Isolation of Mesophyll Cells from Sedum telephium Leaves   总被引:4,自引:4,他引:0       下载免费PDF全文
A technique is described for mechanically isolating metabolically active individual spongy mesophyll cells from the Crassulacean acid metabolism plant, Sedum telephium. Mature leaves are selected at about 2 PM when acidity is low, and three different media are used to reduce the problem of leaf acidity and to maintain isotonic conditions. The upper and lower epidermis is peeled from chilled leaves and the leaves are suspended in a buffered “soaking medium,” then gently ground with a mortar and pestle. Cells and debris are separated using a “washing medium,” with cells being filtered through a 136 micron net and collected on an 80 micron net. Cells then are suspended in a “cell suspension medium” and concentrated by centrifugation. Approximately 2 hours are required for the isolation procedure, and activity in CO2 fixation is constant for up to 4 hours after isolation. Microscopic examination shows about 65% of the isolated cells appear intact and unplasmolyzed and are similar to leaf msophyll cells. The yield of cells on a leaf chlorophyll basis is about 1%. Iranian mangrove forests occur between longitude 25°19′ and 27°84′, in the north part of the Persian Gulf and Oman Sea. In 2002, it was estimated that 93.37 km2 of Iranian shorelines were covered with mangrove forests, with the largest area (67.5 km2) occurring between the Khamir Port and the northwest side of Qeshm Island, and the smallest area (0.01 km2) in the Bardestan estuary. Only two species of mangrove are found in the Persian Gulf: Avicennia marina from Avicenniaceae and Rhizophora macrunata from Rhizophoraceae. A. marina is the dominant specie in these forests whereas Rh. macrunata is found only in the Sirik region. Overexploitation of mangrove leaves and oil pollution are the main causes of mangrove destruction in this region.  相似文献

Avicenna recommended honey in the treatment of tuberculosis. Honey has potent bactericidal activity against many pathogenic organisms, but its antimycobacterial effect has never been studied. In this study the antimycobacterial effect of honey was evaluated in vitro. Standard Lowenstein-Jenson media and L-J media containing various concentrations of honey were prepared. Two bacilli from positive cultures and two from positive smears of the affected patients were inoculated on each of the prepared plates. It was demonstrated that the growth of mycobacteria was inhibited by adding 10% honey to the media. Mycobacteria did not grow in culture media containing 10% and 20% honey while it grew in culture media containing 5%, 2.5% and 1% honey. The good antimycobacterial effect, sterility, low cost, and easy availability of honey makes it an ideal antimycobacterial agent.  相似文献

Morphine can influence immediate early genes (IEG) of activity-regulated cytoskeletal-associated protein (Arc) and brain-derived neurotrophic factor (BDNF) which are activated in response to physiological stimuli during learning, as well as the nerve growth factor (NGF) gene which increases the expression of several IEGs for memory formation. The purpose of the current study was first to evaluate the effect of acute (1-day) and subchronic (15-days) morphine administration on memory retrieval of rats and second to determine the hippocampal expression of NGF, BDNF and Arc genes as potential contributors in the observed effects in each setting. The effects of morphine (intraperitoneal, 10, 15 and 20 mg/kg) on memory function and gene expression were assessed using inhibitory avoidance test and real-time polymerase chain reaction, respectively. We found that a single dose of morphine at the highest dose of 20 mg/kg decreases the post-training step-through-latency, while repeated administration of the same dose for 15 successive days increases this indicator of memory retrieval. We did not detect a significant change in the hippocampal expression of Arc, BDNF or NGF genes after a single episode of morphine treatment. However, subchronic morphine administration (15 and 20 mg/kg) increased the expression of Arc and BDNF genes in a dose dependent manner. A higher mRNA expression for the NGF was observed at the higher dose of 20 mg/kg. We hypothesize that the subchronic effects were morphine-induced behavioral sensitization which may have been enhanced through increased hippocampal Arc expression.

Crystal structures are reported for the D85S and D85S/F219L mutants of the light-driven proton/hydroxyl-pump bacteriorhodopsin. These mutants crystallize in the orthorhombic C222(1) spacegroup, and provide the first demonstration that monoolein-based cubic lipid phase crystallization can support the growth of well-diffracting crystals in non-hexagonal spacegroups. Both structures exhibit similar and substantial differences relative to wild-type bacteriorhodopsin, suggesting that they represent inherent features resulting from neutralization of the Schiff base counterion Asp85. We argue that these structures provide a model for the last photocycle intermediate (O) of bacteriorhodopsin, in which Asp85 is protonated, the proton release group is deprotonated, and the retinal has reisomerized to all-trans. Unlike for the M and N photointermediates, where structural changes occur mainly on the cytoplasmic side, here the large-scale changes are confined to the extracellular side. As in the M intermediate, the side-chain of Arg82 is in a downward configuration, and in addition, a pi-cloud hydrogen bond forms between Trp189 NE1 and Trp138. On the cytoplasmic side, there is increased hydration near the surface, suggesting how Asp96 might communicate with the bulk during the rise of the O intermediate.  相似文献

We derive a formula giving the frequency with which random drift shifts a population between alternative equilibria. This formula is valid when such shifts are rare (Ns much greater than 1), and applies over a wide range of mutation rates. When the number of mutations entering the population is low (4 N mu much less than 1), the rate of stochastic shifts reduces to the product of the mutation rate and the probability of fixation of a single mutation. However, when many mutations enter the population in each generation (4 N mu much greater than 1), the rate is higher than would be expected if mutations were established independently, and converges to that given by a gaussian approximation. We apply recent results on bistable systems to extend this formula to the general multidimensional case. This gives an explicit expression for the frequency of stochastic shifts, which depends only on the equilibrium probability distribution near the saddle point separating the alternative stable states. The plausibility of theories of speciation through random drift are discussed in the light of these results.  相似文献

We investigate the probability of fixation of a chromosome rearrangement in a subdivided population, concentrating on the limit where migration is so large relative to selection (m ? s) that the population can be thought of as being continuously distributed. We study two demes, and one- and two-dimensional populations. For two demes, the probability of fixation in the limit of high migration approximates that of a population with twice the size of a single deme: migration therefore greatly reduces the fixation probability. However, this behavior does not extend to a large array of demes. Then, the fixation probability depends primarily on neighborhood size (Nb), and may be appreciable even with strong selection and free gene flow (≈exp(-B ≈ Nbs) in one dimension, ≈exp(-B ≈ Nb) in two dimensions). Our results are close to those for the more tractable case of a polygenic character under disruptive selection.  相似文献
