水痘-带状疱疹病毒(varicella zoster virus,VZV)糖蛋白E(glycoprotein E,gE)是VZV亚单位疫苗的主要候选蛋白,但目前原核表达系统制备的gE蛋白以包涵体形式为主,可溶性差。本研究采用去除第1~30氨基酸序列的VZV gE胞外域基因,将其与原核表达载体pET32a连接,并转化至感受态细胞BL21(DE3)中。使用异丙基-β-D-硫代半乳糖苷(Isopropylβ-D-thiogalactoside,IPTG)诱导表达,His-tag柱纯化重组gE蛋白,蛋白质印迹法(Western blot,WB)检测其特异性。用该重组gE蛋白免疫BALB/c小鼠制备多克隆抗体,酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)和间接免疫荧光法检测多克隆抗体效价及特异性。结果显示,BL21/pET32a-VZV gE工程菌可以表达可溶性重组gE蛋白,纯化后纯度约为90%。WB鉴定该重组蛋白具有良好的免疫反应性。ELISA检测显示小鼠抗VZV gE多克隆抗体效价>1∶10 000,间接免疫荧光实验结果显示该抗体特异性较高。结果表明,本研究在原核表达系统中成功表达可溶性重组VZV gE蛋白,同时该蛋白具有较强的免疫原性,这为VZV gE亚单位疫苗的研制和大规模生产奠定了基础。 相似文献

Myofibrillogenesis, the process of sarcomere formation, requires close interactions of sarcomeric proteins and various components of sarcomere structures. The myosin thick filaments and M-lines are two key components of the sarcomere. It has been suggested that myomesin proteins of M-lines interact with myosin and titin proteins and keep the thick and titin filaments in order. However, the function of myomesin in myofibrillogenesis and sarcomere organization remained largely enigmatic. No knockout or knockdown animal models have been reported to elucidate the role of myomesin in sarcomere organization in vivo. In this study, by using the gene-specific knockdown approach in zebrafish embryos, we carried out a loss-of-function analysis of myomesin-3 and slow myosin heavy chain 1 (smyhc1) expressed specifically in slow muscles. We demonstrated that knockdown of smyhc1 abolished the sarcomeric localization of myomesin-3 in slow muscles. In contrast, loss of myomesin-3 had no effect on the sarcomeric organization of thick and thin filaments as well as M- and Z-line structures. Together, these studies indicate that myosin thick filaments are required for M-line organization and M-line localization of myomesin-3. The nectin and nectin-like molecule (Necl) family includes important cell adhesion molecules (CAMs) characterized by their Ig-like nature. Such CAMs regulate a broad spectrum of cell-cell interactions, including the interaction between NK cells and cytotoxic T lymphocytes (CTLs) and their target cells. CAM members nectin-2 (CD112) and Necl-5 (CD155) are believed to form homodimers (for nectin-2) or heterodimers in their functions for cell adhesion, as well as to interact with immune costimulatory receptor DNAX accessory molecule 1 (DNAM-1) (CD226) to regulate functions of both NK and CTL cells. However, the structural basis of the interactive mode of DNAM-1 with nectin-2 or Necl-5 is not yet understood. In this study, a soluble nectin-2 Ig-like V-set domain (nectin-2v) was successfully prepared and demonstrated to bind to both soluble ectodomain and cell surface-expressed full-length DNAM-1. The 1.85-? crystal structure of nectin-2v displays a perpendicular homodimer arrangement, revealing the homodimer characteristics of the nectin and Necls. Further mutational analysis indicated that disruption of the homodimeric interface of nectin-2v led to a failure of the homodimer formation, as confirmed by crystal structure and biochemical properties of the mutant protein of nectin-2v. Interestingly, the monomer mutant also loses DNAM-1 binding, as evidenced by cell staining with tetramers and surface plasmon resonance assays. The data indicate that interaction with DNAM-1 requires either the homodimerization or engagement of the homodimeric interface of nectin-2v. These results have implications for immune intervention of tumors or autoimmune diseases in the DNAM-1/nectin-2-dependent pathway. 相似文献

Mannosylerythritol (ME) is the hydrophilic backbone of mannosylerythritol lipids as the most promising biosurfactants produced by different Pseudozyma yeasts, and has been receiving attention as a new sugar alcohol. Different Pseudozyma yeasts were examined for the sugar alcohol production using glucose as the sole carbon source. P. hubeiensis KM-59 highly produced a conventional type of ME, i.e., 4-O-β-d-mannopyranosyl-d-erythritol (4-ME). Interestingly, P. tsukubaensis KM-160 produced a diastereomer of 4-ME, i.e., 1-O-β-d-mannopyranosyl-d-erythritol (1-ME). In shake flask culture with 200 g/l of glucose, strain KM-59 produced 4-ME at a yield of 33.2 g/l (2.2 g/l/day of the productivity), while strain KM-160 produced 1-ME at 30.0 g/l (2.0 g/l/day). Moreover, the two strains were found to produce ME from glycerol; the maximum yields of 4-ME and 1-ME from 200 g/l of glycerol were 16.1 g/l (1.1 g/l/day) and 15.8 g/l (1.1 g/l/day), respectively. The production of 1-ME as the new diastereomer was further investigated in fed batch culture using a 5-l jar-fermenter. Compared to the flask culture, strain KM-160 gave three times higher productivity of 1-ME at 38.0 g/l (6.3 g/l/day) from glucose and at 31.1 g/l (3.5 g/l/day) from glycerol, respectively. This is the first report on the selective production of two diastereomers of ME, and should thus facilitate the functional development and application of the disaccharide sugar alcohol in the food and relative industries. 相似文献

Photoacclimation characteristics of Sargassum thunbergii germlings under different light intensities

The photosynthesis and growth responses of Sargassum thunbergii germlings to different light intensities (10, 60, and 300 μmol photons m?2 s?1) were investigated. Maximum photochemical efficiency (F v/F m), rapid light curves (RLCs), and photochemical and non-photochemical quenching (qP and NPQ) were estimated by a pulse amplitude-modulated fluorometer. The photosynthesis of S. thunbergii germlings exhibited different properties to optimize light capture and utilization. The excitation pressure (1???qP) was rapidly increased to approximately 0.27 showing that germlings responded to high light by chronic photoinhibition with an accumulation of closed reaction centers, which ultimately resulted in a slow growth. This was accompanied by a reduced F v/F m with time and a development of high capacity for NPQ. Although F v/F m in moderate-light germlings did not fully recover overnight, germlings demonstrated a less severe chronic photoinhibition considering the reduced degree of excitation pressure accumulation of approximately 0.15. The relative stability of photosynthetic capacity (rETRmax, E k, and α) could endow germlings with the highest relative growth rate (RGR) of approximately 9.3 % day?1 in moderate light. By contrast, low-light germlings demonstrated high F v/F m and F o, corresponding high α collectively suggested greater efficiency of light absorption and energy transformation. Sustained increases in electron transport capacity (rETRmax and E k) occurred in low-light germlings, which resulted in a stable RGR of over 8.2 % day?1. Consequently, S. thunbergii germlings are considered to prefer low light regimes and have a relative capacity of moderate and high light tolerance. 为评估班氏跳小蜂Aenasius bambawalei Hayat对其寄主扶桑绵粉蚧Phenacoccus solenopsis Tinsley的控制作用, 在实验室条件下(温度25±1℃, 相对湿度70%±5%, 光周期14L∶10D), 研究了班氏跳小蜂对扶桑绵粉蚧3龄若虫、 雌成虫的寄生功能反应及其自身密度效应。结果表明: 寄生功能反应均符合Holling Ⅱ型方程, 且受寄主密度和寄生物密度的影响。当扶桑绵粉蚧 3龄若虫和雌成虫的密度分别大于15头/容器和10头/容器时, 班氏跳小蜂的寄生量增加幅度开始减小; 以瞬间攻击率/处理时间(α/Th)为评价指标, 班氏跳小蜂雌蜂寄生扶桑绵粉蚧3龄若虫时, α/Th为21.1307, 且大于雌蜂寄生雌成虫的情况(α/Th为6.2506); 班氏跳小蜂寄生功能反应有较强的种内干扰作用, 随着自身密度的增加, 寄生效能逐渐降低; 通过方程E=QP-m能很好地模拟班氏跳小蜂的寻找效应(E)与其自身密度(P)之间的关系, 对于3龄若虫和雌成虫的模拟结果分别为E=0.2931P-0.6240和E=0.0944P-0.4840。本研究为利用班氏跳小蜂开展扶桑绵粉蚧生物防治提供了基础数据和方法。 相似文献
