The AMP-activated protein kinase (AMPK) is an alphabetagamma heterotrimer that is activated by low cellular energy status and affects a switch away from energy-requiring processes and toward catabolism. While it is primarily regulated by AMP and ATP, high muscle glycogen has also been shown to repress its activation. Mutations in the gamma2 and gamma3 subunit isoforms lead to arrhythmias associated with abnormal glycogen storage in human heart and elevated glycogen in pig muscle, respectively. A putative glycogen binding domain (GBD) has now been identified in the beta subunits. Coexpression of truncated beta subunits lacking the GBD with alpha and gamma subunits yielded complexes that were active and normally regulated. However, coexpression of alpha and gamma with full-length beta caused accumulation of AMPK in large cytoplasmic inclusions that could be counterstained with anti-glycogen or anti-glycogen synthase antibodies. These inclusions were not affected by mutations that increased or abolished the kinase activity and were not observed by using truncated beta subunits lacking the GBD. Our results suggest that the GBD binds glycogen and can lead to abnormal glycogen-containing inclusions when the kinase is overexpressed. These may be related to the abnormal glycogen storage bodies seen in heart disease patients with gamma2 mutations.  相似文献

用0℃冷冻处理2—3 h,一些PcaSE-1和BEL-7404细胞的角蛋白纤维能部分地转化成凝聚颗粒,但在HeLa 和CNE 细胞中不发生这种角蛋白纤维结构转化。当回复温度到37℃15—30 min 时,PcaSE-1 和BEL-7404细胞的这种结构转化能快速回复。相反,在HeLa 和CNE 细胞有丝分裂时,角蛋白纤维能转化成凝聚颗粒,但PcaSE-1细胞和BEL-7404细胞的角蛋白纤维网始终维持纤维状态,且围绕纺锤体分布。上述结果表明:两类上皮细胞角蛋白纤维结构的转化似由不同因子所引起。我们的结果还指出:(1)单用秋水仙素或用秋水仙素和细胞松弛素D 合并处理PcaSE-1细胞不能引起角蛋白纤维凝聚。但经秋水仙素解聚微管后,会增强细胞对冷处理的凝聚反应。(2)冷处理时角蛋白纤维凝聚的形成与细胞是否具有两套不同的中间纤维无关。(3)予先用TritonX-100抽提细胞,角蛋白纤维在冷冻后不能转化成凝聚颗粒。(4)冷冻处理引起的结构转化可能是某些上皮细胞系的角蛋白纤维的一种特殊性质。  相似文献

利用1976年Landsat MSS、1989年Landsat TM、2000年Landsat ETM+和2010年TM遥感影像,运用GIS和景观生态学方法,分析了1976-2010年疏勒河中游玉门市土地利用/覆被和景观格局的变化.结果表明:1976-2010年间,玉门市土地利用类型转移的主要方向是草地和戈壁转化为耕地、耕地转化为建设用地、草地转化为戈壁;土地利用变化经历了“缓慢变化-急剧变化-显著变化”的过程,景观密度持续增大,最大斑块指数先增大后减小,面积加权形状指数增大,形状趋于不规则;斑块间的最邻近距离减小,景观逐渐向具有多种要素的密集格局演变,更加破碎;不同斑块间的分离度减小;景观的多样性和均匀度先减小后增加.农业人口增长和经济发展是研究区土地利用/覆被变化的最直接驱动力,气候和政策因素也是重要的影响因素.  相似文献

Background

Recently, several studies have demonstrated that two long non-coding RNAs (lncRNAs), HULC and MALAT1, may participate in hepatocellular carcinoma (HCC) development and progression. However, genetic variations in the two lncRNAs and their associations with HCC susceptibility have not been reported. In this study, we hypothesized that single nucleotide polymorphisms (SNPs) in HULC and MALAT1 may contribute to HCC risk.

Methods

We conducted a case-control study and genotyped two SNPs, rs7763881 in HULC and rs619586 in MALAT1, in 1300 HBV positive HCC patients, 1344 HBV persistent carriers and 1344 subjects with HBV natural clearance to test the associations between the two SNPs and susceptibility to HCC and HBV chronic infection.

Results

The variant genotypes of rs7763881 were significantly associated with decreased HCC risk in a dominant genetic model [AC/CC vs. AA: adjusted odds ration (OR)  =  0.81, 95% confidence intervals (CIs)  =  0.68–0.97, P  =  0.022]. Furthermore, the variant genotypes of rs619586 was associated with decreased HCC risk with a borderline significance (AG/GG vs. AA: adjusted OR  =  0.81, 95% CIs  =  0.65–1.01, P  =  0.057). However, no significant association was found between the two SNPs and HBV clearance.

Conclusions

The variant genotypes of rs7763881 in HULC may contribute to decreased susceptibility to HCC in HBV persistent carriers.  相似文献

Most of the maize kernel oil is located in the embryo while the majority of starch is located in the endosperm. Maize kernel composition and value are affected significantly by the ratio of the embryo size to the endosperm size; however, the genetic regulation of embryo to endosperm ratio (EER) in maize is unknown. Here we identified ZmGE2 gene, which encodes a cytochrome p450 protein, as a gene associated with EER variation in maize. We first expressed rice Giant Embryo (GE) gene driven by oleosin promoter in maize and detected a 23.2?% reduction in EER in transgenic seeds, demonstrating the existence of evolutionarily conserved mechanisms for EER determination in rice and maize. We next identified maize GE2, a homolog of rice GE sharing 70?% identity in amino sequence, as a candidate based on the similar expression pattern and co-localization with a previously detected QTL for EER. Followed by linkage and association mapping, a 247-bp transposable element (TE) insertion in 3′-untranslated region of ZmGE2 gene was identified to be associated with increase in EER and kernel oil content. Expression level of the favorable ZmGE2 allele containing the 247-bp TE insertion was strongly reduced. In addition, the 247-bp TE insertion site was a selection target during the artificial long-term selection for the high EER trait in a high oil population. This is the first report that demonstrates an association of ZmGE2 with EER variation in maize and identifies ZmGE2 gene as a promising target for manipulation of EER and grain composition by either transgenic approach or molecular breeding in maize.  相似文献
