High-dose chemotherapy (HDT) and autologous bone marrow rescue (ABMR) is routinely used as consolidation treatment in advanced neuroblastoma. This study is presently examining the efficacy and toxicity of combined [131I]metaiodobenzylguanidine (131I-MIBG) therapy with HDT and ABMR. Five children (4 male, 1 female), median age of 8 years (range 4-11 years) were treated, 3 at relapse and 2 after initial chemotherapy. A single infusion of 131I-MIBG (median activity 11.1 GBq, range 7.4-11.2 GBq) was followed by HDT and ABMR 14-32 days later. High-dose chemotherapy consisted of carboplatin and melphalan in 4 patients, and vincristine, etoposide, carboplatin and melphalan in 1. One patient developed a septicaemia and died, and another failed to engraft; both had extensive bone marrow infiltration at the time of 131I-MIBG therapy. The combined therapy was well tolerated by the three other patients. Two children have relapsed and died (including one who failed to engraft), and 2 are alive 17 and 41 months after ABMR. In the absence of extensive bone marrow metastases, combined therapy offers potential as a means of consolidating treatment in advanced neuroblastoma. 相似文献

The objective of this study was to characterise the antagonistic activity of cellular components of potential probiotic bacteria isolated from the gut of healthy rohu (Labeo rohita), a tropical freshwater fish, against the fish pathogen, Aeromonas hydrophila. Three potential probiotic strains (referred to as R1, R2, and R5) were screened using a well diffusion, and their antagonistic activity against A. hydrophila was determined. Biochemical tests and 16S rRNA gene analysis confirmed that R1, R2, and R5 were Lactobacillus plantarum VSG3, Pseudomonas aeruginosa VSG2, and Bacillus subtilis VSG1, respectively. Four different fractions of cellular components (i.e. the whole-cell product, heat-killed whole-cell product [HKWCP], intracellular product [ICP], and extracellular product) of these selected strains were effective in an in vitro sensitivity test against 6 A. hydrophila strains. Among the cellular components, the ICP of R1, HKWCP of R2, and ICP of R5 exhibited the strongest antagonistic activities, as evidenced by their inhibition zones. The antimicrobial compounds from these selected cellular components were partially purified by thin-layer and high-performance liquid chromatography, and their properties were analysed. The ranges of pH stability of the purified compounds were wide (3.0–10.0), and compounds were thermally stable up to 90 °C. Considering these results, isolated probiotic strains may find potential applications in the prevention and treatment of aquatic aeromonosis. 相似文献

Sol-gel derived silica has tremendous applications as a biocompatible scaffold for the immobilization of cells. The use of xerogel as a matrix in the blueprint of biosensors is an appealing proposition due to several inimitable characteristics of xerogels, primarily because of their high porous nature, amendable pore size, and exceptionally large internal surface area. Morphological (X-Ray Diffraction and Thermogravimmetric Analysis) and optical (Fourier Transform Infrared and UV-Vis absorption) studies of the silica matrices with entrapped Rhizobial (Rz) structure of the biomaterial has been made. Temporal and concentration dependent studies were conducted for impregnated samples; it showed that the response time for the new biosensor for determining the concentration of Rz is less than 20 min. One of three preparations—silicone oil, hydrocortisone acetate, and hydrocortisone acetate plus saline—was injected into 22 osteoarthritic and 49 rheumatoid knees. Silicone injected into stiff, dry, grating knees which were the site of chronic arthritis did not promote better movement than did hydrocortisone alone. Hydrocortisone plus saline, however, appeared to be more effective than silicone or hydrocortisone alone in increasing movement in rheumatoid knees.
