Light critically affects the physiology of plants. Using two-dimensional gel electrophoresis, we used a proteomics approach to analyze the responses of Arabidopsis thaliana to red (660 nm), far-red (730 nm) and blue (450 nm) light, which are utilized by type II and type I phytochromes, and blue light receptors, respectively. Under specific light treatments, the proteomic profiles of 49 protein spots exhibited over 1.8-fold difference in protein abundance, significant at p <0.05. Most of these proteins were metabolic enzymes, indicating metabolic changes induced by light of specific wavelengths. The differentially-expressed proteins formed seven clusters, reflecting co-regulation. We used the 49 differentially-regulated proteins as molecular markers for plant responses to light, and by developing a procedure that calculates the Pearson correlation distance of cluster-to-cluster similarity in expression changes, we assessed the proteome-based relatedness of light responses for wild-type and phytochrome mutant plants. Overall, this assessment was consistent with the known physiological responses of plants to light. However, we also observed a number of novel responses at the proteomic level, which were not predicted from known physiological changes.  相似文献

This study aimed to investigate precision of a newly developed gold nanoparticle-based immunoassay for measuring urinary N-telopeptides (NTx) and its correlation with ELISA measurement. NTx concentrations were determined by absorption changes using spectroscopy at 535 nm, which were measured three times with an interval of two hours for precision test, using urine samples of 60 subjects (32 males and 28 females; mean age 30.4 years, SD 21.9 years). The correlation of the immunoassay with the measurement of ELISA kit was also analyzed. Precision of the immunoassay and its correlation with ELISA were 0.924 (ICC, 95% confidence interval, 0.661-0.971) and 0.90 (p<0.001). Our findings suggested that this immunoassay of urinary NTx could be used as a means of screening for bone resorption and point-of-care testing.  相似文献

Synaptic response to regular stimulation of midbrain and bulbar locomotor sites (LS) and a pontine inhibitory site (IS) was recorded in medial and lateral bulbar neurons in cats (mesencephalic decerebellate preparation). Excitatory post-synaptic potentials (PSP) and discharges were usually noted in medial neurons; mixed PSP also occurred when stimulating the IS. Almost 50% of lateral and over 25% of medial neurons showed a change in background firing rate, failing to generate response time-locked to stimulus. Medial neurons producing a response time-locked to the stimulus showed equal sensitivity to stimulation of midbrain and bulbar LT and very little reaction to IS stimulation. Medial neurons with a response not time-locked to stimuli together with lateral neurons were most receptive to input from the bulbar LS, less sensitive to stimulation of the midbrain LS, and least responsive of all to IS stimulation. Convergence between influences from midbrain and bulbar LS was the same in neurons of all populations. Nodakenin is a coumarin compound initially isolated from the roots of Angelica gigas. In the present study, we investigated the effects of nodakenin on learning and memory impairments induced by scopolamine (1 mg/kg, i.p.) using the passive avoidance test, the Y-maze test, and the Morris water maze test in mice. Nodakenin (10 mg/kg, p.o.) administration significantly reversed scopolamine-induced cognitive impairments in the passive avoidance test and the Y-maze test (P<0.05), and also reduced escape latency during training in the Morris water maze test (P<0.05). Moreover, swimming times and distances within the target zone of the Morris water maze were greater in the nodakenin-treated group than in the scopolamine-treated group (P<0.05). In an in vitro study, nodakenin was found to inhibit acetylcholinesterase activity in a dose-dependent manner (IC(50)=84.7 microM). In addition, nodakenin was also found to inhibit acetylcholinesterase activity for 6 h in an ex-vivo study. These results suggest that nodakenin may be a useful for the treatment of cognitive impairment, and that its beneficial effects are mediated, in part, via the enhancement of cholinergic signaling.  相似文献

Hyperhomocysteinemia is associated with the risk of skeletal health problems, such as osteoporosis, low body mineral density, and fracture. 5, 10-Methylenetetrahydrofolate reductase (MTHFR) and thymidylate synthase (TS) are involved in homocysteine metabolism. We hypothesized that certain genetic polymorphisms of MTHFR and TS that cause altered enzyme activity may lead to hyperhomocysteinemia and affect bone metabolism. Therefore, we determined whether MTHFR 677C>T, MTHFR 1298A>C, TS enhancer region (TSER), and TS 3??-UTR 6 bp insertion/deletion polymorphisms are associated with osteoporotic vertebral compression fracture (OVCF) in postmenopausal Korean women. A total of 308 postmenopausal Korean women were enrolled as study subjects. Among them, 84 were patients with OVCF and 224 were controls. The polymorphisms were analysed by PCR-RFLP methods. Single mutations of MTHFR or TS were not associated with the occurrence of OVCF. However, the combined genotypes 2R3R+2R2R/0bp6bp+6bp6bp (TSER/TS 3??-UTR) and AC+CC/0bp6bp+6bp6bp (MTHFR 1298A>C/TS 3??-UTR) were associated with decreased risk for OVCF. 2R-0bp and 2R-6bp haplotype frequencies of TS were significantly different between the cases and controls. In the present study, the combined genotype of TSER/TS 3??-UTR and MTHFR 1298A>C/TS 3??-UTR was associated with a decreased risk for OVCF in postmenopausal Korean women. However, due to the several limitations of our study including the moderately small sample size, our findings should be considered with caution and further research is needed to draw more definitive conclusions.  相似文献

The soluble β-glucan contents in the cell wall of yeasts were estimated by treating cells with Glucanex® 200G that contained mainly β1,3-glucanase and some β1,6-glucanase. The sensitivity of cell walls of 11 yeasts to various concentrations of β-glucanase was compared. The yeasts that are resistant to β-glucanase treatment are expected to contain higher β-glucan content and those that are sensitive to the β-glucanase treatment are expected to contain lower β-glucan content. Two yeast strains were selected for further study by comparing the sensitivity of cell wall to β-glucanase; Candida bombicola and Candida albicans. Candida bombicola was more resistant and C. albicans was more sensitive to the Glucanex® 200G treatment. The results of enzyme sensitivity tests were verified by quantification of soluble β-glucan content purified from the yeasts. Much larger amount of soluble β-glucan was obtained from the cell walls of C. bombicola (0.08 g g−1 dried cell) than C. albicans (0.025 g g−1 dried cell).  相似文献
