain structure families per domain sequence family increases from 84% for domains 1-100 residues long to 100% for domains greater than 600 residues. This is in keeping with the idea that longer chains will have more alternative folds available to them. Of the representative domains from the domain sequence families, 49% are in the range of 51-150 residues, whereas 64% of the representative chains over 200 residues have more than 1 domain. Of the representative chains, 8.5% are part of multichain domains. The largest multichain domain in the database has 14 chains and 1,400 residues, whereas the largest single-chain domain has 907 residues. The largest number of domains found in a protein is 13. The analysis shows that over the history of the PDB, new domain folds have been discovered at a slower rate than by random selection of all known folds. Between 1992 and 1997, a constant 1 in 11 new domains deposited in the PDB has shown no sequence similarity to a previously known domain sequence family, and only 1 in 15 new domain structures has had a fold that has not been seen previously. A comparison of the September 1996 release of 3Dee to the Structural Classification of Proteins (SCOP) showed that the domain definitions agreed for 80% of the representative protein chains. The effects of rhizobacteria, i.e. Pseudomonas fluorescens, Azotobacter chyroococcum and Azospirillum brasilense, alone and in combination with root symbionts, Rhizobium sp. and Glomus mosseae, on the growth of chickpea, Cicer arietinum, and reproduction of Meloidogyne jaranica were studied. When added alone G. mosseae was better at improving plant growth and reducing galling and nematode reproduction than any other tested organism. Application of P. fluorescens caused an almost similar increase in plant growth to that caused by Rhizobium sp., while use of A. chroococcum was better than A. brasilense in improving growth of nematode --infected plants. Combined use of P. fluorescens with G. mosseae was better at improving plant growth and reducing galling and nematode multiplication than any other combined treatment.  相似文献
