The medaka fish albino mutant, i1 is one of the Tomita collection of medaka pigmentation mutants which exhibits a complete albino phenotype, because of inactivation of the tyrosinase gene due to insertion of a transposable element, Tol‐1. Recently, mosaic black‐pigmented i1 medaka fish have arisen in one of our laboratory breeding populations. Their pigmented cells have been observed in all of the tissues, including the eye and skin, in which melanin is detectable in the wild type. In this study, we analyzed the tyrosinase gene of revertants and showed Tol‐1 to have been precisely excised from the gene, suggesting a causal relationship. Mosaic patterns of pigmentation indicate spontaneous somatic excision of the element from the tyrosinase gene. To our knowledge, this is the first transposable element with somatic excision activity demonstrated phenotypically in vertebrates. The pattern of pigmentation in mosaic revertants indicates frequencies of melanin pigments to be consistent with the numbers of melanophores per unit area of body sites, such as the eyes, head and dorsal trunk.  相似文献

The effects of endurance training on the expression of myosin were electrophoretically analyzed in the deep portion of vastus lateralis muscle from the rat. A 10-wk running program led to increases (P < 0.01) in myosin heavy chain (MHC) 2a and 2d with a decrease (P < 0.01) in MHC(2b). Training also evoked a rearrangement of the isomyosin pattern with decreases in fast isomyosin (FM) 1 (P < 0.01) and FM2 (P < 0.05) and a rise in intermediate isomyosin (P < 0.01). These changes were accompanied by a 61% decrease (P < 0.01) in myosin light chain (MLC) 3F (11.8 +/- 2.7 vs. 4.6 +/- 4.2%). Two-dimensional electrophoresis made it possible to separate the triplet of isomyosins (FMb) consisting of MHC(2b). Training elicited a 26% decrease (P < 0.05) in the FM1b fraction within FMb, i.e., FM1b/(FM1b + FM2b + FM3b) (24.2 +/- 5.5 vs. 18.0 +/- 4.3%). These changes resulted in a 10% decrease (P < 0.05) in the MLC(3F) fraction, i.e., MLC(3F)/(MLC(1F) + MLC(3F)), in FMb (44.9 +/- 4.5 vs. 40.3 +/- 3.2%). These results suggest that endurance training may exert the depressive effect on the contractile velocity of type IIB fibers and that a training-induced decrease in the contractile velocity of whole muscle may be caused by alterations in fast alkali MLC complements within a given fiber type as well as by transitions in MHC-based fiber populations.  相似文献

Five clones that hybridized weakly with the human retinoblastoma (Rb) gene were obtained by screening a human genomic library in a non-stringent condition. The DNAs of two of these clones were partially sequenced and found to contain a region with considerable homology to part of the Rb gene. These two clones were found to contain L1 family repeating sequences. This finding is discussed in relation with possible functions of the L1 family. As the L1 family is transposable, the Rb gene may be inactivated by recombination at this homologous region. Another possibility related with the DNA binding properties of Rb and L1 family proteins is also discussed.  相似文献

Across the globe, producers are moving from individual housing to group housing for sows during gestation. Producers typically group sows of a range of parities together, although the impacts are largely unknown. This study examined the behavioral expression at mixing for young, midparity, and older sows. Ten mixed-parity groups were filmed at mixing on a commercial piggery. One-minute clips were edited from continuous footage where focal sows of known parity could be identified, and scored for qualitative behavioral expression. Parity 2 and 6 sows were more calm/tired than Parity 4 sows, who were more active/energetic. Parity 2 sows were more curious/inquisitive than Parity 4 and 6 sows, who were more anxious/frustrated. Correlations between qualitative behavioral expression and activity indicated sows scored as more calm/tired spent a greater proportion of time standing, while sows scored as more active/energetic spent more time performing avoidance behavior. Different body language is likely to reflect physical or affective differences in how sows cope with mixing.  相似文献
