In epidermis, Ras can influence proliferation and differentiation; however, regulators of epidermal Ras function are not fully characterized, and Ras effects on growth and differentiation are controversial. EGF induced Ras activation in epidermal cells along with phosphorylation of the multisubstrate docking protein Gab1 and its binding to SHP-2. Expression of mutant Gab1Y627F deficient in SHP-2 binding or dominant-negative SHP-2C459S reduced basal levels of active Ras and downstream MAPK proteins and initiated differentiation. Differentiation triggered by both Gab1Y627F and SHP-2C459S could be blocked by coexpression of active Ras, consistent with Gab1 and SHP-2 action upstream of Ras in this process. To study the role of Gab1 and SHP-2 in tissue, we generated human epidermis overexpressing active Gab1 and SHP-2. Both proteins stimulated proliferation. In contrast, Gab1Y627F and SHP-2C459S inhibited epidermal proliferation and enhanced differentiation. Consistent with a role for Gab1 and SHP-2 in sustaining epidermal Ras/MAPK activity, Gab1-/- murine epidermis displayed lower levels of active Ras and MAPK with postnatal Gab1-/- epidermis, demonstrating the hypoplasia and enhanced differentiation seen previously with transgenic epidermal Ras blockade. These data provide support for a Ras role in promoting epidermal proliferation and opposing differentiation and indicate that Gab1 and SHP-2 promote the undifferentiated epidermal cell state by facilitating Ras/MAPK signaling. 相似文献

Previous work of del Castillo and co-workers has shown that bilayer lipid membranes (BLM) can be used as transducers for detection of antigen-antibody reactions. The present experiments extend the previous work by incorporating complement into the BLM system. The results indicate that the antigen-antibody complex or the complement has no ability to affect the BLM system separately, but when carefully combined they will destabilize the BLM even at a much reduced concentration. Traumatic experience has overall far reaching consequences on personality. In particular, it has significant impact on teenagers that are just approaching the phase of solving their identity problems. This research examines the relation of traumatic experience and attitude towards the future in two groups of adolescents. The first group consists of 20 adolescents-refugees from the East Slavonia that were settled in Rijeka area with their parents during the last six years. The second group consists of 20 adolescent's local inhabitants that were influenced by the war only indirectly. Results show significant difference between refugees and non-refugees in expressed interpersonal trust, frustration tolerance, and formation of close contacts, adaptability, precaution, bitterness, and social desirability. In the present study, the performances of conventional purification methods, packed bed adsorption (PBA), and expanded bed adsorption (EBA) for the purification of the nucleocapsid protein (NP) of Newcastle disease virus (NDV) from Escherichia coli homogenates were evaluated. The conventional methods for the recovery of NP proteins involved multiple steps, such as centrifugation, precipitation, dialysis, and sucrose gradient ultracentrifugation. For the PBA, clarified feedstock was used for column loading, while in EBA, unclarified feedstock was used. Streamline chelating immobilized with Ni2+ ion was used as an affinity ligand for both PBA and EBA. The final protein yield obtained in conventional and PBA methods was 1.26% and 5.56%, respectively. It was demonstrated that EBA achieved the highest final protein yield of 9.6% with a purification factor of 7. Additionally, the total processing time of the EBA process has been shortened by 8 times compared to that of the conventional method. 相似文献

MOTIVATION: Missing data in genotyping single nucleotide polymorphism (SNP) spots are common. High-throughput genotyping methods usually have a high rate of missing data. For example, the published human chromosome 21 data by Patil et al. contains about 20% missing SNPs. Inferring missing SNPs using the haplotype block structure is promising but difficult because the haplotype block boundaries are not well defined. Here we propose a global algorithm to overcome this difficulty. RESULTS: First, we propose to use entropy as a measure of haplotype diversity. We show that the entropy measure combined with a dynamic programming algorithm produces better haplotype block partitions than other measures. Second, based on the entropy measure, we propose a two-step iterative partition-inference algorithm for the inference of missing SNPs. At the first step, we apply the dynamic programming algorithm to partition haplotypes into blocks. At the second step, we use an iterative process similar to the expectation-maximization algorithm to infer missing SNPs in each haplotype block so as to minimize the block entropy. The algorithm iterates these two steps until the total block entropy is minimized. We test our algorithm in several experimental data sets. Homocysteine, cytokines (IL-18, IL-6, IL-8) are involved in vascular inflammation and coronary artery disease. Homocysteine influences endothelial IL-6 and IL-8 cytokine expression and release, however, an association between homocysteine and IL-18 has not been previously investigated in endothelial/smooth muscle cells and or in coronary artery disease. We report in 9 coronary artery bypass surgery (CABG) patients a positive correlation r = 0.86 between homocysteine and IL-18 plasma levels (p < 0.05). Plasma IL-18 levels are significantly higher in those patients with elevated homocysteine compared to those with normal levels (p < 0.02; 153 +/- 19 pg/ml versus 116 +/- 14 pg/ml respectively). Our in vitro cell culture studies suggest that the source of IL-18 in CABG patients with elevated homocysteine is not from vascular smooth muscle or endothelial cells. 相似文献

本文将某一种小兽 (小型哺乳动物 )宿主体表的全部蚤作为一个特定的蚤类群落 ,在此基础上运用模糊聚类分析方法对云南人间鼠疫流行区 10种小兽体表的蚤类群落之间的相似性进行了比较。研究结果表明 ,在动物分类上比较接近的、隶属同一属 (鼠属 )的黄胸鼠、大足鼠和斯氏家鼠体表 3种蚤类群落之间的相似性较大 ,在模糊聚类的谱系图上明显聚为一类。动物分类上距离较远、隶属不同属的微尾、臭、灰麝、中华姬鼠、滇绒鼠、社鼠和锡金小鼠体表的 7种蚤类群落之间的相似性较小。结果提示 ,宿主因素可能是影响体表寄生蚤群落之间相似性大小的一个主要因子 ,当宿主在动物分类上彼此接近时 ,其体表寄生蚤群落的相似程度就比较高 ,当宿主在动物分类上距离较大时 ,其体表蚤类群落的相似程度就比较低。本文中不同属小兽体表寄生蚤类形成了不同的群落类型。 相似文献
