Autophagy is a unique intracellular protein degradation system accompanied by autophagosome formation. Besides its important role through bulk degradation in supplying nutrients, this system has an ability to degrade certain proteins, organelles, and invading bacteria selectively to maintain cellular homeostasis. In yeasts, Atg8p plays key roles in both autophagosome formation and selective autophagy based on its membrane fusion property and interaction with autophagy adaptors/specific substrates. In contrast to the single Atg8p in yeast, mammals have 6 homologs of Atg8p comprising LC3 and GABARAP families. However, it is not clear these two families have different or similar functions. The aim of this study was to determine the separate roles of LC3 and GABARAP families in basal/constitutive and/or selective autophagy. While the combined knockdown of LC3 and GABARAP families caused a defect in long-lived protein degradation through lysosomes, knockdown of each had no effect on the degradation. Meanwhile, knockdown of LC3B but not GABARAPs resulted in significant accumulation of p62/Sqstm1, one of the selective substrate for autophagy. Our results suggest that while mammalian Atg8 homologs are functionally redundant with regard to autophagosome formation, selective autophagy is regulated by specific Atg8 homologs.  相似文献

Glioma is a huge threat for human being because it was hard to be completely removed owing to both the infiltrating growth of glioma cells and integrity of blood brain barrier. Thus effectively imaging the glioma cells may pave a way for surgical removing of glioma. In this study, a fluorescent probe, Cy3, was anchored onto the terminal of AS1411, a glioma cell targeting aptamer, and then TGN, a BBB targeting peptide, was conjugated with Cy3-AS1411 through a PEG linker. The production, named AsT, was characterized by gel electrophoresis, 1H NMR and FTIR. In vitro cellular uptake and glioma spheroid uptake demonstrated the AsT could not only be uptaken by both glioma and endothelial cells, but also penetrate through endothelial cell monolayer and uptake by glioma spheroids. In vivo, AsT could effectively target to glioma with high intensity. In conclusion, AsT could be used as an effective glioma imaging probe.  相似文献

Endo-α-N-acetylgalactosaminidase catalyzes the release of Galβ1-3GalNAc from the core 1-type O-glycan (Galβ1-3GalNAcα1-Ser/Thr) of mucin glycoproteins and synthetic p-nitrophenyl (pNP) α-linked substrates. Here, we report the enzymatic syntheses of core 1 disaccharide-containing glycopeptides using the transglycosylation activity of endo-α-N-acetylgalactosaminidase (EngBF) from Bifidobacterium longum. The enzyme directly transferred Galβ1-3GalNAc to serine or threonine residues of bioactive peptides such as PAMP-12, bradykinin, peptide-T and MUC1a when Galβ1-3GalNAcα1-pNP was used as a donor substrate. The enzyme was also found to catalyze the reverse-hydrolysis reaction. EngBF synthesized the core 1 disaccharide-containing oligosaccharides when the enzyme was incubated with either glucose or lactose and Galβ1-3GalNAc prepared from porcine gastric mucin using bifidobacterial cells expressing endo-α-N-acetylgalactosaminidase. Synthesized oligosaccharides are promising prebiotics for bifidobacteria.  相似文献

目的:探讨中国近15年腹茧症的流行病学特征和诊疗经验.方法:总结中国1994年1月-2009年1月问的腹茧症文献资料.结果:中国近15年共报道903例腹茧症,男女比例为1:1.35,平均年龄33.14岁,51.7%分布在华东地区,90.5%以不同表现形式的肠梗阻为主要表现,70.5%属于弥漫型腹茧症,40.2%患者无大网膜,手术以包膜切除为主.结论:腹茧症主要分布在华东地区,术前诊断困难,切除包膜和松解粘连是治疗本病的有效方法.  相似文献

【目的】本研究利用Asd+平衡致死系统构建表达巴氏杆菌毒素(Pasteurella multocida toxin,PMT)的重组猪霍乱沙门氏菌株,并对重组菌株的生物学特性进行比较研究。【方法和结果】通过基因克隆的方法构建表达PMT的重组质粒pYA-PmtC,再将其电转化减毒猪霍乱沙门氏菌C500的asd基因缺失株C501,构建口服活疫苗菌株C501(pYA-PmtC)。研究结果表明重组菌株C501(pYA-PmtC)的生化特性、血清型和生长速度与亲本菌株C500一致;在没有选择压力的条件下,C501(pYA-PmtC)能够稳定遗传重组质粒及其外源基因片段,并能稳定、高效、分泌性表达30.5kDa的外源保护性抗原rPmtC。C501(pYA-PmtC)腹腔感染BALB/c小鼠的LD50为8.5×106CFU,毒力稍低于C500(LD50为4.4×106CFU);口服接种C501(pYA-PmtC)和C500的所有仔猪未见任何发病症状,两者没有显著差别。【结论】本研究利用Asd+平衡致死系统的原理构建表达T+Pm保护性抗原重组猪霍乱沙门氏菌弱毒菌株C501(pYA-PmtC),为进一步开发猪萎缩性鼻炎-副伤寒的双价基因工程疫苗奠定基础。  相似文献

目的:研究TGF-α、TGF-β1与胰腺癌临床病理的关系.方法:免疫组织化学SABC法检测41例胰腺癌组织和12例正常胰腺组织中TGF-α、TGF-β1的表达情况,并分析其与病人的年龄、性别、肿瘤部位、病理分级和分期(UICC)等指标的相关性.结果:在41例胰腺癌组织中TGF-αTGF-β1的阳性表达率分别为73.2%、63.4%,在12例正常胰腺组织中的阳性表达率分别为16.7%、25.0%,两组之间存在显著差异(P=0.001).在胰腺癌组织中,TGF-β1的表达在不同分期中存在显著差异(P=0.019);TGF-α的表达与胰腺癌患者的年龄、性别、肿瘤部位、大小、分期及组织学分级无相关性(P>0.05).结论:TGF-α、TGF-β1在胰腺癌中高表达.TGF-β1与胰腺癌病理分期有关.  相似文献

Time-dependent changes of phytoavailability of Sr after adding it to the soil were studied for better prediction of radioactive Sr behavior in soil–plant systems. Strontium-86 enriched stable isotope was added to a soil sample collected from a field of humus-rich Andosol. Plant uptake and extractability of Sr in the soil were assessed at given times after the addition during a 12-month period including repeated drying-rewetting. Proportion of the added Sr extracted by 1 M ammonium acetate solution from the soil was 45–58% throughout the period, which corresponded to that of long-time aged 90Sr in the soil. Pot cultivations of orchardgrass (Dactylis glomerata) and red clover (Trifolium pratense) were carried out starting with uncropped soil for four weeks five times during the experiment. Clear time-dependent changes were not observed in uptake of the added Sr by the plants. Aging is unlikely to decrease Sr availability over time. The labile form of Sr determined with the isotopic dilution method was approximately 15% of total soil Sr, and more than the percentage of the extractable Sr in the soil. The results suggest that a part of the non-extractable forms of Sr in the soil would be available to the plant.  相似文献
