Studies were made on the effects of human chorionic somatomammotropin (HCS) on normal and malignant cells and tissues in Swiss mice. HCS was found to produce a significant increase in the fresh weight of normal liver, kidney, spleen, testis, ovary and uterus. Total cell counts of leukocytes and erythrocytes were elevated. The percentage of granulocytes in blood was found to be increased and the percentage of lymphocytes was decreased following HCS treatment. HCS stimulated the growth of ascitic Ehrlich's carcinoma and Sarcoma 180, and nucleic acid synthesis by these tumor cells. A depression in the mitogen induced blastogenesis of lymphocytes was also noted following HCS treatment.  相似文献

The sensitive plant Mimosa pudica is made insensitive by a brief treatment with colchicine. A high concentration of colchicine binding protein is present in the fresh actively moving leaves of M. pudica. This protein was partially characterized and compared with the animal brain tubulin. This colchicine binding activity is very low in the insensitive variety of Mimosa, namely Mimosa rubricaulis.  相似文献

A human mAb (HmAb) termed F105 was obtained by fusion of antibody-producing EBV-transformed cells with the HMMA2.11TG/O cell line. F105 is an IgG1 kappa antibody that binds to the surfaces of cells infected with all HIV-1 strains tested: MN, RF, IIIB, and SF2, but not uninfected cells. The HmAb immunoprecipitates GP120 from all four strains. F105 does not react with denatured GP120 on Western blots, but does react with viral lysates and purified GP120 dotted onto nitrocellulose filter paper under nondenaturing conditions. rGP120 from SF2 and soluble rCD4 inhibit antibody binding to infected cells in a dose-dependent manner. F105 inhibits the binding of free, infectious virions to uninfected HT-H9 cells with 50% of maximal (100%) inhibition at approximately 1 microgram/ml. F105 inhibits infection of HT-H9 cells by 100 tissue culture infective dose 50% units of MN and IIIB strains with 50% inhibition at concentrations of HmAb readily achievable in man. It appears that the F105 HmAb reacts with a conformationally defined epitope on HIV-1/GP120 that is exposed on the free virion and is important for binding to the cell surface by the virion. The epitope, which is immunogenic in humans, appears to be within, or topographically near, the CD4-binding site. F105 and the F105 epitope are potentially useful in therapy and in the design of peptide or anti-Id based vaccines; monitoring of the expression of the Id may prove useful in evaluating immune responses in infected individuals or vaccinated volunteers.  相似文献

Collagen stimulation of blood platelets resulted in significant increases in malondialdehyde (MDA) formation and activity of glucose-6-phosphate dehydrogenase (G6PDH) and a decrease in catalase and glutathione peroxidase (GPx). Retinoic acid (RA) pretreatment did not show any appreciable changes except for a decrease in G6PDH activity as compared with collagen alone. RA pretreatment of human blood platelets resulted in an increase in the activities of catalase and GPx, two important radical scavenging enzymes, with significant decrease in MDA formation when compared with ADP alone. It is suggested that RA has a significant effect on the antioxidant defence system in ADP stimulated platelets but not in the collagen stimulated platelets.  相似文献

Concerns have been raised about the use of relative abundance data derived from next generation sequencing as a proxy for absolute abundances. For example, in the differential abundance setting, compositional effects in relative abundance data may give rise to spurious differences (false positives) when considered from the absolute perspective. In practice however, relative abundances are often transformed by renormalization strategies intended to compensate for these effects and the scope of the practical problem remains unclear. We used simulated data to explore the consistency of differential abundance calling on renormalized relative abundances versus absolute abundances and find that, while overall consistency is high, with a median sensitivity (true positive rates) of 0.91 and specificity (1—false positive rates) of 0.89, consistency can be much lower where there is widespread change in the abundance of features across conditions. We confirm these findings on a large number of real data sets drawn from 16S metabarcoding, expression array, bulk RNA-seq, and single-cell RNA-seq experiments, where data sets with the greatest change between experimental conditions are also those with the highest false positive rates. Finally, we evaluate the predictive utility of summary features of relative abundance data themselves. Estimates of sparsity and the prevalence of feature-level change in relative abundance data give reasonable predictions of discrepancy in differential abundance calling in simulated data and can provide useful bounds for worst-case outcomes in real data.  相似文献

Pulse radiolytically generated halogenated alkylperoxyl radicals, namely CCl3OO*, CBr3OO*, CHCl2OO*, CHBr2OO*, etc. have been employed for a study of the oxidation of beta-carotene in a quaternary microemulsion. All these halogenated alkylperoxyl radicals produce a radical cation (absorption maximum at 840 nm), either via the initial formation of an adduct (absorption maximum at 740 nm), or by direct reaction. There was a considerable blue shift of both absorption peaks in the present system as compared to those earlier reported in non-polar as well as in micellar media. An additional intense absorption peak at approximately 345 nm was noted at a longer time scale and was stable up to 5 ms. On irradiation with a large number of electron pulses, a stable product with an absorption maximum at 315 nm appeared. On excitation at 315 nm, this product gave a fluorescence spectrum with an emission maximum at 525 nm. The absorption and fluorescence spectra of the stable product compared with those of retinol; this was further confirmed by HPLC analysis. A suitable mechanism has been proposed.  相似文献
