Summary The secretion of sperm maturation-related molecules by the epididymis is subjected to developmental and hormonal regulation. In the BALB/c mouse, we found that GP-83 and GP-49, two sperm maturationrelated glycoproteins, were secreted by the epididymis. The present study investigated the postnatal development and testosterone-dependence of these two molecules. Histochemical localization in paraffin sections revealed that wheat-germ agglutinin (WGA)-binding sites were first present in the epididymis of 4-week-old mice. The distribution of WGA-binding sites was the same as that of more mature mice, i.e., it was first found in the principal cells of the corpus epididymidis, and gradually appeared in the contents of epididymal tubules. On WGA blots, GP-83 and GP-49 were identified in the corpus, and GP-83 was identified in the cauda of the epididymis. In mice that had received unilateral orchiectomy at 4 weeks of age, GP-83 and GP-49 were present in both intact and orchiectomized epididymides 4 weeks after the operation. In the epididymides of mice that had received bilateral orchiectomy, GP-83 and GP-49 were barely identifiable. However, the presence of these two molecules was restored if testosterone was supplemented immediately after orchiectomy. These results indicate that GP-83 and GP-49 are secreted de novo in the epididymis, and that the secretion of these two molecules is developmentally regulated and androgen-dependent.  相似文献

A large number of fresh water and saline ponds are located in the Labyrinth (77° 33 S, 160° 50 E) of the upper Wright Valley in the Dry Valleys region of southern Victoria Land, Antarctica. It is situated near the terminus of the Wright Upper Glacier between 800–1000 m above sea level. From a limnological point of view, the most interesting problems concerning these saline ponds are the origin of their salts and their evolutional history.Chloride ion contents vary remarkably among the ponds ranging from 0.0049 to 52.4 g kg–1. Surprisingly, more than a half of the ponds are saline with the highest chloride ion content being 2.7 times greater than that of seawater. The D and 18O values of the pond waters indicate a snow and/or glacial meltwaters origin, and that the ponds underwent subsequent alteration due to evaporation or freezing. The composition of chemical components reveal no evidence of trapped seawater. Thus the salt concentrations in the Labyrinth pond waters must be explained principally by the accumulation of atmospheric salts and subsequent repeated cycles of evaporation and freezing of the pond waters over considerable time periods.  相似文献

Polysaccharides-based membranes of chitosan and cellulose blends were prepared using trifluoroacetic acid as a co-solvent. Morphology and mechanical property of prepared membranes were studied by Instron and dynamic mechanical thermal analysis. The mechanical and dynamic mechanical thermal properties of the cellulose/chitosan blends appear to be dominated by cellulose, suggests that cellulose/chitosan blends were not well miscible. It is believed that the intermolecular hydrogen bonding of cellulose is supposed to be break down to form cellulose–chitosan hydrogen bonding; however, the intra-molecular and intra-strand hydrogen bonds hold the network flat. The reduced water vapor transpiration rate through the chitosan/cellulose membranes indicates that the membranes used as a wound dressing may prevent wound from excessive dehydration. The chitosan/cellulose blend membranes demonstrate effective antimicrobial capability against Escherichia coli and Staphylococcus aureus, as examined by the antimicrobial test. These results indicate that the chitosan/cellulose blend membranes may be suitable to be used as a wound dressing with antibacterial properties.  相似文献

Recent studies of cyclooxygenase-2 (COX-2) inhibitors suggest that the balance between thromboxane and prostacyclin is a critical factor in cardiovascular homeostasis. Disruption of prostacyclin signaling by genetic deletion of the receptor or by pharmacological inhibition of COX-2 is associated with increased atherosclerosis and restenosis after injury in animal models and adverse cardiovascular events in clinical trials (Vioxx). Human vascular smooth muscle cells (VSMC) in culture exhibit a dedifferentiated, migratory, proliferative phenotype, similar to what occurs after arterial injury. We report that the prostacyclin analog iloprost induces differentiation of VSMC from this synthetic, proliferative phenotype to a quiescent, contractile phenotype. Iloprost induced expression of smooth muscle (SM)-specific differentiation markers, including SM-myosin heavy chain, calponin, h-caldesmon, and SM alpha-actin, as determined by Western blotting and RT-PCR analysis. Iloprost activated cAMP/protein kinase A (PKA) signaling in human VSMC, and the cell-permeable cAMP analog 8-bromo-cAMP mimicked the iloprost-induced differentiation. Both myristoylated PKA inhibitor amide-(14-22) (PKI, specific PKA inhibitor), as well as ablation of the catalytic subunits of PKA by small interfering RNA, opposed the upregulation of contractile markers induced by iloprost. These data suggest that iloprost modulates VSMC phenotype via G(s) activation of the cAMP/PKA pathway. These studies reveal regulation of VSMC differentiation as a potential mechanism for the cardiovascular protective effects of prostacyclin. This provides important mechanistic insights into the induction of cardiovascular events with the use of selective COX-2 inhibitors.  相似文献

Dengue virus (DV) causes a non-specific febrile illness known as Dengue fever (DF), and a severe life-threatening illness, Dengue hemorrhagic fever/Dengue shock syndrome (DHF/DSS). Hemostatic changes induced by this virus involve three main factors: thrombocytopenia, endothelial cell damage, and significant abnormalities of the coagulation and fibrinolysis systems. The pathogenesis of bleeding in DV infections remains unknown. In this article, we focused on the DV activating endothelial cells and altering the parameters of hemostasis system. The expression of hemostasis-related factors, Thrombomodulin, TF, TFPI, t-PA, and PAI-1, in DV-infected cells were determined by RT-PCR. Flow cytometry analysis and immunofluorescence staining confirmed that the expression levels of TM in the DV-infected HMEC-1 and THP-1 cells were increased. In addition, the purified recombinant domain III of the envelope glycoprotein of DV (EIII) could induce the expression of TM in the HMEC-1 cells and THP-1 cells. The TM expression induced by DV or EIII in the endothelial cells and monocytic cells suggests that the EIII of DV plays an important role in the pathogenesis of DHF/DSS.  相似文献

The bimolecular fluorescence complementation (BiFC) assay is a powerful tool for visualizing and identifying protein interactions in living cells. This assay is based on the principle of protein-fragment complementation, using two nonfluorescent fragments derived from fluorescent proteins. When two fragments are brought together in living cells by tethering each to one of a pair of interacting proteins, fluorescence is restored. Here, we provide a protocol for a Venus-based BiFC assay to visualize protein interactions in the living nematode, Caenorhabditis elegans. We discuss how to design appropriate C. elegans BiFC cloning vectors to enable visualization of protein interactions using either inducible heat shock promoters or native promoters; transform the constructs into worms by microinjection; and analyze and interpret the resulting data. When expression of BiFC fusion proteins is induced by heat shock, the fluorescent signals can be visualized as early as 30 min after induction and last for 24 h in transgenic animals. The entire procedure takes 2-3 weeks to complete.  相似文献

Studies of protein interactions have increased our understanding and knowledge of biological processes. Assays that utilize fluorescent proteins, such as fluorescence resonance energy transfer (FRET) and bimolecular fluorescence complementation (BiFC), have enabled direct visualization of protein interactions in living cells. However, these assays are primarily suitable for a pair of interacting proteins, and methods to visualize and identify multiple protein complexes in vivo are very limited. This protocol describes the recently developed BiFC-FRET assay, which allows visualization of ternary complexes in living cells. We discuss how to design the BiFC-FRET assay on the basis of the validation of BiFC and FRET assays and how to perform transfection experiments for acquisition of fluorescent images for net FRET calculation. We also provide three methods for normalization of the FRET efficiency. The assay employs a two-chromophore and three-filter FRET setup and is applicable to epifluorescence microscopes. The entire protocol takes about 2-3 weeks to complete.  相似文献

This study was designed to determine the effects of 17beta-estradiol (E2) in overcoming the cardiac over-loading and cardiac fibrosis in rats. E2 (100 ng/kg) or oil was applied in female Sprague-Dawley rats with or without bilateral ovariectomy and with or without coarctation of the abdominal aorta after 4 or 8 days. By post-operative day 4, the heart weight, the left ventricular weight, the latent form of MMP-2 in rat hearts with or without the ovary intact had significantly increased while these changes were reversed after E2 treatment. Although animals with the ovaries intact overcame the hypertrophic effects and the consumption of MMP-2, these effects were not restored in ovariectomized animals in which more fibrosis could be found by day 8. Among the IGF-I signaling, the levels of IGF-I, the activities of PI3K-Akt for cardiomyocyte survival, and MEK-ERKs for non-cardiomyocyte proliferation pathways had significantly increased by day 4. These increasing trends were enhanced by E2 treatment. However, down-regulation was only observed on day 8 in ovariectomized animals. Similarly, elevated expressions of the steady-state mRNA of IGF-I, IGF-IR, and Cox vb were observed on day 4 in animals with the ovaries intact and these expressions were enhanced by E2 treatment. In contrast, down-regulation on day 8 in ovariectomized animals was not enhanced by E2. The calcineurin/NFAT-3 pathway was suppressed on day 4 but was elevated on day 8 in ovariectomized animals. These findings indicate that signaling pathways may be plausible mechanisms for the cardiac protective effects of E2 administration.  相似文献
