Summary The selective formation of three tetrapyrroles, Co-containing corrinoids, Ni-containing factor F430 and Fe-containing cytochromes (haems) by Methanosarcina barkeri Fusaro (DSM 804) was achieved as a function of the concentrations of Co2+, Ni2+ and Fe2+ in a methanol minimmum medium. It was found that about 70% of the total tetrapyrroles synthesized was excreted into the culture supernatant. Hence, the continuous production of tetrapyrroles in a fixed-bed reactor (supporter: porous diatomaceous clay) was carried out at a dilution rate of 10 day-1 (850 ml medium/85 ml column/day). The effluent discharged from the reactor contained the excreted tetrapyrroles, the concentrations of which were dependent upon the Co2+, Ni2+ and Fe2+ concentrations in the feed medium. The maximum productivities from the reactor (1 l basis) were 52 M corrinoids/day, 24 M F430/day and 8 M haems/day, respectively.  相似文献

Hormone autonomous callus was institutioniated reproduciblyon MS agar medium with 0.25 mg/liter of BA as the sole planthormone (AI medium) from young leaf explants of sugar beets.When leaf explants were inoculated into AI medium and culturedon a reciprocal shaker, single cells began to be released fromthe cut surfaces of the leaf pieces after 6 days, followed byactive release. When the single cells which had been releasedwere transferred to fresh liquid MS medium without any planthormones, they could divide and grow autonomously, giving riseto hormone-autonomous suspension cultures. Specimens ofCanthigaster rivulata (Temminck et Schlegel) were collected from Kominato and Hayama, central Japan, from May, 1985 to October, 1986. On the basis of the gonadosomatic index, gonadal histology and results of artificial fertilization of these specimens, the spawning season is considered to extend from late June to mid-September. The specimens exhibited the following dimorphic differences associated with sex: 1) The male is larger than the female. 2) Ventral side of the body is brownish orange in the male with vermiculated or reticulated patterns of bright violet, while it is white in the female. 3) The male has a well-developed skin fold along the mid-dorsal and mid-ventral lines, which is greatly elevated during courtship; whereas the female’s skin folds are not or slightly developed and conspicuous only during courtship. In an aquarium with the water temperatures of 22 to 26°C, a pair of fish spawned every four days late in the morning for three consecutive months. Courtship and spawning occurred in a pair. The male swam in front of the female, and elevated the skin folds both dorsally and ventrally, fully spreading the unpaired fins, with the ventral side of the body flashing bright blue and the dorsal side turning dark. Both fish swam in a circular fashion, elevating the skin folds. The male followed the female nudging her abdomen with his snout. Both fish turned upward, and released gametes. The eggs are spherical, 0.53–0.73 mm in diameter, demersal, adhesive, transparent, and pale yellowish orange in color, and contain a cross-shaped or asteroid cluster of oil globules. The egg membrane was thick and consisted of about 14 concentric layers. The incubation period ranged from 73.5 hours at 28.2–28.5°C to 145.0 hours at 22.1–22.4°C. The newly hatched larvae were 1.38–1.98 mm in total length (TL) with 84-11-13 = 19–21 myomeres. The yolk was absorbed when the larvae attained 1.49–2.22 mm TL, three days after hatching. The larvae were fed on oyster larvae, blue mussel larvae, sea-urchin larvae and rotifers, but all of them died in 16 days. During the embryonic and early larval stages, the only pigment cells that appeared on the body were the black chromatophores.  相似文献

Summary Amino acid residues of the carboxyl-terminal region of kanamycin nucleotidyltransferase were modified using segment-directed mutagenesis. Six different mutant enzymes with single amino acid replacements were selected out of 59 clones by DNA sequence analyses. The mutant enzymes were purified and it was found that the thermostability of one mutant enzyme was identical to the wild type, whereas the other five were less thermostable at varying degrees. The data suggested that changes in the enzyme thermostability depend not only on the position but also on the species of amino acid residue replaced.  相似文献

The endogenous calcium ion (Ca2+) in horseradish peroxidase (HRP) was removed to cause substantial changes in the proton NMR spectra of the enzyme in various oxidation/spin states. The spectral changes were interpreted as arising from the substantial alterations in the heme environments, most likely the heme proximal and distal sides. The comparative kinetic and redox studies revealed that these conformational changes affect the reduction process of compound II, resulting in the decrease of the enzymatic activity of HRP. It is also revealed from the ESR spectrum and the temperature dependences of the NMR and optical absorption spectra of the Ca2+-free enzyme that the heme iron atom of the Ca2+-free enzyme is in a thermal spin mixing between ferric high and low spin states, in contrast to that of the native enzyme. These results show that Ca2+ functions in maintaining the protein structure in the heme environments as well as the spin state of the heme iron, in favor of the enzymatic activity of HRP.  相似文献

Summary Mouse lymphoma L1210 cells maintained in vitro at a high cell density for a certain time period adapted themselves to the in vitro environment and were able to grow indefinitely. From these adapted cells, more than 30 clones were isolated. They all had much higher activity to take up cystine than the original L1210 cells, supporting a previous view that the deficiency of the cystine uptake limits the survival and growth of L1210 cells in vitro. The cystine uptake of one cloned cell line was characterized. The enhanced uptake of cystine in these cells was mainly mediated by a Na+-independent, saturable system and was potently inhibited by glutamate and some other anionic amino acids, but less by aspartate. Such activity of cystine uptake was not observed in the original L1210 cells. The results suggest that, upon adaptation in vitro, L1210 cells acquire a new cystine transport activity necessary for survival and growth in vitro.  相似文献

Cynoglossus abbreviatus spawns from mid-March to mid-April in the Sea of Shimabara in Kyushu. During the spawning season ovarian maturation was successfully induced by injection of the pituitary homogenate ofHypophthalmichthys molitrix. The dose of the aceton-dried pituitary homogenate was 6.5 mg/kg body weight ofC. abbreviatus. It took about 2 days for ovulation after injection at a water temperature of 14 to 16°C. Artificial fertilizations were accomplished on March 29, 1974 and again on April 7, 1984, using the females matured by hormone injection in the latter case only. The larvae were reared on the rotifers,Artemia nauplii,Tigriopus japonicus and copepods collected from the sea over a period of 113 days in 1974 and 58 days in 1984. The eggs were pelagic, spherical, 1.19–1.23 mm in diameter and had 30–50 oilglobules of 0.068–0.095 mm in diameter, and the perivitelline space was narrow. The incubation period was 90–98 hours at a water temperature of 14 to 16°C. The newly hatched larvae were 3.18–3.45 mm TL and had 61–64 myomeres. The larvae had many melanophores and xanthophores on the body, forming three bands on the caudal region, but were lacking chromatophores on the finfolds. The yolk was completely absorbed when the larvae attained a size of 4.7–5.6 mm TL 8 days after hatching. A single elongated dosai fin ray developed on the head in the 8-day old larvae. The ray was reduced in size as long as the other rays 1 or 2 days after metamorphosis. The rudiment of pectoral fins were found on the both sides of the body in the 2-day old larvae, but two of them disappeared after metamorphosis. A pelvic fin first appeared as a ventral bud just anterior to the gut in the larva of 8.39 mm TL. The full count of 4 rays was observed on the larva of 10.83 mm TL. Metamorphosis began 22 days after hatching when the larvae were 11.20 mm TL. The right eye began to shift the left side of the head at night and reached to the final place after 8.5 hours. It took about 36 hours to complete the metamorphosis, including the eye movement and fusion of the hole in the rostral beak. At the last stage of metamorphosis, the dosal, caudal, anal and ventral fins became confluent. The larvae reached the juvenile stage at a size of 13.5–14.0 mm TL, approximately 28 days after hatchling. The growth of larvae reared in 1974 is expressed by the following equations: Y1 = 3.448 · 1.0507x (8≦X≦28) Y2 = 6.3322 · 1.0275x (28≦X≦75) where Y is the total length (mm) and X is the number of days after hatching. Development of fin-supports and fin-rays was observed in larval and juvenileChanos chanos, Chondrification of the caudal complex started at 4.70 mm SL. Ossification of the caudal elements started at 7.80 mm SL and was nearly completed at about 30 mm SL. Cartilaginous fusion of caudal elements, which occurs in hypurals of higher teleostean fishes but is not seen in lower teleosts, was observed between the neural arch of the preural centrum 1 and that of the ural centrum 1 via a small cartilage bridging the distal tips of the two arches. Caudal finrays began to develop at 6.60 mm SL, and an adult complement of principal rays was attained at 7.35 mm SL. Dorsal and anal pterygiophore elements were first evident at 6.70 mm and 6.65 mm SL, respectively. All proximal radiais were formed at 8.15 mm SL in both fins. Formation of dorsal and anal fin-rays started simultaneously at 8.60 mm SL, and adult fin-ray complements were attained at 10,00 mm and 10.70 mm SL, respectively. In the pectoral fin, the cleithrum, coraco-scapular cartilage and blade-like cartilage (fin plate) had already been formed at 4.65 mm SL. The mesocoracoid was observed to originate from the coraco-scapular cartilage and become detached from it in the course of ossification. Pectoral fin-ray formation started at 13.80 mm SL and was completed in number of rays at 20.00 mm SL. In the pelvic fin, the basipterygium was first evident at 13.00 mm SL. Pelvic fin-rays appeared at 13.80 mm SL and attained their adult count at 17.15 mm SL.  相似文献

In 1987, a study on buried seed populations was conducted in the crater basin of Mt. Usu, a volcano located in northern Japan, where the vegetation had beeen almost completely destroyed by eruptions in 1977 and 1978. The former vegetation had consisted of grassland and broad-leaved forest. In the areas formerly occupied by this grassland and forest, 2128.0 and 1985.3 seeds per square meter, respectively, were extracted from 12 blocks of the former topsoil using a floattion method. This revealed that many seeds were still viable even after ten years of burial under thick volcanic deposits. The seeds were distributed more in sandy soil than in rocky soil of the former topsoil. Twenty-five species, most of which favored grassland, were detected in both the former grassland and forest. From comparison of α-and β-diversities between the grassland and forest, the structure of the buried seed population was considered likely to have been more diverse in the forest than in the grassland. The determinants of composition of the buried seed populations were discussed with reference to the former vegetation and soil characteristics.  相似文献

Soon after the 1977–78 eruptions of the volcano Usu, there were created many gullies in which former topsoil, i.e., well-developed soil accumulated before the eruptions, was eroded in the crater basin, whereas the outside of the gully was covered with thick volcanic deposits. The short-dispersal-seed plants were the most abundant in the inside of gully where the former topsoil was exposed, however, they have not immigrated from external environments. The germination tests of the seeds extracted from the former topsoil demonstrated that viable seeds were buried at 1683.3/m2 for 9 years after the eruptions. At least, 12 herbaceous species, e.g.,Rumex obtusifolius, Geum macrophyllum var.sachalinense andPoa annua, were derived from the buried seeds in the former topsoil. Their contribution to revegetation was estimated to be 40.0% in the inside of gully where the former topsoil was exposed in 1983, suggesting that buried seeds are one of the most important seed source for revegetation even in the case of a volcano which produced pumice and ash by eruption.  相似文献
