Application of phosphatic (P) fertilizers and biosolids is known to enhance cadmium (Cd) contamination in saline soils. Increased concentration of dissolved chloride (Cl?) in soil solution significantly influences Cd bioavailability in P fertilizer- or biosolid-amended soils. Arbuscular mycorrhizal (AM) fungi have an ability to protect plants against salinity and heavy metals by mediating interactions between toxic ions and plant roots. The effects of Glomus mosseae (AM) and NaCl and Cd stresses on Cd uptake and osmolyte and phytochelatin (PCs) synthesis in Cajanus cajan (L.) Millsp. (pigeonpea) were studied under greenhouse conditions. Two genotypes [Sel 85?N (tolerant) and ICP 13997 (sensitive)] were subjected to NaCl (4 and 6?dS?m?1) and Cd (CdCl2, 25 and 50?mg?kg?1 dry soil) treatments. NaCl and Cd applied individually as well as in combination caused dramatic reductions in plant biomass and induced membrane peroxidation, ionic perturbations, and metabolite synthesis in both genotypes, although Sel 85?N was less affected than ICP 13997. Cadmium uptake was enhanced when NaCl was added along with Cd. The protection of growth in Sel 85?N was associated with restricted accumulation of Na+, Cl?, and Cd2+ and higher concentrations of stress metabolites (sugars, proteins, free amino acids, proline, glycine betaine). Cd led to a significant increase in biothiols (NP-SH) and glutathione (GSH), with a larger pool of NP-SH which strongly induced accumulation of phytochelatins, whereas no significant effects in their concentrations were detectable under NaCl stress. The interactive effects of NaCl and Cd on all parameters were larger than those of individual treatments. Fungal inoculations improved plant growth and reduced accumulation of toxic ions. Higher stress metabolite synthesis and PCs observed in AM plants of Sel 85?N indicated the role of an efficient AM symbiosis capable of attenuating NaCl and Cd stresses. 相似文献

A method is described for the construction of a highly sensitive amperometric sensor for the detection of tyrosine, employing a magnetic nanoparticles-zinc oxide/zinc hexacyanoferrate (Fe3O4NP-ZnO/ZnHCF) hybrid film electrodeposited on the surface of a Pt electrode as working electrode. The sensor is based on electrocatalytic mechanism initiated by electrochemical oxidation of the reduced form of the hybrid film at +0.2 V vs. Ag/AgCl followed by completion of chemical oxidation of tyrosine. The sensor showed optimum response within 2 s at pH 2. The working/linear range of the sensor was 0.02–2.76 mM with a detection limit of 4 μM. The sensor measured tyrosine level in serum, a potential biomarker of phenylketonuria. The working electrode lost only 5 % of its initial activity, when stored at 4 °C, after its regular use over a period of 100 days. 相似文献

Sperm cells exhibit extremely high sensitivity in response to slight changes in temperature, osmotic pressure and/or presence of various chemical stimuli. In most cases throughout the evolution, these physico-chemical stimuli trigger Ca2+-signaling and subsequently alter structure, cellular function, motility and survival of the sperm cells. Few reports have recently demonstrated the presence of Transient Receptor Potential (TRP) channels in the sperm cells from higher eukaryotes, mainly from higher mammals. In this work, we have explored if the sperm cells from lower vertebrates can also have thermo-sensitive TRP channels. In this paper, we demonstrate the endogenous presence of one specific thermo-sensitive ion channel, namely Transient Receptor Potential Vanilloid family member sub type 1 (TRPV1) in the sperm cells collected from fresh water teleost fish, Labeo rohita. By using western blot analysis, fluorescence assisted cell sorting (FACS) and confocal microscopy; we confirm the presence of this non-selective cation channel. Activation of TRPV1 by an endogenous activator NADA significantly increases the quality as well as the duration of fish sperm movement. The sperm cell specific expression of TRPV1 matches well with our in silico sequence analysis. The results demonstrate that TRPV1 gene is conserved in various fishes, ranging from 1–3 in copy number, and it originated by fish-specific duplication events within the last 320 million years (MY). To the best of our knowledge, this is the first report demonstrating the presence of any thermo-sensitive TRP channels in the sperm cells of early vertebrates as well as of aquatic animals, which undergo external fertilization in fresh water. This observation may have implications in the aquaculture, breeding of several fresh water and marine fish species and cryopreservation of fish sperms. 相似文献

Brucellosis is a worldwide zoonotic disease. No Brucella vaccine is available for use in humans, and existing animal vaccines have limitations. There is a need to develop a safe and effective vaccine against human and animal brucellosis. In the present study, we generated recombinant cysteine synthase A (rCysK) of Brucella abortus in Escherichia coli and purified it up to homogeneity by metal affinity chromatography. The immunogenicity and protective efficacy of purified rCysK were evaluated in BALB/c mice with Freund’s adjuvant, aluminium hydroxide gel or without any adjuvant. High titres of anti-rCysK IgG antibody predominated by IgG1 were observed in all immunized mice. After stimulation with rCysK, the spleen lymphocytes of mice immunized with CysK formulated with aluminium hydroxide gel produced significant levels of IFN-γ. Protection against challenge with virulent B. abortus strain 544 was determined in BALB/c mice, and significant protection was observed in all CysK immunized groups when compared with PBS control. Among all the CysK vaccine groups, comparatively better protection was observed in mice immunized with aluminium hydroxide gel (1.064 units of protection). Overall, the results of the study suggest that rCysK induces primarily Th2 type of immune response and provides partial protection against B. abortus challenge. 相似文献

This paper reports, the Laser Induced Breakdown Spectroscopy (LIBS) studies and structure elucidation of compounds isolated
from the fruit extract of Moringa oleifera and also deals with their possible effects on some bacterial strains viz. Staphylococcus aureus, Klebsiella pneumonia, Escherichia coli and Pseudomonas aeruginosa. The extract was found to be active against all four microorganisms used. Extent of inhibitory effect of extract was assessed
at different concentrations of 25, 50, 75 mg/ml by measuring diameter of inhibition zone (DIZ). Our results clearly showed
that the 75 mg/ml concentration of the extract had 14, 12 and 18 mm of the DIZ against Staphylococcus aureus, Klebsiella pneumonia and Pseudomonas aeruginosa and 14 mm with 50 mg/ml concentration against Escherichia coli. The results were compared with the standard antibiotic ‘ampicillin’ of 1 mg/ml concentration. LIBS was recorded with high
power pulsed laser beam from Nd: YAG Laser (Continuum Surelite III-10), focused on the surface of the material, which was
in liquid form, to generate plasma on the surface of the sample. LIBS data clearly demonstrate the presence of trace elements,
magnesium and iron, in high concentration in the extract. Whereas, from the phytochemical profile reveals the presence of
two new compounds, S-ethyl-N-{4-[(α-L-rhamnosyloxy) benzyl]} thiocarbamate and 2-acetoxy {4-[(2′,3′,4′-tri-O-acetyl-α-L-rhamnosyloxy)
benzyl]} acetonitrile as the major constituents. This study is the first report on synergetic effect of the phytoconstituents
