Novel triphosphate derivatives bearing bulky or small groups at alpha-position attached to the triphosphate residue through linkers of different structures and lengths were synthesized and studied as substrates toward terminal deoxynucleotidyltransferase. The substrate efficacy depends on the structure of substituents, linker length, and nature of metal activator. The replacement of hydrophobic groups by small substituents decreased the substrate efficacy by about 20 times in respect to hydrophobic residues. The dependence on metal activator is the following: Co(2+) > Mn(2+) > Mg(2+). The model of interaction of alkyl triphosphates with linkers of different lengths bearing TdT active site is presented.  相似文献

Time course of changes in intracellular water, K+ and Na+ of U937 cells incubated in hyperosmolar medium with addition of 200 mM sucrose was studied. Ouabain-sensitive and ouabain-resistant Rb+ (K+) influxes were measured during regulatory cell volume increase (RVI) and apoptotic volume decrease (AVD). Microscopy of cells stained by Acrydine orange, Ethydium bromide, APOPercenrage Dye and polycaspase marker FLICA was performed. We found that initial osmotic cell shrinkage induced both RVI and AVD responses. RVI dominated at the early stage whereas AVD prevailed at the later stage. In view of the data obtained in U937 cells the current opinion that RVI "dysfunction" is a prerequisite for apoptosis and AVD (Subramanyam et al., 2010) should be revised. U937 cells are capable to trigger of apoptosis and AVD in spite of the unimpaired RVI response. It is concluded that AVD plays a significant role in preventing osmotic lysis of apoptotic cells rather than in the initiation of apoptosis.  相似文献

Theoretical and experimental analysis of interaction of modified D- and L- dNTP as substrates for template-dependent and template-independent DNA polymerases was performed. It is shown that if the modified nucleoside 5'-triphosphates do not contain a substituent in position 3' DNA chains can be extended by both strereoisomeric series with the same kinetic parameters. But the presence of even a 3'-hydroxy group in L-dNTP prevents their incorporation into the DNA chain.  相似文献

Plants of buckwheat Fagopyrum esculentum possessing diploid and tetraploid genotypes were studied at the initial ontogenetic stages. They were compared in their morphophysiological characteristics, accumulation of phenolic compounds (including their main classes—phenylpropanoids and flavonoids), and activity of L-phenylalanine ammonia-lyase. An apparent resemblance in morphophysiological characteristics of seedlings was found between the two specimens, but diploid plants tended to faster linear growth than tetraploid ones. Differences in the accumulation of phenolic compounds in the hypocotyl and cotyledonous leaves were revealed. In most cases, in the course of seedling growth, the changes in phenylalanine ammonialyase activity did not correlate with the changes in the levels of these secondary metabolites. The effects of gene dosage were established towards accumulation of phenylpropanoids and anthocyanins in hypocotyls of seedlings and flavonoid accumulation in cotyledonous leaves. It is concluded that buckwheat seedlings with a tetraploid genotype have higher capacity than diploid seedlings for biosynthesis of phenolics.  相似文献

A study was conducted to determine in situ rates of Fe(II) oxidation and Fe(III) precipitation along a 5.0 m reach of a ferruginous groundwater discharge zone under two distinct conditions; (i) the natural state featuring abundant flocculent mats of bacteriogenic iron oxides (BIOS) produced by Fe(II)-oxidizing bacteria, and (ii) after a manual washout of the streambed to remove the microbial mat. Examination of mat samples by differential interference contrast light microscopy revealed tangled meshworks of filamentous Leptothrix sheaths and helical Gallionella stalks intermixed with fine-grained hydrous ferric oxide (HFO) precipitates. The greatest accumulation of BIOS mat was 1.0 m downstream of the groundwater spring. Redox potential (Eh) increased sharply from 200 mV to over 300 mV over the last 2.0 m of the reach. Similarly, dissolved oxygen increased from < 10% saturation to almost 100% saturation over the last 2.0 m of the reach, whereas pH increased from 6.4 to 7.3. Pseudo-first-order rate constants determined on the basis of analytical solutions to sequential partial differential advection-dispersion-reaction equations for the linear Fe(II)→Fe(III)→HFO reaction network yielded in situ Fe(II) oxidation rate constants (kox) of 1.70 ± 0.20 min?1 in natural conditions and 0.48 ± 0.14 min?1 after washout. Corresponding Fe(III)-precipitation rates (kp) before and after washout were 3.45 ± 0.10 min?1 and 0.90 ± 0.01 min?1, respectively. These values for kox and kp are higher than estimates obtained from closed batch microcosm and laboratory experiments, underscoring the crucial dependence of in situ Fe(II) oxidation and Fe(III) precipitation rates on advective and dispersive mass transport. The results also highlight the influence that BIOS microbial mats exert on the reaction kinetics of the multiple heterogeneous reactions contributing not only to Fe(II)/Fe(III) redox transformations in groundwater discharge zones, but also the precipitation of HFO.  相似文献

Rose vervain is a little-known ornamental annual plant. The adaptive properties (drought and cold resistance) and long period of flowering make this species promising for growing in flower gardens and containers. Chemical mutagenesis widely used for various plant species was applied to induce character variation in Rose vervain. The properties of development of flowers and inflorescences in lines descending from the å3–å5 mutants generated by the seed exposure to chemical mutagens diethyl sulfate and nitrosomethylurea were considered.  相似文献
