A high gastric cancer mortality in Fujian province (Peoples Republic of China) has been associated with the consumption of certain salted fermented fish products such as fish sauce (FS). We have investigated the levels and nature of N-nitroso compounds (NOC) and genotoxins present, before and after nitrosation, in 49 FS samples collected from villages in this high-risk area, pooled into six samples. The concentrations of total NOC before nitrosation ranged from 0.2 to 16 mumoles/l, and after nitrosation at pH 2 and pH 7, they rose by up to 4800- and 100-fold, respectively. In nitrosated samples, 40-50% of total NOC was not extractable into organic solvents; volatile N nitrosamines accounted for 1-2% and N-nitrosamino acids for 8-16% of total NOC. None of the FS samples exhibited genotoxic activity, but after nitrosation all were weakly active in the SOS chromotest. The highest SOS-inducing potency was observed with nitrosated ethyl acetate extracts of most samples. The formation of methylating agents was measured by incubation of nitrosated FS with DNA and subsequent analysis of 7-methylguanine adduct. 2 of the 6 nitrosated FS samples caused a slight increase in DNA methylation. 1 pooled home-made FS sample (the only one tested) contained tumour promoter-like substances, as measured by expression of certain EBV genes in Raji cells. HPLC fractionation of ethyl acetate extracts of FS samples allowed identification of three UV-absorbing peaks that, upon nitrosation, produced direct-acting genotoxins. This genotoxicity was partly ascribed to the formation of nitrite-derived arene diazonium cations that were characterized by a coupling reaction with N-ethyl-1-naphthylamine and thin-layer chromatography.  相似文献

Nitric oxide (NO*) is a diatomic free radical which has recently been found to have a key role in both normal physiological processes and disease states. The presence of NO in biological systems leads to the formation of reactive nitrogen species (RNS) such as peroxynitrite which reacts avidly with tyrosine residues in proteins to form nitrotyrosine (NTYR). Since peroxynitrite has a very short half-life at neutral pH, the presence of NTYR has been used as a marker of RNS production in various tissues. A number of methods for separation, detection, and quantitation of NTYR in biological samples have been developed. These methods include immunochemical techniques such as immunhistochemistry, ELISA, and Western blotting, high-performance liquid chromatography (HPLC) in combination with various detection systems including UV and electrochemical detection (ECD), gas chromatography (GC), gas chromatography-mass spectrometry (GC-MS), and electrospray mass spectrometry. In terms of sensitivity and specificity, it would appear that methods based on combinations of HPLC and various types of ECD are very versatile giving a limit of detection of 20 fmol per injection of protein hydrolysate. They are only limited by the sample quantity and the preparation that is required to achieve acceptable chromatograms. In addition to the detection of NTYR as a marker of RNS, its role in biological systems may be more subtle with nitration of key tyrosine residues likely to profoundly affect cellular function such as signaling cascades. Further advances are likely to be made in the localization of NTYR residues in peptide fragments using mass spectrometry.  相似文献

Improvements in analytical methodology have led to the detection and quantification of 'background' levels of a number of DNA and protein adducts. Many of these adducts are derived from 'low molecular weight' reactive species which may be generated during normal physiological processes, metabolic pathways or inflammatory processes. The adducts have been detected using gas chromatography-mass spectrometry, HPLC in combination with various detection systems, 32P-postlabelling and immunoassay methods. The reliability and accuracy of many widely used methods for adduct measurements are discussed with reference to several examples where human data is available, namely 4-aminobiphenyl, malondialdehyde, methylating agents, ethylene oxide and hydroxyl radical damage. The accurate and specific quantitation of 'background' levels of damage is essential if reliable estimates of increases in risk associated with incremental increases in exposure to exogenous agents are to be calculated. In experimental studies using low dose exposures to carcinogens, such as N-nitrosodimethylamine, adduct levels in liver correlate closely with tumour incidence. In all likelihood, such relationships need to be established for each exposure and, in order to be relevant to human risk assessment, need to take into account factors such as DNA repair and mutagenic efficiency. Finally, in order to estimate the increase in cancer attributable to a given level of external exposure, it is clearly important to establish background levels of corresponding DNA damage so that the scale of the incremental increase can be calculated.  相似文献

Explaining cooperation is one of the greatest challenges for evolutionary biology. It is particularly a problem in species such as humans, where there is cooperation between nonrelatives. Numerous possible solutions have been suggested for the problem of cooperation between nonrelatives, including punishment, policing, and various forms of reciprocity. Here, we suggest that local competition for resources can pose a problem for these hypotheses, analogous to how it can select against cooperation between relatives. We extend the prisoner's dilemma (PD) game to show that local competition between interacting individuals can reduce selection for cooperation between nonrelatives. This is because, with local competition, fitness is relative to social partners, and cooperation benefits social partners. We then test whether nonrelated humans adjust their level of cooperation facultatively in response to the scale of competition when playing the PD for cash prizes. As predicted, we found that individuals were less likely to cooperate when competition was relatively local. Cooperation between humans will therefore be most likely when repeated interactions take place on a local scale between small numbers of people, and competition for resources takes place on a more global scale among large numbers of people.  相似文献

Female mating rate is an important variable for understanding the role of females in the evolution of mating systems. Polyandry influences patterns of sexual selection and has implications for sexual conflict over mating, as well as for wider issues such as patterns of gene flow and levels of genetic diversity. Despite this, remarkably few studies of insects have provided detailed estimates of polyandry in the wild. Here we combine behavioural and molecular genetic data to assess female mating frequency in wild populations of the two-spot ladybird, Adalia bipunctata (Coleoptera: Coccinellidae). We also explore patterns of sperm use in a controlled laboratory environment to examine how sperm from multiple males is used over time by females, to link mating with fertilization. We confirm that females are highly polyandrous in the wild, both in terms of population mating rates (approximately 20% of the population found in copula at any given time) and the number of males siring offspring in a single clutch (three to four males, on average). These patterns are consistent across two study populations. Patterns of sperm use in the laboratory show that the number of mates does not exceed the number of fathers, suggesting that females have little postcopulatory influence on paternity. Instead, longer copulations result in higher paternity for males, probably due to the transfer of larger numbers of sperm in multiple spermatophores. Our results emphasize the importance of combining field and laboratory data to explore mating rates in the wild.  相似文献

It is now widely accepted that post-zygotic reproductive isolation is the result of negative epistatic interactions between derived alleles fixed independently at different loci in diverging populations (the Dobzhansky-Muller model). What is less clear is the nature of the loci involved and whether the derived alleles increase in frequency through genetic drift, or as a result of natural or sexual selection. If incompatible alleles are fixed by selection, transient polymorphisms will be rare and clines for these alleles will be steep where divergent populations meet. If they evolve by drift, populations are expected to harbour substantial genetic variation in compatibility and alleles will introgress across hybrid zones once they recombine onto a genetic background with which they are compatible. Here we show that variation in male sterility in a naturally occurring Chorthippus parallelus grasshopper hybrid zone conforms to the neutral expectations. Asymmetrical clines for male sterility have long tails of introgression and populations distant from the zone centre show significant genetic variation for compatibility. Our data contrast with recent observations on 'speciation genes' that have diverged as a result of strong natural selection.  相似文献

Environmental factors play an integral role, either directly or indirectly, in structuring faunal assemblages. Water chemistry, predation, hydroperiod and competition influence tadpole assemblages within waterbodies. We surveyed aquatic predators, habitat refugia, water height and water chemistry variables (pH, salinity and turbidity) at 37 waterbodies over an intensive 22‐day field survey to determine which environmental factors influence the relative abundance and occupancy of two habitat specialist anuran tadpole species in naturally acidic, oligotrophic waterbodies within eastern Australian wallum communities. The majority of tadpoles found were of Litoria olongburensis (wallum sedge frog) and Crinia tinnula (wallum froglet) species, both habitat specialists that are associated with wallum waterbodies and listed as Vulnerable under the IUCN Red List. Tadpoles of two other species (Litoria fallax (eastern sedge frog), and Litoria cooloolensis (cooloola sedge frog)) were recorded from two waterbodies. Tadpoles of Litoria gracilenta (graceful treefrog) were recorded from one waterbody. Relative abundance and occupancy of L. olongburensis tadpoles were associated with pH and water depth. Additionally, L. olongburensis tadpole relative abundance was negatively associated with turbidity. Waterbody occupancy by C. tinnula tadpoles was negatively associated with predatory fish and water depth and positively associated with turbidity. Variables associated with relative abundance of C. tinnula tadpoles were inconclusive and further survey work is required to identify these environmental factors. Our results show that the ecology of specialist and non‐specialist tadpole species associated with ‘unique’ (e.g. wallum) waterbodies is complex and species specific, with specialist species likely dominating unique habitats.  相似文献

Niche construction refers to the activities of organisms that bring about changes in their environments, many of which are evolutionarily and ecologically consequential. Advocates of niche construction theory (NCT) believe that standard evolutionary theory fails to recognize the full importance of niche construction, and consequently propose a novel view of evolution, in which niche construction and its legacy over time (ecological inheritance) are described as evolutionary processes, equivalent in importance to natural selection. Here, we subject NCT to critical evaluation, in the form of a collaboration between one prominent advocate of NCT, and a team of skeptics. We discuss whether niche construction is an evolutionary process, whether NCT obscures or clarifies how natural selection leads to organismal adaptation, and whether niche construction and natural selection are of equivalent explanatory importance. We also consider whether the literature that promotes NCT overstates the significance of niche construction, whether it is internally coherent, and whether it accurately portrays standard evolutionary theory. Our disagreements reflect a wider dispute within evolutionary theory over whether the neo‐Darwinian synthesis is in need of reformulation, as well as different usages of some key terms (e.g., evolutionary process).  相似文献
