黏虫Mythimna separata是为害禾本科作物的重大迁飞性害虫。汉江中游流域是湖北小麦等作物的主产区,也是黏虫的常年发生地。2012年至2017年,连续6年在汉江中游湖北省宜城市利用糖醋液、频振式杀虫灯调查了黏虫成虫种群动态。结果表明:宜城地区黏虫成虫可分为4代:越冬代及1代至3代。越冬代成虫峰期出现在2月中下旬至3月底,其中最早始见期为2017年2月14日,最晚为2015年3月10日。1代成虫峰期为5月中旬至6月底,且1代为黏虫累积虫量最多的世代,7月至10月的2代、3代黏虫成虫数量稀少,没有明显的成虫峰期,但2017年7月底至8月初出现较明显2代成虫峰期。汉江中游流域宜城地区属于1代黏虫多发区;1月份温度不足以影响此地黏虫越冬,此地可能为黏虫常年越冬地;在田间变温环境下,夏季高温对宜城黏虫成虫种群无显著抑制作用;黏虫在宜城越冬代及1代成虫量是全国黏虫发生量的重要预警指标。 相似文献

Metformin is currently a strong candidate anti-tumor agent in multiple cancers. However, its anti-tumor effectiveness varies among different cancers or subpopulations, potentially due to tumor heterogeneity. It thus remains unclear which hepatocellular carcinoma (HCC) patient subpopulation(s) can benefit from metformin treatment. Here, through a genome-wide CRISPR-Cas9-based knockout screen, we find that DOCK1 levels determine the anti-tumor effects of metformin and that DOCK1 is a synthetic lethal target of metformin in HCC. Mechanistically, metformin promotes DOCK1 phosphorylation, which activates RAC1 to facilitate cell survival, leading to metformin resistance. The DOCK1-selective inhibitor, TBOPP, potentiates anti-tumor activity by metformin in vitro in liver cancer cell lines and patient-derived HCC organoids, and in vivo in xenografted liver cancer cells and immunocompetent mouse liver cancer models. Notably, metformin improves overall survival of HCC patients with low DOCK1 levels but not among patients with high DOCK1 expression. This study shows that metformin effectiveness depends on DOCK1 levels and that combining metformin with DOCK1 inhibition may provide a promising personalized therapeutic strategy for metformin-resistant HCC patients.Supplementary InformationThe online version contains supplementary material available at 10.1007/s13238-022-00906-6. 相似文献

【目的】利用特异性核酸染料叠氮溴乙锭(Ethidium monoazide bromide, EMA)与实时荧光定量PCR技术相结合, 建立一种能有效区分青枯菌死活细胞的检测方法。【方法】样品DNA制备前经EMA渗透预处理, 再进行实时荧光定量PCR特异扩增菌体DNA。【结果】终浓度为2.0 mg/L的EMA能有效排除1.0×107 CFU/mL灭活青枯菌细胞DNA的扩增, 对活细胞和不可培养状态(Viable but non-culturable, VBNC)活菌的DNA扩增均没有影响。当每个定量PCR反应体系中的活细胞在5.0×100?5.0×104 CFU范围内时, 扩增Ct值与定量PCR反应体系中活细胞CFU对数值呈良好的负相关性(R2=0.992 5)。比较EMA-qPCR法和平板计数法对经过不同温度短期保存的青枯菌检测结果发现, 待检样品可在24 °C与4 °C冷藏条件下短期保存。【结论】本研究建立的EMA-qPCR方法能有效检测青枯菌VBNC细胞和有效区分死活菌, 避免或减少青枯菌PCR检测的假阳性和假阴性。 相似文献

Scrub typhus is the most common endemic vector-borne disease in Taiwan. We identified a total of 4,857 laboratory-confirmed cases during 2006–2016 with hyperendemic foci on offshore islands, including Penghu (778 cases, 16.0%) and Kinmen (716 cases, 14.7%), and eastern Taiwan, including Taitung (628 cases, 12.9%) and Hualien (508 cases, 10.5%). Scrub typhus cases occur year-round throughout Taiwan, with a summer peak in June and July. A total of 545 O. tsutsugamushi isolates were successfully obtained from patients infected in diverse geographic areas, including Taiwan and three offshore islands, and the complete open reading frame of the 56 kDa type-specific antigen gene (tsa56) sequence of these isolates was examined. High phylogenetic diversity was found in these isolates, which could be grouped into 36 distinct sequence types. Most isolates belonged to the Karp (49.9%; 272/545), followed by the TW-22 (17.8%; 97/454) and Kawasaki (14.7%; 80/545) genotypes. In conclusion, our data indicate the widespread presence of tsa56 genotypes closely related to Thailand and Korean strains and the presence of the unique endemic strains TW-12, TW-22, TW-29, and TW-36 in Taiwan. 相似文献

Glioblastoma multiforme (GBM) is the most aggressive and highly vascularized brain tumor with poor prognosis. Endothelial cell-dependent angiogenesis and tumor cell-dependent Vasculogenic mimicry (VM) synergistically contribute to glioma vascularization and progression. However, the mechanism underlying GBM vascularization remains unclear. In this study, GBM stem cells (GSCs) were divided into high and low β8 integrin (ITGB8) subpopulations. Co-culture assays followed by Cell Counting Kit-8 (CCK-8), migration, Matrigel tube formation, and sprouting assays were conducted to assess the proliferative, migratory and angiogenic capacity of GBM cells and human brain microvascular endothelial cells (hBMECs). An intracranial glioma model was constructed to assess the effect of ITGB8 on tumor vascularization in vivo. Our results indicated that ITGB8 expression was elevated in GSCs and positively associated with stem cell markers in glioma tissues, and could be induced by hypoxia and p38 activation. ITGB8 in GSCs inhibited the angiogenesis of hBMECs in vitro, while it promoted the ability of network formation and expression of VM-related proteins. The orthotopic GBM model showed that ITGB8 contributed to decreased angiogenesis, meanwhile enhanced invasiveness and VM formation. Mechanistic studies indicated that ITGB8-TGFβ1 axis modulates VM and epithelial-mesenchymal transition (EMT) process via Smad2/3-RhoA signaling. Together, our findings demonstrated a differential role for ITGB8 in the regulation of angiogenesis and VM formation in GBM, and suggest that pharmacological inhibition of ITGB8 may represent a promising therapeutic strategy for treatment of GBM.Subject terms: Cancer stem cells, CNS cancer 相似文献

Advances in the design of special cryobiomedical apparatus and a review of the trend of developments in the field of cryosurgery in China are discussed. The typical structure of two special cryoprobes for treatment deep in the body and the technology of designing these probes are presented in detail. Some cases which are treated successfully with the above cryoprobes will also be discussed. The experimental aspects of heat transfer in frozen tissue and of the temperature profiles both of a human brain during surgery and of the cryoprobe are described. Other improvements in the field of cryosurgical devices, e.g., four main ways of attaching freezing tips to cryoprobes during surgery and an LN2 transfer tube with high dexterity are also presented. 黄土丘陵区不同侵蚀环境下土壤有机碳对植被恢复的响应 总被引:4,自引:0,他引:4
植被恢复是提高土壤有机碳累积和储存的重要措施。以黄土丘陵区两个典型侵蚀环境下的小流域即黄土区坊塌流域和砒砂岩区满红沟流域退耕坡面为研究对象,分析了土壤有机碳含量(SOCC)、有机碳密度(SOCD)在同一侵蚀环境不同群落下的变化以及在同一群落不同侵蚀环境间的差异,旨在探明不同侵蚀环境下土壤有机碳对植被恢复的响应。结果表明:1)同一侵蚀环境下,与坡耕地相比,自然恢复方式下退耕地植被恢复初期SOCC、SOCD均显著降低,之后随植被恢复均显著升高(P0.05);人工恢复方式下退耕地20—25年柠条锦鸡儿群落和13—14年刺槐群落SOCC、SOCD均显著升高(P0.05),说明同一侵蚀环境内,退耕地在两种恢复方式下均能显著提高土壤有机碳累积和储存。2)同一侵蚀环境下,与相近恢复年限的自然恢复群落相比,刺槐群落SOCC、SOCD均显著高于长芒草+铁杆蒿群落(P0.05),砒砂岩区柠条锦鸡儿群落SOCC、SOCD均显著低于铁杆蒿群落(P0.05),黄土区柠条锦鸡儿群落SOCC显著低于而SOCD显著高于铁杆蒿群落(P0.05),说明相同恢复时间内,相对于自然恢复方式,人工刺槐造林在两种侵蚀环境下均能累积与储存较多的土壤有机碳,而柠条锦鸡儿造林在两种侵蚀环境下累积土壤有机碳的效果均不佳,在黄土区储存土壤有机碳效果好于砒砂岩区。3)同一群落下,黄土区人工和自然恢复群落SOCC均高于砒砂岩区;黄土区人工恢复群落SOCD均显著高于而自然恢复群落SOCD均低于砒砂岩区(P0.05),说明黄土区人工恢复累积和储存土壤有机碳及自然恢复累积土壤有机碳的效果较好,而砒砂岩区自然恢复储存土壤有机碳的效果较好。 相似文献
