Bumblebees are widely distributed across the world and have great economic and ecological importance as pollinators in the forest as well as in agriculture. The insect midgut consists of three cell types, which play various important roles in digestion, absorption, and hormone production. The present study characterized the anterior and posterior midgut regions of the bumblebee, Bombus morio. The digestive, regenerative and endocrine cells in the midgut showed regional differences in their number, nuclear size, as well as the size of the striated border. Ultrastructurally, the digestive cells contained many mitochondria and long microvilli; however, in the anterior midgut region, these cells showed dilated basal labyrinths with a few openings for the hemocoel, whereas the labyrinths of the basal posterior region remained inverse characteristics. Thus, the characterization of the midgut of B. morio supported an ecto-endoperitrophic circulation, contributing to a better understanding of the digestive process in this bee. 相似文献

Trophic interactions play a critical role in the structure and function of ecosystems. Given the widespread loss of biodiversity due to anthropogenic activities, understanding how trophic interactions respond to natural gradients (e.g., abiotic conditions, species richness) through large‐scale comparisons can provide a broader understanding of their importance in changing ecosystems and support informed conservation actions. We explored large‐scale variation in reef fish trophic interactions, encompassing tropical and subtropical reefs with different abiotic conditions and trophic structure of reef fish community. Reef fish feeding pressure on the benthos was determined combining bite rates on the substrate and the individual biomass per unit of time and area, using video recordings in three sites between latitudes 17°S and 27°S on the Brazilian Coast. Total feeding pressure decreased 10‐fold and the composition of functional groups and species shifted from the northern to the southernmost sites. Both patterns were driven by the decline in the feeding pressure of roving herbivores, particularly scrapers, while the feeding pressure of invertebrate feeders and omnivores remained similar. The differential contribution to the feeding pressure across trophic categories, with roving herbivores being more important in the northernmost and southeastern reefs, determined changes in the intensity and composition of fish feeding pressure on the benthos among sites. It also determined the distribution of trophic interactions across different trophic categories, altering the evenness of interactions. Feeding pressure was more evenly distributed at the southernmost than in the southeastern and northernmost sites, where it was dominated by few herbivores. Species and functional groups that performed higher feeding pressure than predicted by their biomass were identified as critical for their potential to remove benthic biomass. Fishing pressure unlikely drove the large‐scale pattern; however, it affected the contribution of some groups on a local scale (e.g., large‐bodied parrotfish) highlighting the need to incorporate critical functions into conservation strategies. 相似文献

An α-glucosidase enzyme produced by the fungus Thermoascus aurantiacus CBMAI 756 was purified by ultra filtration, ammonium sulphate precipitation, and chromatography using Q Sepharose, Sephacryl
S-200, and Superose 12 columns. The apparent molecular mass of the enzyme was 83 kDa as determined in gel electrophoresis.
Maximum activity was observed at pH 4.5 at 70°C. Enzyme showed stability stable in the pH range of 3.0–9.0 and lost 40% of
its initial activity at the temperatures of 40, 50, and 60°C. In the presence of ions Na+, Ba2+, Co2+, Ni2+, Mg2+, Mn2+, Al3+, Zn2+, Ca2+ this enzyme maintained 90–105% of its maximum activity and was inhibited by Cr3+, Ag+, and Hg2+. The enzyme showed a transglycosylation property, by the release of oligosaccharides after 3 h of incubation with maltose,
and specificity for short maltooligosaccharides and α-PNPG. The Km measured for the α-glucosidase was 0.07 μM, with a Vmax of 318.0 μmol/min/mg. 相似文献

Complexes of general formula [{CuX}2(YNC10H14O)] (X = Cl; Y = NHMe, NH2 or X = Br; Y = NH2) were synthesised from camphor hydrazone ligands (YNC10H14O) by reaction with the suitable copper(I) halide. Structural analysis by X-rays performed on a red crystal of [{CuCl}2(Me2NNC10H14O)] revealed that the complex is a one-dimensional copper polymer formed by two rather different copper units bridged by chloride. One of the copper units displays a tetrahedral geometry while the other is linear. Although the geometries and neighbourhoods of the two copper units are very different the oxidation state of the metal is the same, i.e. Cu(I) as corroborated by magnetic and electrochemical measurements.The ability of [{CuCl}2(Me2NNC10H14O)] to promote the activation of 4-pentyn-1-ol towards cyclization was studied under homogeneous or heterogeneous experimental conditions. Candida albicans and C. tropicalis obtained from whole saliva of patients presenting signs of oral candidosis were assayed for quantification of colony forming
units, exoenzyme activity (phospholipase and proteinase) and antifungal drug sensitivity (amphotericin B, fluconazole and
itraconazole) by the reference method of the Clinical and Laboratory Standards Institute. The number of colony forming units
per milliliter varied according to the Candida species involved and whether a single or mixed infection was present. Proteinase activity was observed in both C. albicans and C. tropicalis, but phospholipase activity was noted only in C. albicans. Interest in producing large quantities of supercoiled plasmid DNA has recently increased as a result of the rapid evolution of gene therapy and DNA vaccines. Owing to the commercial interest in these approaches, the development of production and purification strategies for gene-therapy vectors has been performed in pharmaceutical companies within a confidential environment. Consequently, the information on large-scale plasmid purification is scarce and usually not available to the scientific community. Studies investigating the impacts of biodiversity loss on ecosystem processes have often reached different conclusions, probably
because insufficient attention has been paid to some aspects including (1) which biodiversity measure (e.g., species number,
species identity or trait) better explains ecosystem functioning, (2) the mechanisms underpinning biodiversity effects, and
(3) how can environmental context modulates biodiversity effects. Here, we investigated how species number (one to three species)
and traits of aquatic fungal decomposers (by replacement of a functional type from an unpolluted site by another from a metal-polluted
site) affect fungal production (biomass acumulation) and plant litter decomposition in the presence and absence of metal stress.
To examine the putative mechanisms that explain biodiversity effects, we determined the contribution of each fungal species
to the total biomass produced in multicultures by real-time PCR. In the absence of metal, positive diversity effects were
observed for fungal production and leaf decomposition as a result of species complementarity. Metal stress decreased diversity
effects on leaf decomposition in assemblages containing the functional type from the unpolluted site, probably due to competitive
interactions between fungi. However, dominance effect maintained positive diversity effects under metal stress in assemblages
containing the functional type from the metal-polluted site. These findings emphasize the importance of intraspecific diversity
in modulating diversity effects under metal stress, providing evidence that trait-based diversity measures should be incorporated

of human immunodeficiency virus in experimental candidiasis suggested reduction in fungal infection, and medicinal plants

C. albicans from clinical isolates and the action of plant extracts against strains of C. albicans and its synthesized proteinases, comparing with antifungal fluconazole and amphotericin B and proteinase inhibitors pepstatin
A, amprenavir, and ritonavir. The results reported here showed that these extracts have a certain kind of action and that
