The role of CD14 in the phagocytosis and killing of microorganisms was investigated using macrophage-like cell lines, CD14-positive J774.1 cells and CD14-negative mutant J7.DEF3 cells derived from J744.1 cells. The cells were infected with Salmonella typhimurium organisms of the smooth (S)-form LT2, mutant rough (R)-form TV148 or Staphylococcus aureus 248βH. At 30 or 180 min incubation, the cells were washed and disrupted. Colony-forming units (CFUs) liberated from the disrupted cells were determined by quantitative cultivation, and the phagocytic index and killing rate were calculated. Both the phagocytic index and killing rate of J774.1 cells against LT2 organisms were greater than those of J7.DEF.3 cells. However, the index and rate of J774.1 cells against TV148 and 248βH organisms were similar to those of the J7.DEF.3 cells. The phagocytosis of LT2 organisms by J774.1 cells was partially inhibited by S-form LPS (S-LPS) and anti-CD14 antibody, but not by R-chemotype LPS (R-LPS). Bacteroides vulgatus has been shown to be involved in the aggravation of colitis. Previously, we separated two potent virulence factors, capsular polysaccharide (CPS) and lipopolysaccharide (LPS), from a clinical isolate of B. vulgatus and characterized the structure of CPS. In this study, we elucidated the structures of O-antigen polysaccharide (OPS) and lipid A in the LPS. LPS was subjected to weak acid hydrolysis to produce the lipid A fraction and polysaccharide fraction. Lipid A was isolated by preparative TLC, and its structure determined by MS and NMR to be similar to that of Bacteroides fragilis except for the number of fatty acids. The polysaccharide fraction was subjected to gel-filtration chromatography to give an OPS-rich fraction. The structure of OPS was determined by chemical analysis and NMR spectroscopy to be a polysaccharide composed of the following repeating unit: [-->4)alpha-L-Rhap(1-->3)beta-D-Manp(1-->]. 相似文献

Recently, neuroligins (NLs)3 and 4X have received much attention as autism-related genes. Here, we identified syntrophin-gamma2 (SNTG2) as a de novo binding partner of NL3. SNTG2 also bound to NL4X and NL4Y. Berberine is an isoquinoline alkaloid isolated from Coptidis rhizoma, a major herb widely used in Chinese herbal medicine. Berberine's biological activity includes antidiarrheal, antimicrobial, and anti-inflammatory effects. Recent findings show that berberine prevents neuronal damage due to ischemia or oxidative stress and that it might act as a novel cholesterol-lowering compound. The accumulation of amyloid-beta peptide (Abeta) derived from amyloid precursor protein (APP) is a triggering event leading to the pathological cascade of Alzheimer's disease (AD); therefore the inhibition of Abeta production should be a rational therapeutic strategy in the prevention and treatment of AD. Here, we report that berberine reduces Abeta levels by modulating APP processing in human neuroglioma H4 cells stably expressing Swedish-type of APP at the range of berberine concentration without cellular toxicity. Unique pillar-like colonies of aquatic mosses, rising from cyanobacterial and algal mats, have been discovered in some freshwater
lakes in the vicinity of Syowa Station (69°00′S, 39°35′E), continental Antarctica. These moss pillars are about 40 cm in diameter
and up to 60 cm high and occur at the lake bottoms mainly between 3 and 5 m depth. The primary component is a species of Leptobryum, a genus unknown in the continental Antarctic terrestrial bryoflora and as an aquatic genus elsewhere in the world. Bryum pseudotriquetrum is often an associated species. In longitudinal section the pillars reveal several whitish layers formed by mineral sediment
and dead cyanobacteria. It is speculated that the biomass of aquatic mosses at the bottom of many Antarctic lakes is considerably
greater than that previously estimated.

1 We used isozyme variation to examine the genet structure of Uvularia perfoliata patches in gap and closed canopy habitats in a temperate deciduous forest in Maryland, USA.
2 A large patch in a gap habitat was composed of a small number of widely spread genets with many ramets, and a large number of genets with more restricted distribution and few ramets. Genets with many ramets were patchily distributed at a metre scale. Analysis of genet structure on a scale of square centimetres, however, revealed that the genets were highly intermingled with no clear boundaries between them. The presence at both scales of sampling of many genets with unique multilocus genotypes indicated continuing genet recruitment within the population.
3 In the closed canopy habitat, the patches examined were each composed of a single unique multilocus genotype, suggesting that each had developed by asexual propagation following the establishment of a single genet.
4 The clonal structure of U. perfoliata patches in both gap and closed canopy habitats therefore appears to depend on recruitment patterns of genets. Populations in closed canopy habitats are characterized by a 'waiting' strategy, in which asexual ramet production maintains populations until genet recruitment by seed production can occur under the more optimal conditions associated with canopy gaps. Abstract : To examine the physiological roles of the δ subunit of Ca2+ /calmodulin-dependent protein kinase ∥ (CaM kinase ∥δ) in brain, we examined the localization of CaM kinase ∥δ in the rat brain. A specific antibody to CaM kinase ∥δ1-δ4 isoforms was prepared by immunizing rabbits with a synthesized peptide corresponding to the unique carboxyl-terminal end of these isoforms. The prepared antibody did not recognize the α, β, and γ subunits, which were each overexpressed in NG108-15 cells. Immunoblot analysis on various regions and the nuclear fractions from rat brains suggested that some isoforms of CaM kinase ∥δ1-δ4 were abundant in the nucleus in the cerebellum. Total RNA from the cerebellum was analyzed by RT-PCR with a primer pair from variable domain 1 to variable domain 2. We detected the three PCR products δ3.1, δ3.4, and δ3 that contained the nuclear localization signal. These CaM kinase ∥δ3 isoforms were localized in the nuclei in transfected NG108-15 cells. Immunohistochemical study suggested the existence of these isoforms in the nuclei in cerebellar granule cells. These results suggest that CaM kinase ∥δ3 isoforms are involved in nuclear Ca2+ signaling in cerebellar granule cells. 相似文献

Stability constants for the 1:1 complexes of dibenzo-30-crown-10 (DB30C10) with alkali metal ions have been determined at 25 °C in nitromethane and water by conductometry and capillary electrophoresis, respectively. Transfer activity coefficients of DB30C10 and its complexes from nitromethane to S (S = water, acetonitrile, propylene carbonate, methanol, and N,N-dimethylformamide) have been determined at 25 °C to evaluate the solvation properties. The stability constant in the poorly solvating solvent, nitromethane, decreases with increasing metal ion size, Na+ > K+ > Rb+ > Cs+, reflecting the intrinsic selectivity governed by electrostatic interaction between the metal ion and the ether oxygen atoms. It is also suggested that a part of the ether oxygen atoms does not bind to the metal ion in the Na(DB30C10)+ complex. The aqueous stability constant varies as Na+ ? K+ ≈ Rb+ ≈ Cs+; this selectivity pattern is similar to that in acetonitrile, propylene carbonate, and methanol. The complex stability in water is very low compared to that in the nonaqueous solvents, owing to hydrogen bonding of water to the oxygen atoms of the free crown ether. The transfer activity coefficient values show that DB30C10 shields all the metal ions effectively from the solvents and lead to the conclusion that the complexation selectivity in S receives a significant contribution from the solvation of the free metal ions. The Na(DB30C10)+ complex has specific interaction with water, causing much lower K+/Na+ selectivity in H2O than in MeOH. 相似文献
