本研究构建了水牛卵巢黄体形成及退化过程的定量蛋白质表达谱,从中寻找与水牛发情周期相关的重要蛋白质。为研究水牛卵巢周期性变化的分子机制提供蛋白质水平的数据支撑,进而为提高水牛繁殖效率奠定实验基础。以水牛排卵后的红体期(corpus hemorrhagicum,CH)、黄体期(corpus luteum,CL)和白体期(corpus fibrosum,CF)的卵巢作为研究对象,利用串联质谱标签(tandem mass tag,TMT)标记结合液质联用((liquid chromatography/mass spectrometry,LC-MS/MS)的定量蛋白质组学技术,采用相关软件对得到的差异表达蛋白质进行生物信息学分析。同时对与水牛发情周期相关的重要差异蛋白质:单核巨噬细胞分化抗原(CD14)、通用转录因子Ⅱ-Ⅰ(GTF2I)、羟基类固醇(17β)脱氢酶1(HSD17B1)、U6 sn RNA相关Sm样蛋白质(LSM1)和纤溶酶原激活物抑制物(SERPINE1)进行了qRT-PCR分析验证。结果显示,共鉴定得到2 343种蛋白质,其中差异表达蛋白质有284种(差异倍数≥2.0),初步构建了水牛卵巢黄体形成和退化的定量蛋白质表达谱。对其中的五种重要差异蛋白质(CD14、GTF2I、HSD17B1、LSM1和SERPINE1)进行qRT-PCR验证,结果有四种(CD14、HSD17B1、LSM1和SERPINE1)的qPCR结果与质谱结果相一致,仅GTF2I与质谱结果不一致,可能是由于GTF2I存在转录后调控。本研究首次从蛋白质水平对水牛卵巢排卵后的周期性变化的分子机制进行了探究,同时为其他家畜品种发情周期的研究提供了新的研究思路。  相似文献

Docosahexaenoic acid (DHA) plays an important role in visual function but has a highly oxidation-prone chemical structure. Therefore, we investigated how dietary DHA affects the generation of lipid peroxides in rat retina under oxidative stress in diabetes with/without vitamin E (VE) deficiency. Streptozotocin-induced (50 mg i.p./kg B.W.) diabetic Sprague-Dawley (SD) rats were assigned to four groups: (i) control/VE(+), (ii) DHA/VE(+), (iii) control/VE( - ) and (iv) DHA/VE( - ), and raised for 28 days. We then measured lipid peroxide levels in the retina, serum and liver. With a normal intake of VE, dietary DHA increased only the retinal level of thiobarbituric acid-reactive substances (TBARS) slightly. In contrast, in rats with VE deficiency, dietary DHA increased serum and liver lipid peroxide levels but not in the retina. These results suggest that dietary DHA does not necessarily promote lipid peroxidation in the retina even under high oxidative stress.  相似文献

An extremely low‐frequency magnetic field (ELF‐MF) is generated by power lines and household electrical devices. Many studies have suggested an association between chronic ELF‐MF exposure and anxiety and/or depression. The mechanism of these effects is assumed to be a stress response induced by ELF‐MF exposure. However, this mechanism remains controversial. In the present study, we investigated whether chronic ELF‐MF exposure (intensity, 3 mT; total exposure, 200 h) affected emotional behavior and corticosterone synthesis in mice. ELF‐MF‐treated mice showed a significant increase in total immobility time in a forced swim test and showed latency to enter the light box in a light–dark transition test, compared with sham‐treated (control) mice. Corticosterone secretion was significantly high in the ELF‐MF‐exposed mice; however, no changes were observed in the amount of the adrenocorticotropic hormone and the expression of genes related to stress response. Quantification of the mRNA levels of adrenal corticosteroid synthesis enzymes revealed a significant reduction in Cyp17a1 mRNA in the ELF‐MF‐exposed mice. Our findings suggest the possibility that high intensity and chronic exposure to ELF‐MF induces an increase in corticosterone secretion, along with depression‐ and/or anxiety‐like behavior, without enhancement of the hypothalamic–pituitary–adrenal axis. An increasing number of studies are being conducted to examine the density- or trait-mediated indirect effects of large herbivores on phytophagous insects. However, these effects are highly context dependent and no general trends have been made clear. We conducted a meta-analysis focusing on three factors capable of affecting detection of the indirect effects of large herbivores on phytophagous insects: type of response variable, experimental scale, and characteristics of study organisms. Overall, large herbivores exerted a negative effect on insects, a trend that was prominent in studies using insect abundance as a response variable. No particular trends were observed in studies using herbivory rate as a variable, and these studies often focused on plant trait-mediated effects more than density-mediated ones. Experimental scale affected the strength of indirect effects: within-year or individual tree level experiments did not follow any trends, whereas 1–10 year experiments or 0–10 ha scale experiments show a negative impact on insects. Characteristics of large herbivores and growth forms of transmitter plants also contributed to variations in the observed effect size; negative effects were reported in livestock-grassland ecosystems and neutral effects in tree-dominated systems. There was a close association between response variable, experimental scale, and characteristics of study organisms, and these effects jointly contributed to the apparent trends. To predict the impacts of large herbivores at ecosystem level, it is necessary to eliminate these biases arising from study design and to evaluate the effect on insect densities at large spatial and temporal scales.  相似文献

Species composition and fruiting season of ammonia fungi were investigated in Iriomote Island of the southern Ryukyus, Okinawa Prefecture, Japan.Castanopsis andPinus forests were surveyed and 10 species of ammonia fungi were collected, including one new agaric species,Hebeloma luchuense sp. nov. This new fungus is characterized by having a rooting, squamulose-scaly stipe and cortinate veil and forms ectomycorrhizae withCastanopsis cuspidata var.sieboldii. Although the general mushroom season in theCastanopsis forest in Iriomote island was very short and restricted to summer, ammonia fungi were observed to fruit throughout the year in urea-treated plots.  相似文献

Phosphatidylcholine‐specific phospholipase C (PC‐PLC) is the major enzyme in the Phosphatidylcholine (PC) cycle and is involved in many long‐term cellular responses such as activation, proliferation, and differentiation events. Cell division cycle 20 homolog (Cdc20) is an essential cell‐cycle regulator required for the completion of mitosis. Our previous studies identified the interaction between PC‐PLC and Cdc20. Through the interaction, Cdc20 could mediate the degradation of PC‐PLC by Cdc20‐mediated ubiquitin proteasome pathway (UPP). In this study, we found that PC‐PLC might not be involved in cancer metastasis. Inhibition of PC‐PLC by D609 could cause cell proliferation inhibition and apoptosis inhibition in CBRH‐7919 cells. Inhibition of PC‐PLC could also influence the cell cycle by arresting the cells in G1 phase, and Cdc20 might be involved in these processes. Taken together, in this report, we provided new evidence for the functional roles of PC‐PLC and Cdc20 in the cell cycle, proliferation, and apoptosis in CBRH‐7919 cells. In order to understand how inter-subunit association stabilizes oligomeric proteins, a single polypeptide chain variant of heptameric co-chaperonin GroES (tandem GroES) was constructed from Escherichia coli heptameric GroES by linking consecutively the C-terminal of one subunit to the N-terminal of the adjacent subunit with a small linker peptide. The tandem GroES (ESC7) showed properties similar to wild-type GroES in structural aspects and co-chaperonin activity. In unfolding and refolding equilibrium experiments using guanidine hydrochloride (Gdn-HCl) as a denaturant at a low protein concentration (50 microg ml(-1)), ESC7 showed a two-state transition with a greater resistance toward Gdn-HCl denaturation (Cm=1.95 M) compared to wild-type GroES (Cm=1.1 M). ESC7 was found to be about 10 kcal mol(-1) more stable than the wild-type GroES heptamer at 50 microg ml(-1). Kinetic unfolding and refolding experiments of ESC7 revealed that the increased stability was mainly attributed to a slower unfolding rate. Also a transient intermediate was detected in the refolding reaction. Interestingly, at the physiological GroES concentration (>1 mg ml(-1)), the free energy of unfolding for GroES heptamer exceeded that for ESC7. These results showed that at low protein concentrations (<1 mg ml(-1)), the covalent linking of subunits contributes to the stability but also complicates the refolding kinetics. At physiological concentrations of GroES, however, the oligomeric state is energetically preferred and the advantages of covalent linkage are lost. This finding highlights a possible advantage in transitioning from multi-domain proteins to oligomeric proteins with small subunits in order to improve structural and kinetic stabilities.  相似文献

Rubiscolin-6 (Tyr-Pro-Leu-Asp-Leu-Phe) is a delta opioid peptide derived from the large subunit of spinach d-ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco). We previously reported that rubiscolin-6 had an analgesic effect and stimulated memory consolidation. Here we show that intraperitoneally (i.p.) or orally administered rubiscolin-6 has an anxiolytic effect at a dose of 10 mg/kg or 100 mg/kg, respectively, in the elevated plus-maze test in mice. The anxiolytic effects of rubscolin-6 after i.p. (10 mg/kg) and oral (100 mg/kg) administration were blocked by a delta opioid receptor antagonist, naltrindole (1 mg/kg, s.c.), suggesting that the anxiolytic activity of rubiscolin-6 is mediated by delta opioid receptor. The anxiolytic effect of rubiscolin-6 (10 mg/kg, i.p.) was also blocked by a dopamine D(1) antagonist, SCH23390 (30 microg/kg, i.p.), but not by a dopamine D(2) antagonist, raclopride (15 microg/kg, i.p.). The anxiolytic effect of rubiscolin-6 (10 mg/kg, i.p.) was blocked by sigma(1) receptor antagonist, BMY14802 (0.5 mg/kg, i.p.) or BD1047 (10 mg/kg, i.p.). Taken together, the anxiolytic effect of rubiscolin-6 is mediated by sigma(1) and dopamine D(1) receptors downstream of delta opioid receptor.  相似文献
