With the aim to address an undesired cardiac issue observed with our related compound in the recently disclosed novel series of renin inhibitors, further chemical modifications of this series were performed. Extensive structure–activity relationships studies as well as in vivo cardiac studies using the electrophysiology rat model led to the discovery of clinical candidate trans-adamantan-1-ol analogue 56 (DS-8108b) as a potent renin inhibitor with reduced potential cardiac risk. Oral administration of single doses of 3 and 10 mg/kg of 56 in cynomolgus monkeys pre-treated with furosemide led to significant reduction of mean arterial blood pressure for more than 12 h. 相似文献

When mt+ and mt gametes of Chlamydomonas reinhardtiiwere mixed, shedding of cell walls took place in both matingtypes during massive agglutination and/or pairing. This wascaused by a cell wall lytic factor that had been induced byflagellar agglutination and excreted into the medium by cellsconcurrently with their cell wall release. When glutaraldehyde-fixed gametes and isolated flagella of onemating type caused isoagglutination of live gametes of the othermating type, the live mt+ gametes induced the lytic factor andshed their walls, whereas none of the live mt did this.The cell walls of mt gametes were lost only when thelytic factor, which had been excreted by mt+ gametes into themedium, acted from the outside. Bacteria living in soil collected from a rice paddy in Fukuoka, Japan, were examined by electron microscopy using a freeze-substitution fixation method. Most of the observed bacteria could be categorized, based on the structure of the cell envelope and overall morphology, into one of five groups: (i) bacterial spore; (ii) Gram-positive type; (iii) Gram-negative type; (iv) Mycobacterium like; and (v) Archaea like. However, a few of the bacteria could not be readily categorized into one of these groups because they had unique cell wall structures, basically resembling those of Gram-negative bacteria, but with the layer corresponding to the peptidoglycan layer in Gram-negative bacteria being extremely thick, like that of the cortex of a bacterial spore. The characteristic morphological features found in many of these uncultured, soil-dwelling cells were the nucleoid being in a condensed state and the cytoplasm being shrunken. We were able to produce similar morphologies in vitro using a Salmonella sp. by culturing under low-temperature, low-nutrient conditions, similar to those found in some natural environments. The first step in developing regenerative medicine approaches to treat renal diseases using pluripotent stem cells must be the generation of intermediate mesoderm (IM), an embryonic germ layer that gives rise to kidneys. In order to achieve this goal, establishing an efficient, stable and low-cost method for differentiating IM cells using small molecules is required. In this study, we identified two retinoids, AM580 and TTNPB, as potent IM inducers by high-throughput chemical screening, and established rapid (five days) and efficient (80% induction rate) IM differentiation from human iPSCs using only two small molecules: a Wnt pathway activator, CHIR99021, combined with either AM580 or TTNPB. The resulting human IM cells showed the ability to differentiate into multiple cell types that constitute adult kidneys, and to form renal tubule-like structures. These small molecule differentiation methods can bypass the mesendoderm step, directly inducing IM cells by activating Wnt, retinoic acid (RA), and bone morphogenetic protein (BMP) pathways. Such methods are powerful tools for studying kidney development and may potentially provide cell sources to generate renal lineage cells for regenerative therapy. 相似文献

Copulatory behavior unrelated to conception is sometimes observed in some non-human primates including the Japanese monkey.
In the present study, the authors examined whether a mature follicle or a newly formed fresh corpus luteum was observed in
the ovaries of female Japanese monkeys which displayed the copulatory behavior unrelated to conception.
Post-conception copulatory behaviors were observed in three out of four females usually kept in individual cages in an air-conditioned
room, and in two out of three females without infants kept in an outdoor group cage. However, neither a mature Graafian follicle
nor a fresh corpus luteum formed newly after conception was observed in any of these females by laparoscopic examinations
conducted immediately after termination of the copulatory behavior. In females with infants born in the preceding birth season,
copulatory behaviors were observed in three out of four females kept in the outdoor group cage, and in two out of four females
in a free-ranging troop. Ovulation was confirmed in one case out of the three kept in the outdoor group cage, but neither
a mature follicle nor a newly formed corpus luteum was observed in the remaining four females.
These findings suggest that copulatory behavior in the Japanese monkey is not always controlled by the development of a follicle
