Dolichol-phosphate mannose (Dol-P-Man) is a key mannosyl donor for the biosynthesis of N-linked oligosaccharides as well as for O-linked oligosaccharides on yeast glycoproteins, and for the synthesis of the glycosyl-phosphatidylinositol anchor found on many cell surface glycoproteins. It is synthesized by Dol-P-Man synthase which is the only glycosyltransferase in the dolichol pathway that has been expressed as an active protein, solubilized and purified in large enough quantities for structural investigations. Earlier studies showed that the enzyme is closely associated with membranes of endoplasmic reticulum with unique lipid requirements for its maximal activity. This potential target of antibiotic therapy is now being investigated at the molecular level to establish information about the structure of the enzyme as well as determine the nature and properties of the enzyme-phospholipid interactions. In this paper, we have determined the activities of the fluorescent labeled dolichyl-phosphate derivatives as well as the intramolecular distances between amino acid residues near the active site and/or the fluorophores of the substrate derivatives using fluorescence energy resonance transfer. These results also show that the conserved consensus sequence is not required by Dol-P-Man synthase neither for the recognition of Dol-P nor for the catalytic activity.  相似文献

Parental selection is crucial for hybrid breeding, but the methods available for such a selection are not very effective. In this study, a 6×6 incomplete diallel cross was designed using 12 rapeseed germplasms, and a total of 36 hybrids together with their parental lines were planted in 4 environments. Four yield-related traits and seed oil content (OC) were evaluated. Genetic distance (GD) was estimated with 359 simple sequence repeats (SSRs) markers. Heterosis levels, general combining ability (GCA) and specific combining ability (SCA) were evaluated. GD was found to have a significant correlation with better-parent heterosis (BPH) of thousand seed weight (TSW), SCA of seeds per silique (SS), TSW, and seed yield per plant (SY), while SCA showed a statistically significant correlation with heterosis levels of all traits at 1% significance level. Statistically significant correlations were also observed between GCA of maternal or paternal parents and heterosis levels of different traits except for SS. Interestingly, maternal (TSW, SS, and OC) and paternal (siliques per plant (SP) and SY) inheritance of traits was detected using contribution ratio of maternal and paternal GCA variance as well as correlations between GCA and heterosis levels. Phenotype and heterosis levels of all the traits except TSW of hybrids were significantly correlated with the average performance of parents. The correlations between SS and SP, SP and OC, and SY and OC were statistically significant in hybrids but not in parents. Potential applications of parental selection in hybrid breeding were discussed.  相似文献

目的用生物芯片技术分析胚胎干细胞定向分化为神经干细胞过程中microRNA(miRNA)的表达变化,筛选调控的分化的miRNA,研究分化调控机制。方法胚胎干细胞在含LIF培养基中培养3d后,采用经典5步培养方法定向诱导向神经干细胞分化,采用nestin作为神经干细胞标记进行鉴定,送检胚胎干细胞及神经干细胞,提取总RNA以及小分子RNA,经荧光标记后与miRNA基因芯片杂交,获得胚胎干细胞诱导前后miRNA表达谱。结果1)胚胎干细胞在含LIF培养过程中保持未分化状态,Oct-4、碱性磷酸酶表达阳性;2)经典五步法诱导胚胎干细胞定向分化为神经干细胞,nestin阳性细胞为85%;3)通过基因微阵列分析,有90个miRNA的改变显著,其中68个表达上调,22个表达下调。结论miRNA可能对胚胎干细胞定向分化为神经干细胞过程起到关键作用。  相似文献

Dendroctonus valens is an invasive pest in coniferous forests of northern China. It was suspected of being responsible for the death of more than three million Pinus tabuliformis trees. The present study sought to identify the ophiostomatoid fungi associated with D. valens in northern China and understand the possible role of these fungi in the pine decline. On the basis of morphology, physiology, mating compatibility and phylogenetic analyses of multiple DNA sequences, seven species of ophiostomatoid fungi were isolated from and around D. valens galleries: Leptographium alethinum, Grosmannia koreana (teleomorph of L. koreanum), L. procerum, L. sinoprocerum, L. truncatum, Pesotum aureum and P. pini. All have been recorded for the first time in China. Among them, the occurrence of the dominant species L. procerum is positively linked to attack intensities of D. valens. The pathogenicity of four species (L. koreanum, L. procerum, L. sinoprocerum and L. truncatum) was tested on mature P. tabuliformis trees by stem inoculation. All inoculated strains caused significant necrotic lesions on the inner bark. However, L. koreanum and L. truncatum induced more extensive lesions than L. procerum and L. sinoprocerum. Their association with D. valens and the P. tabuliformis decline is discussed.  相似文献

The purpose of this study was to use metabonomic profiling to identify a potential specific biomarker pattern in urine as a noninvasive bladder cancer (BC) detection strategy. A liquid chromatography-mass spectrometry based method, which utilized both reversed phase liquid chromatography and hydrophilic interaction chromatography separations, was performed, followed by multivariate data analysis to discriminate the global urine profiles of 27 BC patients and 32 healthy controls. Data from both columns were combined, and this combination proved to be effective and reliable for partial least squares-discriminant analysis. Following a critical selection criterion, several metabolites showing significant differences in expression levels were detected. Receiver operating characteristic analysis was used for the evaluation of potential biomarkers. Carnitine C9:1 and component I, were combined as a biomarker pattern, with a sensitivity and specificity up to 92.6% and 96.9%, respectively, for all patients and 90.5% and 96.9%, respectively for low-grade BC patients. Metabolic pathways of component I and carnitine C9:1 are discussed. These results indicate that metabonomics is a practicable tool for BC diagnosis given its high efficacy and economization. The combined biomarker pattern showed better performance than single metabolite in discriminating bladder cancer patients, especially low-grade BC patients, from healthy controls.  相似文献

Cynandione A, an acetophenone from the roots of Cynanchum auriculatum and other species in the genus attenuates neurotoxicity of a variety of neurotoxic agents such as l-glutamate in vitro. In this study, we sought to further characterize the neuroprotective effects of cynandione A and other acetophenones from the roots of C. auriculatum in pheochromocytoma tumor cell line PC12 and investigate whether cynandione A protected against ischemic injuries in rats with experimentally induced cerebral ischemia. Viability assays using the 2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophen-yl)-2H-tetrazolium monosodium salt method and lactate dehydrogenase (LDH) release assays showed that cynandione A dose-dependently attenuated glutamate-induced cytotoxicity. Comparative proteomic analysis by two-dimensional gel electrophoresis and matrix-assisted laser desorption/ionization-time of flight MS/MS of PC12 cells treated with cynandione A showed 10 μM cynandione A caused broad changes in protein expression in PC12 cells including down-regulation of high mobility group box 1 (HMGB1) and dihydropyrimidinase-like 2 (DPYSL2). Immunoblotting studies showed that 10 μM cynandione A aborted glutamate-induced increase in DPYSL2 and HMGB1 levels in PC12 cells and 30 mg/kg cynandione A also attenuated the rise in HMGB1 levels and mitigated DPYSL2 cleavage in brain tissues of rats with cerebral ischemia. Furthermore, rats with cerebral ischemia treated with 30 mg/kg cynandione A exhibited markedly improved neurological deficit scores at 24 and 72 h compared with control and a 7.2% reduction in cerebral infarction size at 72 h (p < 0.05 vs. control). Our findings demonstrated that cynandione A mitigated ischemic injuries and should be further explored as a neuroprotective agent for ischemic stroke.  相似文献

植物种子从母树掉落形成土壤种子库时,凋落物或土壤是其最初接触的物理环境,种子所处位置(种子在凋落物上层、土壤表层或凋落物下层)影响了幼苗天然更新进程。模拟格氏栲种子在凋落物上层(种子下层铺垫2和4 cm凋落物)、土壤表层(无凋落物)及凋落物下层(种子上层覆盖2、4、6和8 cm凋落物)等3种不同散布位置,探讨种子散布位置对幼苗叶绿素荧光特性、非结构性碳水化合物、比叶面积、叶干物质含量和养分含量的影响。结果表明:不同散布位置的幼苗单位面积的叶氮含量与可溶性糖、非结构性碳水化合物含量呈显著正相关,与比叶面积呈显著负相关。适宜凋落物覆盖(2和4 cm)的幼苗通过提高叶绿素相对含量、可溶性糖含量、非结构性碳水化合物含量、叶干物质含量和单位面积的叶氮含量和叶磷含量,降低比叶面积等的资源获取策略来实现自身快速生长需求。无凋落物和深层凋落物覆盖(6和8 cm)的幼苗采取高单位重量的叶氮含量和比叶面积,低叶干物质含量和非结构性碳水化合物含量的资源保守型策略以截获更多有效光资源,进而弥补深层凋落物带来的郁闭环境,降低幼苗因“碳饥饿”而死亡的几率。下层铺垫凋落物的幼苗通过在叶片储藏淀粉,降低叶片光合组织消耗能量(低PSⅡ最大光化学效率)等维持幼苗生长。熵值法综合分析表明,浅层凋落物覆盖(2 cm)对格氏栲幼苗生长的促进作用最为显著,未来可通过调节天然林凋落物层厚度以促进格氏栲幼苗生长与更新。  相似文献
