A novel HPLC-UV method with pre-column derivatization by using 2-mercaptoethanol was established for determination of 1,2-[bis(1,2-benzisoselenazolone-3(2H)-ketone)]-ethane (BBSKE) in dog plasma. The derivatives were identified by mass spectrometry. The method had a good linear range of 0.05-2 microg/ml (r(2)=0.9995). The lower limit of quantification (LOQ) was 0.05 microg/ml. The precision and accuracy were less than 7%. After dosing of BBSKE (30 mg/kg, p.o. and 0.79 mg/kg, i.v.) in dogs, AUC(0-t) were 5.72+/-2.42 and 1.35+/-0.41 microg h/ml; t(1/2) were 4.6+/-2.1 and 1.7+/-0.6h, respectively. Salt stress is one important factor influencing the growth and development of plants, and salt tolerance of plants is a result of combined action of multiple genes and mechanisms. Rosa rugosa is not only an important ornamental plant, but also the natural aromatic plant of high value. Wild R. rugosa which is naturally distributed on the coast and islands of China has a good salt tolerance due to the special living environment. Here, the vacuolar Na+/H+ reverse transporter gene (NHX1) and the vacuolar H+-ATPase subunit C gene (VHA-c) closely related to plant salt tolerance were isolated from wild R. rugosa, and the expression patterns in R. rugosa leaves of the two genes under NaCl stress were determined by real-time quantitative fluorescence PCR. The results showed that the RrNHX1 protein is a constitutive Na+/H+ reverse transporter, the expression of the RrNHX1 gene first increased and then decreased with the increasing salt concentration, and had a time-controlled effect. The RrVHA-c gene is suggestive of the housekeeping feature, its expression pattern showed a similar variation trend with the RrNHX1 gene under the stress of different concentrations of NaCl, and its temporal expression level under 200 mM NaCl stress presented bimodal change. 以南粳44为供试材料,在粘土和砂土土壤中,设置麦秸秆不还田和全量还田(6000 kg·hm-2)及3种施氮量(0、225、300 kg·hm-2)试验,研究了麦秸秆全量还田的腐解率和有机碳释放量动态变化,及其对稻田0~45 cm土壤溶解有机碳(DOC)含量和水稻产量的影响.结果表明: 麦秸秆还田的前期(0~30 d)其腐解率和有机碳释放量最高,腐解率为35.0%(粘土)和31.7%(砂土),有机碳释放率为34.1%(粘土)和33.1%(砂土);30 d后两者均减小.施用氮肥可显著促进秸秆腐解和有机碳释放量,粘土中麦秸秆腐解率和有机碳释放量明显大于砂土.麦秸秆还田后土壤DOC含量逐渐增加,至25 d达最大值,粘土和砂土分别为60.18和56.62 mg·L-1,此后逐渐减小并趋于稳定.麦秸秆还田处理15 cm处土壤DOC含量显著高于未还田处理,但两者在30和45 cm处土壤DOC含量差异不显著,说明秸秆还田主要增加了稻田0~15 cm土层DOC含量.与不施氮处理相比,施氮处理土壤DOC含量降低,2种施氮处理间差异不显著.秸秆还田减少了水稻前期分蘖发生量,显著降低了有效穗数,增加了穗粒数、结实率和千粒重,显著提高了水稻产量. 相似文献

A Chlamydomonas reinhardtii chloroplast expression vector, pACTBVP1, containing the fusion of the foot and mouth disease virus (FMDV) VP1 gene and the cholera toxin B subunit (CTB) gene was constructed and transfered to the chloroplast genome of C. reinhardtii by the biolistic method. The transformants were identified by PCR, Southern blot, Western blot and ELISA assays after selection on resistant medium and incubation in the dark. The CTBVP1 fusion protein was expressed in C. reinhardtii chloroplast and accounted for up to 3% of the total soluble protein. The fusion protein also retained both GM1-ganglioside binding affinity and antigenicity of the FMDV VP1 and CTB proteins. These experimental results support the possibility of using transgenic chloroplasts of green alga as a mucosal vaccine source. 相似文献
