A solid-phase immunoadsorbent specific for terminal deoxynucleotidyl transferase has been prepared. The enzyme from calf thymus and acute lymphoblastic leukemia cells binds to columns of this material. Bound enzyme can be eluted in an active form. Selective and rapid purification of terminal deoxynucleotidyl transferase from crude extracts of cells containing this enzyme can be achieved by this method since the immunoadsorbent has no affinity for other cellular DNA polymerases.  相似文献

Color-Doppler ultrasonography was used to compare the vascularity of the preovulatory follicle between heifers that became pregnant compared with nonpregnant. Heifers (n=34) received a luteolytic dose of PGF2alpha when the diameter of the largest follicle of the second follicular wave reached > or =11 mm (actual diameter 11.63+/-0.06 mm). An ovulation-inducing injection of GnRH analogue was given 36 h after the PGF2alpha treatment. Artificial insemination (AI) was performed 26 h after GnRH treatment, and ovulation occurred in all heifers within 3h after AI. Follicle blood flow was assessed at Hour 0 (GnRH treatment) and at Hour 26 (AI). Follicle diameter at Hours 0 and 26 was greater (main effect of group; P<0.03) in the group that became pregnant (n=25) compared with nonpregnant (n=9). Percentage of follicle wall with power Doppler signals of blood flow showed an interaction of group-by-hour (P<0.03), reflecting greater blood flow in the pregnant group than in the nonpregnant group at Hour 26 but not at Hour 0. Spectral-Doppler evaluation of the resistance index for a vessel in the follicle wall resulted in a group-by-hour interaction (P<0.03) from a lower index in the pregnant group at Hour 26. The lower index indicated greater downstream vascular perfusion in the follicle wall. Results supported the hypothesis of a positive relationship between the extent of blood flow of the preovulatory follicle and successful establishment of pregnancy in cattle.  相似文献

Glasshouse experiment was conducted to assess the impact of green chopped leaves of four test plants (Lantana camara, Ficus virens, Kigelia pinnata and Ficus bengalensis) and two nematicides (Phorate and Carbofuran) on the plant growth parameters of tomato cv. K25 and on the root-knot development. Results revealed that all the tested treatments significantly (p = 0.05) improved plant growth parameters and reduced root-knot development compared to control. Among the tested organic additives, chopped green leaves of Lantana camara added to soil gave the highest enhancement in plant growth parameters, including plant height, fresh and dry weight, number of fruits and fruit weight with the values of 94.2 cm, 106.8 g, 31.6 g, 7.2 and 153.3 g respectively, as well as a greater reduction of Meloidogyne javanica reproduction and development but exhibiting a lower response compared to nematicides. There was also significant reduction in root-knot development in tomato plants growing in other organic additive amended soil.  相似文献

Subcutaneous pegylated interferon beta-1a (peginterferon beta-1a [PEG-IFN]) 125 μg every two or four weeks has been studied in relapsing-remitting multiple sclerosis (RRMS) patients in the pivotal Phase 3 ADVANCE trial. In the absence of direct comparative evidence, a network meta-analysis (NMA) was conducted to provide an indirect assessment of the relative efficacy, safety, and tolerability of PEG-IFN versus other injectable RRMS therapies. Systematic searches were conducted in MEDLINE, Embase, and the Cochrane Library, and conference proceedings from relevant annual symposia were hand-searched. Included studies were randomized controlled trials evaluating ≥1 first-line treatments including interferon beta-1a 30, 44, and 22 μg, interferon beta-1b, and glatiramer acetate in patients with RRMS. Studies were included based on a pre-specified protocol and extracted by a team of independent reviewers and information scientists, utilizing criteria from NICE and IQWiG. In line with ADVANCE findings, NMA results support that PEG-IFN every 2 weeks significantly reduced annualized relapse rate, and 3- and 6-month confirmed disability progression (CDP) versus placebo. There was numerical trend favoring PEG-IFN every 2 weeks versus other IFNs assessed for annualized relapse rate, and versus all other injectables for 3- and 6-month CDP (6-month CDP was significantly reduced versus IFN beta-1a 30 μg). The safety and tolerability profile of PEG-IFN beta-1a 125 μg every 2 weeks was consistent with that of other evaluated treatments. Study limitations for the NMA include variant definitions of relapse and other systematic differences across trials, assumptions that populations were sufficiently similar, and inability to perform NMA of adverse events. With similar efficacy compared to other RRMS treatments in terms of annualized relapse rate and 3- and 6-month CDP, a promising safety profile, and up to 93% reduction in number of injections (which may improve adherence), PEG-IFN every 2 weeks offers a valuable alternative treatment option for patients with RRMS.  相似文献

Hydrolysis of exogenous phosphatidylcholine (PtdCho) to 1,2-diacylglycerol by rat liver plasma membranes was stimulated by oleate concentrations as low as 0.1 mM. In the presence of 75 mM ethanol, the fatty acid also enhanced phosphatidylethanol (PtdEtOH) formation from PtdCho. These effects were also observed with linoleate and arachidonate, but not with saturated fatty acids or detergents, and were minimal in microsomes or mitochondria. Release of [3H]choline from exogenous Ptd[3H]Cho was stimulated by oleate, whereas phosphoryl[3H]choline formation was inhibited. Oleate and other unsaturated, but not saturated, fatty acids also stimulated the conversion of exogenous [14C]phosphatidic acid to [14C]diacylglycerol. These data are consistent with stimulatory effects of these fatty acids on both phospholipase D and phosphatidate phosphohydrolase in liver plasma membranes. The stimulatory effect of guanosine 5'-O-[3-thio]triphosphate) (20 microM) on PtdEtOH and diacylglycerol formation from PtdCho was enhanced by low concentrations of oleate. Phospholipase A2 also stimulated PtdEtOH and diacylglycerol formation from exogenous PtdCho. It is proposed that unsaturated fatty acids may play a physiological role in the regulation of diacylglycerol production through activation of phospholipase D and phosphatidate phosphohydrolase.  相似文献

Two new natural triterpenes, lantaninilic acid and lantoic acid, along with the known triterpenes lantadene A, and oleanolic, ursolic, betulinic, lantanolic, and camaric acid, were obtained from the aerial parts of Lantana camara through bioassay‐guided isolation, monitoring the in vitro antileishmanial activity against promastigotes of Leishmania major. Oleanolic acid ( 3 ), ursolic acid ( 4 ), lantadene A ( 5 ), and lantanilic acid ( 7 ) showed significant leishmanicidal activities with IC50 values of 53.0, 12.4, 20.4, and 21.3 μM , respectively. The IC50 value of ursolic acid ( 4 ; 12.4 μM ) was found to be comparable with that of the standard drugs, pentamidine (IC50 15.0 μM ) and amphotericin B (IC50 0.31 μM ). The in vitro activities of L. camara and its constituents against promastigotes of Leishmania major are reported here for the first time.  相似文献
