粳稻子预44中稻瘟病数量抗性位点分析   总被引:4,自引:2,他引:2

稻瘟病是世界范围内影响水稻(Oryza sativa)生产的主要病害。抗稻瘟病基因的发掘和育种利用是控制稻瘟病经济、环保的有效措施。为了揭示云南地方水稻品种子预44广谱持久抗瘟机制, 利用江南香糯和子预44杂交构建的F7重组自交群体, 采用苗期稻瘟病菌自然诱发接种法, 通过调查田间抗瘟性表型数据, 结合基因型数据对子预44中的数量抗瘟性位点进行了分析。结果表明, 在连锁系数(logarithm of odds, LOD)大于2.0的域值上, 共检测出13个QTLs, 分别位于第1、2、6、8、12号染色体上。不同位点表型贡献值差异较大, 范围为5.8%-21.9%, 其中8号染色体上标记RM72-RM404之间的QTLs可解释约61.9%的表型变异, 很可能为一个主效抗瘟QTL位点。多个位点的主效和微效抗性相结合可能是子预44持久稻瘟病抗性的分子基础。  相似文献

Four endemic Batrachospermaceae species in China, Batrachospermum hongdongense Xie & Feng, Batrachospermum longipedicellatum Hua & Shi, Kumanoa curvata (Shi) Vis et al., and Kumanoa intorta (Jao) Entwisle et al., were analyzed with multilocus data in order to assess their phylogenetic placement. Multilocus molecular data from one nuclear gene (the ribosomal SSU), one chloroplast gene (rbcL), and two mitochondrial genes (COI and the cox2-3 spacer) as well as morphological studies were used to determine the phylogenetic relationships of these endemic species within the Batrachospermales. Batrachospermum hongdongense and B. longipedicellatum had previously been placed in Batrachospermum section Batrachospermum, but based on the data from this study they should be transferred into section Helminthoidea. Kumanoa curvata and K. intorta clearly belong to the newly established genus Kumanoa.  相似文献

Multiplex PCR of microsatellite is a cost effective and high throughput technique of genotyping. We developed a new 12 plex PCR kit for Populus euphratica, the only tree species in desert area ranging from Western China to Mediterranean coast. Three primers were designed for the expressed sequence tags (ESTs) sequences from the NCBI database and the other nine primers were designed based on the EST sequences of Peuphratica obtained by Solexa. The multiplex kit was tested by 96 samples from three natural populations. The results showed sufficient amplification stability and high polymorphism. All the 12 loci used in this kit showed a high transferability (79%) in other seven species from five sections of the genus. The new 12 plex kit combined with the two eight multiplex kits we had developed in previous studies, should be useful to reveal the genetic mechanism and evolution history of the Peuphratica and related species. During the research, we found that primers selection, amplification efficiency, null allele detection are the essential parts of the multiplex kit development.  相似文献

We examined cluster root formation and root exudation by white lupin (Lupinus albus L. cv. Kiev Mutant) in response to growth medium and phosphorus supply in a sand/solution split-root system. The split-root system consisted of a nutrient solution compartment and a siliceous sand compartment. Phosphorus was applied at 1 (low-P plants) or 50 (high-P plants) μM as KH2PO4 to the solution compartment and at 10, 50 or 250 mg P kg−1 as hydroxyapatite (Ca-P) to the sand compartment. In contrast to the high-P plants, P concentration and P uptake in the low-P plants increased with increasing P supply to the sand compartment. The NaHCO3-extractable P was lower in the rhizosphere of the low-P plants than the high-P ones. The proton extrusion rate by the solution-grown roots of the low-P plants was higher than that of the high-P plants at the early growth stage. For the low-P plants, the proportion of dry root biomass allocated to cluster roots was higher in the solution compartment than that in the sand compartment. The citrate exudation increased in the sand compartment and decreased in the solution compartment with time, showing a lack of synchronization in citrate exudation by two root halves grown in different media. The cluster root proportion and citrate exudation in both compartments decreased with increasing shoot P concentration. An additional experiment with no P added to either root compartment showed that the proportion of cluster roots was about 9% lower in the sand than solution compartments. The results suggest that cluster root formation and citrate exudation can be significantly affected by the root growth medium in addition to being regulated by shoot P status. More P can be exploited from sparingly available Ca-P by the low-P plants than the high-P ones due to greater citrate exudation under P deficiency.  相似文献

针对本实验室筛选得到的一株亚硝酸盐氧化细菌(nitrite-oxidizingbacteria),研究了在28℃~30℃、摇床转速为110r/min时,pH、氮源、碳源、NaCl、有机物对菌体生长的影响。结果表明,培养基pH8·0~8·5、NaNO2含量4,500mg/L、Na2CO3含量1·5g/L、NaCl含量0~0·5%、葡萄糖含量0~0·1%时,亚硝酸盐氧化细菌生长良好,培养9d时,细菌浓度可达4·6×109MPN/mL,且培养基中的NO2--N能全部被硝化为NO3--N。培养基中NaCl含量大于0·5%、葡萄糖含量大于0·1%时,亚硝酸盐氧化细菌对氮源的利用受到抑制。亚硝酸盐氧化细菌降解淡水养殖池塘中的NO2--N试验表明,在水温25℃、pH8·6的池塘中,NO2--N从菌体投放后的第3d开始下降,18d后NO2--N由1·47mol/L下降至0·49mol/L。  相似文献

Cell-density-dependent gene regulation by quorum-sensing systems has a crucial function in bacterial physiology and pathogenesis. We demonstrate here that the Staphylococcus aureus agr quorum-sensing regulon is divided into (1) control of metabolism and PSM cytolysin genes, which occurs independently of the small regulatory RNA RNAIII, and (2) RNAIII-dependent control of additional virulence genes. Remarkably, PSM expression was regulated by direct binding of the AgrA response regulator. Our findings suggest that quorum-sensing regulation of PSMs was established before wide-ranging control of virulence was added to the agr regulon, which likely occurred by development of the RNAIII-encoding region around the gene encoding the PSM delta-toxin. Moreover, the agr regulon in the community-associated methicillin-resistant S. aureus MW2 considerably differed from that previously determined using laboratory strains. By establishing a two-level model of quorum-sensing target gene regulation in S. aureus, our study gives important insight into the evolution of virulence control in this leading human pathogen.  相似文献

The 16S rRNA gene approach was applied to investigate the diversity of Planctomycetes in latitudinal surface seawater of the Western Pacific Ocean. The results revealed that the Pirellula-Rhodopirellula-Blastopirellula clade dominated the Planctomycetes community at all surface seawater sites while the minority genera Gemmata and Planctomyces were only found at sites H5 and H2 respectively. Although the clone frequency of the PRB clade seemed stable (between 83.3% and 94.1%) for all surface seawater sites, the retrieved Pirellula-Rhodopirellula-Blastopirellula clade presented unexpected diversity. Interestingly, low latitude seawater appeared to have higher diversity than mid-latitudes. integral-LIBSHUFF software analysis revealed significantly different diversity patterns between in latitudinal surface seawater and in the sediment of South China Sea station M2896. Our data suggested that different hydrological and geographic features contributed to the shift of Planctomycetes diversity in marine environments. This is, to our knowledge, the first systematic assessment of Planctomycetes in latitudinal surface seawater of the open sea and the first comparison of diversity pattern between surface seawater and sediments and has broadened our understanding of Planctomycetes diversity in marine environments.  相似文献
