A new lupin alkaloid, (+)-5,17-dehydromatrine N-oxide, was isolated from the fresh aerial parts of Euchresta japonica. Its structure was confirmed by spectrometric data and by direct comparison with a synthetic sample, prepared from (+)-sophoranol ((+)-5-hydroxymatrine). It was also concluded that (+)-5,17-dehydromatrine N-oxide and (+)-matrine N-oxide possess the same configuration with respect to the asymmetric nitrogen by NMR spectra. 相似文献

An enzyme, dUTPase, that catalyzes the conversion of dUTP to dUMP and PPi, was partially purified from regenerating rat livers. The molecular weight was estimated by gel filtration to be 60,000. The apparent Km for dUTP was 12 μM. No other deoxyribonucleoside triphosphates served as a substrate. This enzyme is active in the absence of added divalent cations or sulfhydryl reagents; the activity could be inhibited by EDTA and shows a broad pH optimum with no decrease in activity from pH 7 to 11. The specific activity of dUTPase in rat liver begins to rise 16 h after partial hepatectomy and reaches a maximum about 24 h after the operation, rising to at least 5 to 6 times the normal level. 相似文献

A new culture method for the injection of tobacco mesophyll protoplasts has been established. The protoplasts are embedded
in a thin layer of alginate and are nourished from the medium in the underlying basislayer. In the alginate layer the protoplasts
regenerate to calli at a frequency of up to 80%. Embedded protoplasts can be selected either with 50 mg l−1 kanamycin or 5 mg l−1 paromomycin. Single resistant cells can be recovered from about 10 000 sensitive cells in one alginate layer. Injection of
theneo gene (coding for neomycin phosphotransferase II) into protoplast derived single cells in the alginate layer results in kanamycin
resistant colonies that can be regenerated to mature plants. These plants express the neomycin phosphotransferase as shown
by enzyme activity assay. The integration of the transgene into the plant genome could be proved by Southern hybridization
to high molecular weight DNA. With this culture method 100 cells can be injected per hour. Transformation frequencies range
from 2 to 20%. In crossing experiments, it was shown that the foreign gene is transmitted to the next generation in a Mendelian

A highly thermostable neutral protease was found in culture filtrates ofBacillus stearothermophilus. The optimum reaction pH and temperature of this protease were 6.0 and 60°C, respectively, and 90% activity remained even after heat treatment at 90°C for 30 min. The protease was markedly inactivated by diisopropyl fluorophosphate, but EDTA and iodoacetic acid hardly affected it. A new catshark,Scyliorhinus tokubee sp. nov., is described based on specimens from the coast of Shirahama, eastern Izu Peninsula, southern Japan. The present species is distinguished from other congeners in having a particular coloration with dark saddles, blotches and numerous small light spots, a wide oral cleft, the anterior nasal flap not reaching the oral cleft, a short interspace between the dorsal fins, developed clasper hooks, and some meristic characters (number of vertebrae, jaw teeth, and spiral valve turns). Summary Lipase from Rhizopus delemar was immobilized by entrapment with photo-crosslinkable resin prepolymers or urethane prepolymers or by binding to various types of porous silica beads. The immobilized lipase preparations thus obtained were examined for their activity in converting olive oil to an interesterified fat (cacao butter-like fat), whose oleic acid moieties at 1- and 3-positions were replaced with stearic acid moieties, in the reaction solvent n-hexane. Although all of the immobilized preparations exhibited some activity, lipase adsorbed on Celite and then entrapped with a hydrophobic photo-crosslinkable resin prepolymer showed the highest activity, about 75% of that of lipase simply adsorbed onto Celite. (1) Three analogs of merocyanine dyes added to suspensions of chromatophore vesicles showed absorbance changes responding to the change in surface potential induced by salt addition and to the change in membrane potential induced by illumination. (2) The extent of the light-induced absorbance changes of the dyes was linearly related, in the presence and absence of uncouplers, to that of carotenoid spectral shift which is an intrinsic probe of the intramembrane electric field. (3) Comparison of the merocyanine absorbance changes induced by salt addition with those induced by illumination indicated that the surface potential change in the outer surface of chromatophore membranes during illumination was very small. (4) Judging from the spectra of these absorbance and from the low permeabilities of the dyes to membrane, the absorbance change are attributed to change in distribution of the dyes between the medium and the outer surface region in chromatophore membranes. The extent of the light-induced absorbance changes of merocyanine dyes depended on the salt concentration of the medium. The types of dependence were different among three merocyanine analogs. This is explained by the mechanism mentioned above assuming appropriate parameters. It is suggested that, under continuous illumination, an equilibrium of the electrochemical potential of H+ is reached between the bulk aqueous phase and the outer surface region in the membrane where the merocyanine dyes are distributed. 相似文献

In a liquid culture of human bone marrow, the development of fibroblast colonies takes place on days 6 to 9. Twenty percent fetal calf serum is used as the stimulus for fibroblast colony growth. Human bone marrow cells are plated as 2 × 105 cells in the culture. Normal human bone marrow yields 47 ± 4 fibroblasts colonies per 2 × 105 cells plated. Bone marrow fibroblast cultures using agar or methylcellulose restrict colony formation. Marked colony suppression was observed in acute leukemia, and a discrete colony number was observed in hypoplastic anemia. This fibroblast culture method should be applied to a larger number of patients to determine whether it has a pathognomonic value and clinical significance. 相似文献

Two new cage-type lupin alkaloids, (?)-tsukushinamine-B and tsukushinamine-C, have been isolated from the fresh epigeal parts of Sophora franchetiana, along with (?)-cytisine, (?)-N-formylcytisine, (?)-rhombifoline, (?)-anagyrine, (?)-baptifoline and (±)-ammodendrine, as well as (?)-tsukushinamine-A. The structures of these novel tsukushinamine-type lupin alkaloids were determined by spectroscopic data and partly by a chemical reaction.
