Constructing functional materials on a 3D host is an efficient strategy to tackle issues of lithium (Li) metal anodes. Although non-Li-alloying materials provide structural stability during cycling due to reduced lattice distortions, low lithiophilicity and sluggish mass transport kinetics limit their functionality. Herein, a synergistic strategy is proposed to improve intrinsic lithiophilicity and mass transport kinetics of non-Li-alloying nucleation sites and demonstrate its remarkable efficacy. Two carbon fiber (CF) hosts coated by non-Li-alloying nanosheets with and without oxygen-enriched carbon filler (OCF) as lithiophilicity and mass transport booster (OCF-DSC@CF and DSC@CF, respectively) are constructed and their physiochemical properties are systematically evaluated to reveal the efficacy of OCF. By advanced characterization techniques, including 3D tomography and location-dependent electron energy loss spectroscopies, the complex heterostructure of OCF-DSC@CF with distinctive roles of each constituent is clearly identified. As verified by theoretical and electrochemical analyses, the incorporation of OCF endows OCF-DSC@CF with substantially improved lithiophilicity and mass transport kinetics. Moreover, OCF-DSC@CF induces a multifunctional SEI enriched with LiF and LiCx, which exhibits well-balanced electrical resistivity and ionic conductivity. Benefiting from these attributes, OCF-DSC@CF exhibits an unprecedented cyclability under a low N/P ratio of 1.8, achieving 700 cycles at 0.5C with an exceptional capacity retention of 97.8%.  相似文献

Amylin cosecretes with insulin from pancreatic beta-cells and shows high sequence homology with CGRP, adrenomedullin, and salmon calcitonin. This study aimed to investigate the effect of amylin on the atrial hemodynamics and ANP release from rat atria and to identify its receptor subtypes. Isolated perfused left atria from either control or streptozotocin-treated rats were paced at 1.3 Hz. The concentration of ANP was measured by radioimmunoassay and the translocation of ECF was measured by [3H]-inulin clearance. Rat amylin increased atrial contractility and suppressed the release of ANP. Rat CGRP showed similar effects but was approximately 300-fold more potent than amylin. Pretreatment with receptor antagonist for CGRP1 [rat alpha-CGRP (8-37)] or salmon calcitonin [acetyl-(Asn30, Tyr32)-calcitonin(8-32), (AC 187)] blocked the suppressive effect of ANP release and the positive inotropic effect by rat amylin. However, receptor antagonists for amylin [amylin (8-37), acetyl-amylin] did not block those effects. Amylin (8-37), acetyl-amylin, or rat alpha-CGRP (8-37) alone accentuated the release of ANP with no changes in atrial contractility. The effect of rat amylin and rat amylin (8-37) on the ANP release was attenuated in streptozotocin-treated rats. We suggest that amylin suppressed ANP release with increased atrial contractility through receptors for CGRP1 and salmon calcitonin and the attenuation of amylin and its antagonist on ANP release from streptozotocin-treated rat atria may be due to the downregulation of amylin receptor.  相似文献

Epidermal growth factor (EGF) is a member of a structurally related family containing heparin-binding EGF-like growth factor (HB-EGF) and transforming growth factor alpha (TGFalpha) that exerts neurotrophic activity on midbrain dopaminergic neurons. To examine neurotrophic abnormality in Parkinson's disease (PD), we measured the protein content of EGF, TGFalpha, and HB-EGF in post-mortem brains of patients with Parkinson's disease and age-matched control subjects. Protein levels of EGF and tyrosine hydroxylase were decreased in the prefrontal cortex and the striatum of patients. In contrast, HB-EGF and TGFalpha levels were not significantly altered in either region. The expression of EGF receptors (ErbB1 and ErbB2, but not ErbB3 or ErbB4) was down-regulated significantly in the same forebrain regions. The same phenomenon was mimicked in rats by dopaminergic lesions induced by nigral 6-hydroxydopamine infusion. EGF and ErbB1 levels in the striatum of the PD model were markedly reduced on the lesioned side, compared with the control hemisphere. Subchronic supplement of EGF in the striatum of the PD model locally prevented the dopaminergic neurodegeration as measured by tyrosine hydroxylase immunoreactivity. These findings suggest that the neurotrophic activity of EGF is maintained by afferent signals of midbrain dopaminergic neurons and is impaired in patients with Parkinson's disease.  相似文献

A series of 7-alkoxyl-4,5-dihydro-[1,2,4]triazolo[4,3-a]quinoline derivatives was synthesized using 6-hydroxy-3,4-dihydro-1H-quinolin-2-one as a starting material. Their anticonvulsant activities were evaluated by the maximal electroshock test (MES test) and the subcutaneous (s.c.) pentylenetetrazol test (scMet test), and their neurotoxicity was evaluated by the rotarod neurotoxicity test (Tox). MES and scMet tests show that 7-(4-fluorobenzyloxy)-4,5-dihydro-[1,2,4]triazolo[4,3-a]quinoline 4l was found to be the most potent with ED50 value of 11.8 and 6.7 mg kg(-1) and protective index (PI = TD50/ED50) value of 4.6 and 8.1, respectively.  相似文献

We examined cell killing and chromatid damage in primary human bronchial epithelial cells irradiated with high-energy 56Fe ions. Cells were irradiated with graded doses of 56Fe ions (1 GeV/nucleon) accelerated with the Alternating Gradient Synchrotron at Brookhaven National Laboratory. The survival curves for cells plated 1 h after irradiation (immediate plating) showed little or no shoulder. However, the survival curves for cells plated 24 h after irradiation (delayed plating) had a small initial shoulder. The RBE for 56Fe ions compared to 137Cs gamma rays was 1.99 for immediate plating and 2.73 for delayed plating at the D10. The repair ratio (delayed plating/immediate plating) was 1.67 for 137Cs gamma rays and 1.22 for 56Fe ions. The dose-response curves for initially measured and residual chromatid fragments detected by the Calyculin A-mediated premature chromosome condensation technique showed a linear response. The results indicated that the induction frequency for initially measured fragments was the same for 137Cs gamma rays and 56Fe ions. On the other hand, approximately 85% of the fragments induced by 137Cs gamma rays had rejoined after 24 h of postirradiation incubation; the corresponding amount for 56Fe ions was 37%. Furthermore, the frequency of chromatid exchanges induced by gamma rays measured 24 h after irradiation was higher than that induced by 56Fe ions. No difference in the amount of chromatid damage induced by the two types of radiations was detected when assayed 1 h after irradiation. The results suggest that high-energy 56Fe ions induce a higher frequency of complex, unrepairable damage at both the cellular and chromosomal levels than 137Cs gamma rays in the target cells for radiation-induced lung cancers.  相似文献

本研究采用电镜及酶细胞化学的方法观察了鸡胚脾脏不同胚龄组巨噬细胞溶酶体酸性磷酸酶(AcP酶)的变化、凋亡实验组巨噬细胞及其AcP酶与凋亡细胞的关系。取10天、13天和17天鸡胚脾脏,按Gomori法显示AcP酶,各胚龄脾脏巨噬细胞AcP酶细胞化学反应阳性,按AcP酶染色阳性做溶酶体计数,结果显示随着胚龄的增加溶酶体数随之增加,尤以第17天组溶酶体数增加最为明显,所得数据经统计分析表明各胚龄组间溶酶体数的差异有统计学意义。凋亡实验组采用放线菌酮诱导15天鸡胚脾脏细胞凋亡,结果显示凋亡细胞为各类幼稚血细胞,以幼稚淋巴细胞为主。巨噬细胞未见凋亡,而是吞噬了大量的凋亡细胞和凋亡小体,AcP酶反应颗粒不仅出现在巨噬细胞的溶酶体、吞噬体,还见于高尔基复合体、内质网等。细胞AcP酶反应强度数字化结果表明:凋亡组酶活性显著高于对照组,差别有统计学意义,提示胚胎巨噬细胞在凋亡细胞出现时AcP酶活性增强,说明巨噬细胞吞噬和消化凋亡细胞或凋亡小体是通过AcP酶等活性物质来实现的。  相似文献

动物实验发现睾酮能改善干眼病动物模型干眼症状,促进泪腺分泌,但作用机理不明,本研究探讨泪腺细胞中是否存在睾酮受体。雌雄各8只大鼠的泪腺分别制成石蜡切片和超薄切片,睾酮分别采用ABC法及免疫金标记,通过真空负压ABC法光镜观察及免疫金探针超微结构定位进行细胞睾酮受体检查。结果:光镜下泪腺导管上皮细胞的胞浆及胞核中出现免疫染色阳性,泪腺上皮细胞则很少见到;电镜下泪腺细胞胞浆及核中可见金颗粒,对照组则染色阴性。结论 泪腺细胞存在着睾酮受体,睾酮通过受体对泪腺发生作用。  相似文献

HAL1基因是酵母中重要的耐盐基因。以酿酒酵母AS2.375菌株的DNA为模板,根据已发表的序列设计引物,经PCR扩增得到约900 bp的HAL1基因片段,连接到pMD18-T载体上,转化大肠杆菌JM109,筛选重组质粒进行酶切分析和序列测定,结果显示已克隆到完整的可读框,该基因的序列与已知序列同源性达99%。将HAL1基因从T-载体上切下连接到pAM194载体上构建了HAL 1基因的植物表达载体,用于烟草的转化获得了耐盐性提高的转化植株。  相似文献
