The chlL gene product is involved in the light-independent synthesis of chlorophyll in photosynthetic bacteria, green algae and non-flowering plants. The chloroplast genome of Chlorella vulgaris strain C-27 contains the first example of a split chlL gene, which is interrupted by a 951?bp group I intron in the coding region. In vitro synthesized pre-mRNA containing the entire intron and parts of the flanking exon sequences is able to efficiently self-splice in vitro in the presence of a divalent and a monovalent cation and GTP, to yield the ligated exons and other splicing intermediates characteristic of self-splicing group I introns. The 5′ and 3′ splice sites were confirmed by cDNA sequencing and the products of the splicing reaction were characterized by primer extension analysis. The absence of a significant ORF in the long P9 region (522?nt), separating the catalytic core from the 3′ splice site, makes this intron different from the other known examples of group I introns. Guanosine-mediated attack at the 3′ splice site and the presence of G-exchange reaction sites internal to the intron are some other properties demonstrated for the first time by an intron of a protein-coding plastid gene.  相似文献

Multielement analysis was performed on human milk collected on 5-9-d postpartum from 51 Japanese females using inductively coupled plasma (ICP) mass spectrometry (MS), ICP atomic emission spectrometry (ICP-AES) and fluorometry. Thirty-one elements were detected by these analytical methods in milk. Twelve elements (Na, Mg, P, S, K, Ca, Cu, Zn, Se, Sr, Rb, and Mo) were detected in all of the samples. Al, Cs, and Ba were the elements detected by ICP-MS in more than half of the samples. Multiple regression analysis extracted biological attributes of mother and infant, such as maternal stature, maternal wt, or infant's birth wt, as statistically significant factors contributing to the variation in elemental concentration in milk. However, the rates of contribution were small in all cases. It was concluded that the biological attributes of mother and infant examined in this study were not the major factors that contribute to elemental variation in human milk.  相似文献

Expression of 4C9, a Lex[Gal beta 1----4(Fuc alpha 1----3)GlcNAc] antigen, during mouse embryogenesis was studied by immunohistochemical methods. Distribution of 4C9 was similar to, but not identical with that of SSEA-1 (stage-specific embryonic antigen-1). Notably, 4C9 was detected in some of the inner cell mass cells of late blastocysts, ectoderm cells migrating from the primitive streak to the mesoderm space and primordial germ cells just formed from the migrating cells. Thus, 4C9 was considered to be continuously expressed in the cell lineage starting at the totipotent 8 cell stage and leading to primordial germ cells. While 4C9 gradually decreased from the surface of primordial germ cells after they have settled in the gonad, the antigen remained in cytoplasmic granules for some period in a sex determined manner. In male gonads, cytoplasmic granules positive for 4C9 tended to be polarized to one side of cytoplasm. The 4C9 reactive material completely disappeared from male germ cells by day 16 of gestation. In female gonads, granules scattered throughout the cytoplasm and cell surface were positive for 4C9. On day 16 of gestation the cell surface antigenicity was lost, but some cytoplasmic antigenicity still remained. As above, 4C9 is a reliable marker to study the origin, migration and differentiation of primordial germ cells, and to distinguish male and female germ cells. By immunoelectron microscopy, 4C9 was detected at the plasma membrane, the Golgi apparatus, and dense-cored vesicles in primordial germ cells on 10-11 days of gestation.  相似文献

Although the myelin-associated glycoprotein (MAG) cannot be detected in primary cultures of rat Schwann cells in the absence of neurons, MAG expression was demonstrated in some lines of cultured Schwann cells that had been immortalized by repetitive passaging. Radioimmunoassay of one such Schwann cell line, S-16, showed a remarkably high MAG concentration of about 1 ng/microgram of total protein, a level that is comparable to the MAG concentration in adult sciatic nerve. The S-16 cells divide very rapidly, are rounder than normal Schwann cells, and elaborate many processes after reaching high density. The cells are galactocerebroside positive, but express little or no P0 glycoprotein or myelin basic protein. As in nerve, the MAG synthesized by the cultured cells is primarily the shorter isoform (S-MAG). Furthermore, the posttranslational processing resembles that occurring in vivo including a similar degree of glycosylation, sulfation of oligosaccharides, and phosphorylation of the polypeptide. The sensitivity of MAG to treatment of the intact cells with trypsin or neuraminidase, as well as surface labeling with [3H]borohydride reduction after periodate oxidation, demonstrated that most of the MAG expressed by the S-16 cells is located on the cell surface. This line of immortalized Schwann cells expressing a remarkably high level of MAG should be useful for investigating the cell biology and function of this glycoprotein.  相似文献

The morphology of the excretory system of Hysterothylacium haze was examined by serial histological sections. The excretory system was H-shaped and glandular, consisting of lateral filaments and a commissure, with the exretory pore opening posterior to the nerve ring. A large excretory nucleus was present in the left filament. The cuticularized excretory duct was confined to the left side of the commissure. The glandular excretory system is rare among the Raphidascaridinae.  相似文献

Neuropeptide Y (NPY)- and somatostatin (SS)-like immunoreactivities (LI) were investigated in tumor tissues of one ganglioneuroma (GN), 3 ganglioneuroblastomas (GNB) and one neuroblastoma (NB) by radioimmunoassay. NPY-LI was detected from all 5 tumor tissues (16.4-1247 pmol/g wet tissue). Sephadex G-50 column chromatography and reverse phase high performance liquid chromatography (HPLC) revealed that most of the NPY-LI in tumor extracts was eluted in an identical position to synthetic human NPY except one GNB (case 2). In this case, most of the NPY-LI was eluted in a higher molecular weight region than synthetic human NPY in Sephadex G-50 column chromatography and in a more hydrophobic position in HPLC. SS-LI was detected from 4 tumor extracts except one GNB (case 2) (21.3-787 pmol/g wet tissue). Sephadex G-25 column chromatography and reverse phase HPLC revealed that SS-LI in tumor extracts was eluted just after the void volume and then in the same positions as SS-28 and SS-14. These results suggest that NPY, SS-14 and SS-28 exist in tumor tissues of GN, GNB and NB, and most of the NPY-LI in one GNB was a higher molecular and more hydrophobic form of NPY-LI.  相似文献

2-amino-5-carboxymuconic 6-semialdehyde is an unstable intermediate in the meta-cleavage pathway of 4-amino-3-hydroxybenzoic acid in Bordetella sp. strain 10d. In vitro, this compound is nonenzymatically converted to 2,5-pyridinedicarboxylic acid. Crude extracts of strain 10d grown on 4-amino-3-hydroxybenzoic acid converted 2-amino-5-carboxymuconic 6-semialdehyde formed from 4-amino-3-hydroxybenzoic acid by the first enzyme in the pathway, 4-amino-3-hydroxybenzoate 2,3-dioxygenase, to a yellow compound (epsilonmax = 375 nm). The enzyme in the crude extract carrying out the next step was purified to homogeneity. The yellow compound formed from 4-amino-3-hydroxybenzoic acid by this purified enzyme and purified 4-amino-3-hydroxybenzoate 2,3-dioxygenase in a coupled assay was identified as 2-hydroxymuconic 6-semialdehyde by GC-MS analysis. A mechanism for the formation of 2-hydroxymuconic 6-semialdehyde via enzymatic deamination and nonenzymatic decarboxylation is proposed based on results of spectrophotometric analyses. The purified enzyme, designated 2-amino-5-carboxymuconic 6-semialdehyde deaminase, is a new type of deaminase that differs from the 2-aminomuconate deaminases reported previously in that it primarily and specifically attacks 2-amino-5-carboxymuconic 6-semialdehyde. The deamination step in the proposed pathway differs from that in the pathways for 2-aminophenol and its derivatives.  相似文献

Prostaglandin F levels were determined in monkey uterine fluid collected daily from a silicone tubing collection system surgically installed in adult female rhesus monkeys. Sampling was obtained from intact and ovariectomized — hormone treated monkeys. During the primate menstrual cycle, uterine fluid prostaglandin F levels showed a cyclic pattern in concentration with highest values recorded during the 18–20th day of the cycle. Estrogen treatment to the ovariectomized female elicited a striking and marked increase in uterine fluid prostaglandin F levels while progesterone treatment had little effect. These results suggest that the presence of uterine fluid prostaglandin F is estrogen dependent and may be causely related to the control of menstrual cyclicity.  相似文献
