A central feature of the adeno-associated virus (AAV) latent life cycle is persistence in the form of both integrated and episomal genomes. However, the molecular processes associated with episomal long-term persistence of AAV genomes are only poorly understood. To investigate these mechanisms, we have utilized a recombinant AAV (rAAV) shuttle vector to identify circular AAV intermediates from transduced HeLa cells and primary fibroblasts. The unique structural features exhibited by these transduction intermediates included circularized monomer and dimer virus genomes in a head-to-tail array, with associated specific base pair alterations in the 5′ viral D sequence. In HeLa cells, the abundance and stability of AAV circular intermediates were augmented by adenovirus expressing the E2a gene product. In the absence of E2a, adenovirus expressing the E4 open reading frame 6 gene product decreased the abundance of AAV circular intermediates, favoring instead the linear replication form monomer (Rfm) and dimer (Rfd) structures. In summary, the formation of AAV circular intermediates appears to represent a new pathway for AAV genome conversion, which is consistent with the head-to-tail concatemerization associated with latent-phase persistence of rAAV. A better understanding of this pathway may increase the utility of rAAV vectors for gene therapy.  相似文献

Antifreeze proteins have the ability to bind to ice with high affinity and inhibit further crystal growth. The insect antifreeze protein from spruce budworm exhibits very high thermal hysteresis activity and is implicated in the protection of overwintering larvae from freezing. This protein has been crystallized in 20-25% polyethylene glycol (Mr 6000), 0.4 M NaCl, 0.1 M Tris-HCl, pH 8.5, by vapor diffusion using the hanging drop method. The resulting crystals are very thin (typically <0.01 mm in the shortest dimension), and only after repeated seeding could crystals be grown large enough for data collection using synchrotron radiation. The crystals belong to the monoclinic space group C2, with cell dimensions a = 82.28 A, b = 62.29 A, c = 63.63 A, and beta = 113.7 degrees. Molecules in the asymmetric unit are related by a twofold axis of symmetry with two molecules present. Native data to a resolution of 2.6 A have been collected with 90.3% completeness and a Rsym of 6.9%.  相似文献

目的: 验证临床受试者所完成的心肺运动试验(CPET)为最大极限运动,进一步设计完善Max试验验证CPET结果客观定量功能评估的准确性及以某特定指标的特定数值作为停止运动的标准是否可行。方法: 选择2017年9月至2019年1月在阜外医院签署知情同意书后进行CPET和Max试验受试者216例。其中正常受试者41例,因CPET峰值呼吸交换率(RER)≤1.10,或运动中心率和血压不上升,对CPET极限运动结果存在质疑的临床患者175例进行研究。其中60例已初步报告,本研究进一步扩大研究。Max试验方法:完成CPET测试后,先蹬车≥60 r/min,再施加130%峰值功率的恒定功率,鼓励受试者运动至不能坚持的极限状态。计算分析Max试验30 s的最大心率和最大摄氧量、及其与峰值心率和峰值摄氧量之间的差值和百分差值。百分差值=(Max值-峰值值)/Max值× 100%。评测标准:①若心率和摄氧量任一指标的差值百分比≤-10%(Max测试的数值低于CPET峰值数据)则定义Max试验操作失败,否则为成功;2若心率和摄氧量的差值百分比均在-10%~10%,则Max试验操作成功,证明CPET数据为极限运动,CPET 峰值相关数据较为准确;③若心率和摄氧量差值任一指标差值百分比≥10%时,则Max试验操作成功,证明CPET结果为非极限运动。结果: 病例组峰值摄氧量(L/min、ml/(min·kg)、%pred)、无氧阈(L/min、ml/(min·kg)、%pred)、峰值氧脉搏(ml/beat、%pred)、峰值RER、峰值收缩压(mmHg)、峰值运动负荷(W/min)、峰值心率(bpm)、摄氧有效性峰值平台(OUEP)(比值、%pred)低于正常组,二氧化碳通气有效性平均90 s最低值(Lowest Ve/VCO2)(比值、%pred)、二氧化碳通气效率斜率(Ve/VCO2 Slope)(比值、%pred)高于正常组(P<0.05)。所有正常组与病例组均安全无任何事件完成CPET和Max试验。216例受试者中,Max试验成功198例(91.7%),其中证明CPET为极限运动182例,为非极限运动16例;失败18例(8.3%)。结论: 在临床检查中,若对CPET结果是否为最大极限存在质疑,利用Max试验可验证CPET是否为极限运动。Max试验方法安全可行,值得进一步深入研究和临床推广应用。  相似文献

Pyruvate phosphate dikinase (PPDK) catalyzes the reversible reaction: ATP + P(i) + pyruvate <--> AMP + PP(i) + PEP using Mg2+ and NH4+ ions as cofactors. The reaction takes place in three steps, each mediated by a carrier histidine residue located on the surface of the central domain of this three-domain enzyme: (1) E-His + ATP <--> E-His-PP.AMP, (2) E-His-PP.AMP + P(i) <--> E-His-P + AMP + PP(i), (3) E-His-P + pyruvate <--> E-His + PEP. The first two partial reactions are catalyzed at an active site located on the N-terminal domain, and the third partial reaction is catalyzed at an active site located on the C-terminal domain. For catalytic turnover, the central domain travels from one terminal domain to the other. The goal of this work is to determine whether the two connecting linkers direct the movement of the central domain between active sites during catalytic turnover. The X-ray crystal structure of the enzyme suggests interaction between the two linkers that may result in their coordinated movement. Mutations were made at the linkers for the purpose of disrupting the linker-linker interaction and, hence, synchronized linker movement. Five linker mutants were analyzed. Two of these contain 4-Ala insertions within the solvated region of the linker, and three have 3-residue deletions in this region. The efficiencies of the mutants for catalysis of the complete reaction as well as the E-His + ATP <--> E-His-PP.AMP partial reaction at the N-terminal domain and the E-His + PEP <--> E-His-P + pyruvate reaction at the C-terminal domain were measured to assess linker function. Three linker mutants are highly active catalysts at both active sites, and the fourth is highly active at one site but not the other. These results are interpreted as evidence against coordinated linker movement, and suggest instead that the linkers move independently as the central domain travels between active sites. It is hypothesized that while the linkers play a passive role in central domain-terminal domain docking, their structural design minimizes the conformational space searched in the diffusion process.  相似文献

摘要:【目的】研究温度以及返回液添加量对体系中氧化效果以及金(Au)氰化浸出率的影响。【方法】针对某高硫高砷金精矿建立了一套连续生物预氧化体系(氧化时间6 d),比较不同的温度(40℃和45℃)和返回液添加量(0和600mL)对生物预氧化效果的影响:间隔8 h连续测定氧化体系中的氧化液、氧化渣以及氰化渣中的总Fe、总S、总As以及SO4 2-和Au含量;并用16S rRNA基因克隆文库的方法对体系中的浸矿菌群结构进行分析。【结果】结果显示,提高温度和添加氧化返回液都会改变体系:升高温度在一定程度上提高了体系的氧化程度,主要表现为显著降低了尾渣中总铁、总硫含量,显著提高了尾渣中的S6 + 比例,并且降低前三槽氧化体系的耗碱量;投加返回液则会降低体系的氧化程度,主要表现为显著提高了尾渣金品位和铁含量,显著降低了尾渣中的S6 + 以及Au 浸出率;Dissimilarity和DCA结果表明温度变化对体系的影响更为显著;氧化过程中的Au 浸出率、硫氧化率以及As 去除率,三者互为显著正相关;克隆文库结果显示,该浸矿菌群主要包括3类细菌: Acidithiocacillus caldu (71%)、Leptospirillum ferriphilum (23%)和Sulfobacillus thermosulfidooxidans(6%),基于97%相似性的OTU覆盖率达93.67%。【结论】研究结果表明,相对于添加返回液,温度变化会更显著的改变体系反应效率。提高体系温度有利于矿物的氧化,而添加返回液会降低Au浸出率。本文首次研究了连续生物预氧化过程中温度梯度和返回液添加对体系的影响,本研究结果对中温生物预氧化工业生产的工艺优化、降低工业运行成本具有重要的指导意义。  相似文献

Horseweed (Conyza canadensis) is a problem weed in crop production because of its evolved resistance to glyphosate and other herbicides. Although horseweed is mainly self-pollinating, glyphosate-resistant (GR) horseweed can pollinate glyphosate-susceptible (GS) horseweed. To the best of our knowledge, however, there are no available data on horseweed pollen production, dispersion, and deposition relative to gene flow and the evolution of resistance. To help fill this knowledge gap, a 43-day field study was performed in Champaign, Illinois, USA in 2013 to characterize horseweed atmospheric pollen emission, dispersion, and deposition. Pollen concentration and deposition, coupled with atmospheric data, were measured in a source field (180 m by 46 m) and its surrounding areas up to 1 km downwind horizontally and up to 100 m vertically. The source strength (emission rate) ranged from 0 to 140 pollen grains per plant per second (1170 to 2.1×106 per plant per day). For the life of the study, the estimated number of pollen grains generated from this source field was 10.5×1010 (2.3×106 per plant). The release of horseweed pollen was not strongly correlated to meteorological data and may be mainly determined by horseweed physiology. Horseweed pollen reached heights of 80 to100 m, making long-distance transport possible. Normalized (by source data) pollen deposition with distance followed a negative-power exponential curve. Normalized pollen deposition was 2.5% even at 480 m downwind from the source edge. Correlation analysis showed that close to or inside the source field at lower heights (≤3 m) vertical transport was related to vertical wind speed, while horizontal pollen transport was related to horizontal wind speed. High relative humidity prevented pollen transport at greater heights (3–100 m) and longer distances (0–1000 m) from the source. This study can contribute to the understanding of how herbicide-resistance weeds or invasive plants affect ecology through wind-mediated pollination and invasion.  相似文献
