Diacylglycerol kinase (DGK) catalyzes conversion of a lipid second messenger diacylglycerol to another messenger molecule phosphatidic acid. Consequently, DGK plays a pivotal role in cellular pathophysiology by regulating the levels of these two messengers. We reported previously that DGKζ translocates from the nucleus to cytoplasm in hippocampal neurons under ischemic/hypoxic stress. In addition, we also identified nucleosome assembly protein 1 (NAP1)-like proteins NAP1L1 and NAP1L4 as novel DGKζ-interacting partners using a proteomic approach and revealed that these NAP1-like proteins induce cytoplasmic translocation of DGKζ in overexpressed cells because NAP1-like proteins associate with the nuclear localization signal of DGKζ and block its nuclear import via importin α. In the present study, we examined whether NAP1-like proteins are expressed in the brain and whether the molecular interaction of DGKζ and NAP1-like proteins would be changed in the brain after hypoxic stress. Immunohistochemistry revealed that NAP1L1 and NAP1L4 are widely expressed in neurons and glial cells in the brain with some differences. After 3 days of transient whole-body hypoxic stress, DGKζ translocated from the nucleus to cytoplasm in hippocampal pyramidal neurons, whereas NAP1-like proteins remained in the cytoplasm. Contrary to our expectations, NAP1-like proteins showed no change in their expression levels. The molecular interaction between DGKζ and NAP1-like proteins was attenuated after hypoxic stress. These results suggest that DGKζ cytoplasmic translocation in neurons under hypoxic stress is regulated by some mechanism which differs from that mediated by NAP1-like proteins. 相似文献

We calculated the probability of paternity exclusion (P) in 6 troops of rhesus and Japanese macaques housed in open enclosures and 68 wild troops of Japanese, crab-eating, and toque
macaques using 33 genetic loci which encoded the blood protein variations detected by electrophoretic techniques. In the open
enclosures, especially of rhesus troops, we obtained a fairly high probability of paternity exclusion and succeeded in determining
the fathers of offspring. However, we found significant differences between the observed and calculated probabilities in most
of the troops. These differences were ascribed to a situation whereby the Hardy-Weinberg equilibrium had not been attained
in the troops and/or the numbers of variable loci were too small. In the wild troops of Japanese, crab-eating, and toque macaques,
the means ofP were 0.2274 (0.0192–0.5017), 0.4635 (0.1676–0.7151), and 0.7382 (0.6266–0.7954), respectively. We also estimated the number
of loci needed to determine the fathers of offspring with a probability of 0.8 assuming that ten males were present in the
troop. The estimated number was about 13.5 times, 5 times and 1.8 times the number of loci examined on average in the troops
of Japanese, crab-eating and toque macaques, respectively. This means that determination of most of the fathers of offspring
in wild troops of these macaques, even of toque macaques which have a rather high probability of paternity exclusion, is difficult

The entomogenous fungus Nomuraea rileyi reportedly secretes a proteinaceous substance inhibiting larval molt and metamorphosis in the silkworm Bombyx mori. We studied the possibility that N. rileyi controls B. mori development by inactivating hemolymph molting hormone, ecdysteroids. Incubation of ecdysone (E) and 20-hydroxyecdysone (20E) in fungal-conditioned medium resulted in their rapid modification into products with longer retention times in reverse-phase HPLC. Each modified product from E and 20E was purified by HPLC, and identified by NMR as 22-dehydroecdysone and 22-dehydro-20-hydroxyecdysone. Some other ecdysteroids with a hydroxyl group at position C22 were also modified. Injection of the fungal-conditioned medium into Bombyx mori larvae in the mid-4th instar inhibited larval molt but induced precocious pupal metamorphosis, and its injection into 5th instar larvae just after gut purge blocked pupal metamorphosis. In hemolymph of injected larvae, E and 20E disappeared and, in turn, 22-dehydroecdysone and 22-dehydro-20-hydroxyecdysone accumulated. These results indicate that N. rileyi secretes a specific enzyme that oxidizes the hydroxyl group at position C22 of hemolymph ecdysteroids and prevents molting in B. mori larvae. 相似文献

The degradation of proteins of the rod outer segment (ROS) fraction by partially purified cathepsin D [EC 3.4.23.5] from the retinal pigment epithelium was studied. The ROS fraction, prepared from bovine eyes by sucrose density gradient centrifugation, had little cathepsin D activity. Partially purified cathepsin D, obtained from crude extract of bovine retinal pigment epithelium using bovine serum albumin as a substrate, hydrolyzed the porteine of the ROS fraction. The rate of degradation of ROS proteins was proportional to both the enzyme concentration and the incubation time. With ROS proteins as substrate, the optimal pH of cathepsin D was about 3.5. The degradation of ROS proteins was inhibited by pepstatin. 相似文献

development of chimpanzees and compared their skeletal development with humans'. A development pattern of chimpanzees repeated
accelerations and decelerations displaying “early-juvenile trough,” “pre-adolescent peak,” “mid-adolescent trough,” and “post-adolescent
peak” in incremental curves. Sex differences in skeletal development are slower development in males during infant and early
juvenile phases, and greater increment around the adolescent phase in males. Females are fully mature at younger ages than
males, e.g. about one and a half years. In comparison with chimpanzees, humans have such characteristics as a longer slower

The efficient production of GlcNAc by cellulases derived from Trichoderma viride (T) and Acremonium cellulolyticus (A) was observed by HPLC analysis compared to lipase, hemicellulase, and pectinase. β-Chitin showed higher degradability than α-chitin when using cellulase T. The optimum pH of cellulase T was 4.0 on the hydrolysis of β-chitin. The yield of GlcNAc was enhanced by mixing of cellulase T and A. 相似文献

Prolonged administration of dipropylacetic acid, a branched-chain fatty acid, reduced the glycine cleavage activity in the liver of rat to about one-third of the activity in the control rat. The reduction of the activity appeared to be due mainly to reduction of the level of P-protein, a pyridoxal phosphate enzyme, which is responsible for the first step of the glycine cleavage, although dipropylacetic acid was also found to inhibit the activity of P-protein in vitro in a noncompetitive but partially competitive manner with respect to glycine. The rat treated with dipropylacetic acid may provide an experimental approach for the biochemical study of hyperglycinemia which accompanies to metabolic disorders of branchedchain keto acids. In the dipropylacetic acid-treated rat, however, the glycine concentration in the serum was not appreciably elevated and this may be accounted for by the fact that the activities of both the glycine cleavage system and serine dehydratase are considerably higher in the rat liver as compared with those in other animals including human. 相似文献

Information on mechanisms of differentiation or homogenization of urban floras is deficient, despite their importance for urban nature conservation and urban land management. Roads, as a major human promoter of urbanization, can be an initial habitat for plants dispersed by transportations. We assumed that variation in weed vegetation along urban–rural roadside gradients is small, particularly in curbside cracks. We classified vegetation occurring in curbside cracks along the National Route 3 in southern Japan and compared the characteristics of the vegetation types recognized. Species in curbside cracks were recorded on 40 plots. Three vegetation types were classified, in part related to surrounding land-use types. Although the Shannon–Wiener diversity index and the number of native species clearly differed among the vegetation types in the curbside cracks, no significant differences between the three floristic vegetation types were found in the number of non-natives and invasive alien species. This may result from the small specificity and complexity of landscape structures, due to the continuity and connectivity of paved-road networks. Of the 122 species, 44 were non-natives or invasive alien species. The vegetation types generally involved the same plant families, with large numbers of species from the Asteraceae and Poaceae, regardless of vegetation types, but frequency of occurrence of the two families clearly differed between natives and non-natives and invasive alien species. Ephemeral non-natives and invasive alien species, particularly Asteraceae and Poaceae, seem to have advantageous dispersal strategies or low habitat dependency facilitating their occurrence in curbside cracks, regardless of adjacent land uses and the urban–rural landscape gradient. The doublecortin (DCX) gene was recently found to be involved in patients with X-linked lissencephaly and subcortical band
heterotopia or double cortex syndrome. We have studied the coding regions of the DCX gene in 11 Japanese patients with cortical
dysplasia and have identified three different mutations (R186C in exon 3, R272X and R303X in exon 5) in four sporadic female
cases. R272X, which has been detected in two unrelated cases, is a novel mutation. Although the number of cases studied remains
limited, exon 5 may be a common mutational site in Japanese patients in contrast to many previus reports concerning exons
2 and 3.
