Cortactin is an F‐actin binding protein that has been suggested to play key roles in various cellular functions. Here, we generated mice carrying floxed alleles of the cortactin (Cttn) gene (Cttnflox/flox mice). Expression of Cre recombinase in mouse embryonic fibroblasts (MEFs) isolated from Cttnflox/flox embryos depleted cortactin within days, without disturbing F‐actin distribution and localization of multiple actin‐binding proteins. Cre‐mediated deletion of Cttn also did not affect cell migration. To obtain mice with a Cttn null allele, we next crossed Cttnflox/flox mice with transgenic mice that express Cre recombinase ubiquitously. Western blot and immunocytochemical analysis confirmed complete elimination of cortactin expression in MEFs carrying homozygously Cttn null alleles. However, we found no marked alteration of F‐actin organization and cell migration in Cttn null‐MEFs. Spatial pattern changes in aboveground plant biomass in a grazing pasture   总被引:1,自引:0,他引:1  
Using gamma distribution and spatial autocorrelation, it was demonstrated that plant biomass per unit area of a pasture grazed by cattle exhibited two kinds of spatial heterogeneity: small-scale heterogeneity caused by grazing and large-scale heterogeneity caused by topography, land aspect, etc. For each of the 10 measurement times from May to August, 100 quadrats 50cm × 50cm were arranged along a straight line 50m long in a pasture, and the plants within the quadrats were harvested at the height of 3cm above the ground surface to measure the dry weight. The data were aggregated into frequency distributions, and gamma distribution and the parameter values were estimated. This analysis showed that with the progression of grazing the amount of biomass decreased and the degree of spatial heterogeneity in biomass, measured per 0.25m2, increased, and due to plant regrowth the trends were reversed. By rearranging the 100 biomass data in order of weight, it was suggested that plots with an extremely large biomass were not grazed by cattle and remained in the pasture. For the same data, variations of biomass along the straight line were divided into two parts based on the moving average: the spatial trend and the residuals which cannot be explained by the trend. In this analysis, 48–75% of the total spatial variation was explained by the trend along the straight line. Analysis using spatial autocorrelation for the actual biomass changes showed that the biomass changes within a range of about 10m on the straight line gave a positive correlation, which indicates a topographical trend in biomass. Spatial autocorrelation for residuals suggested that the spatial changes in biomass along the straight line followed a wave-like or checker-board pattern. Small-scale spatial heterogeneity in plant biomass may be caused by the uneven deposition of excreta by grazing animals, uneven use of the grassland by grazing animals, and uneven dispersal of plant seeds through faeces over the grassland. The possibility that such unevenness might accelerate energy flow in the grassland ecosystem and contribute to grassland sustainability is discussed.  相似文献

The purpose of this study is to differentiate roles of several growth factors and cytokines in proliferation and differentiation of pulp cells during development and repair. In human pulp cell cultures, laminin and type I collagen levels per cell remained almost constant during the whole culture period (22 days). On the other hand, secreted protein, acidic and rich in cysteine (SPARC/osteonectin) and alkaline phosphatase (ALPase) levels markedly increased after the cultures reached confluence. Laminin and type I collagen, as well as fibronectin, stimulated the spreading of pulp cells within 1 h. Adding transforming growth factor-β (TGF-β) decreased laminin and ALPase levels, whereas it increased SPARC and fibronectin levels 3- to 10-fold. Western and Northern blots showed that TGF-β enhanced SPARC synthesis at the protein and mRNA levels. Basic fibroblast growth factor (bFGF) decreased type I collagen, laminin, SPARC, and ALPase levels without changing the fibronectin level. Platelet-derived growth factor (PDGF) selectively decreased laminin, SPARC, and ALPase levels. Epidermal growth factor (EGF) also decreased SPARC and ALPase levels. Tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) decreased type I collagen and laminin levels, and abolished SPARC and ALPase syntheses. Of these peptides, bFGF and PDGF showed the greatest stimulation of [3H]thymidine incorporation into DNA. TGF-β, EGF, and TNF-α had less effect on DNA synthesis, whereas IL-1β inhibited DNA synthesis. These findings demonstrated that TGF-β, bFGF, EGF, PDGF, TNF-α, and IL-1β have characteristically different patterns of actions on DNA, laminin, type I collagen, fibronectin, ALPase, and SPARC syntheses by pulp cells. It is common for small tandem peptide multimer genes to be indirectly inserted into expression vectors and fused with a protein tag. In this study, a multimer of the tandem angiotensin I-converting enzyme inhibitory peptide (ACE-IP) gene was directly transferred to a commercially available vector and the designed gene was expressed as a repeated peptide in Escherichia coli BL21(DE3)pLysS. The process further developed in our study was the construction of six-repeated ACE-IP synthetic genes and their direct insertion. Protein expression in inclusion bodies was confirmed by SDS-PAGE and Western blot. Acid hydrolysis of inclusion bodies produced single-unit peptides through cleavage of the aspartyl-prolyl bonds. This cleaved recombinant peptide (rACE-IP) was purified using immuno-affinity chromatography followed by reversed phase-HPLC. 105–115 mg of the lyophilized recombinant peptide was obtained from 1 L E. coli culture. In vitro biological activity of rACE-IP was indistinguishable from that of the natural peptide produced by hydrolysis in artificial gastric juice or by acidic hydrolysis. The rACE-IP prepared by recombinant DNA technology and solid-phase synthesis methods showed a similar IC50. This strategy could be used for the expression of important peptides, which have N-terminal proline (P) and C-terminal aspartic acid residues (D) for commercial applications, e.g. functional foods and drinks.  相似文献

Animals associative learning plays a crucial role in many intraspecific or interspecific interactions, involving an animals use of information on its interacting counterparts. Here, we present a theoretical model that captures the basic features of an animals associative learning, which may involve generalization, for a simplest case of warning signals. Specifically, we derive formulae for the average level of associative memory as functions of a few parameters that reflect the population density of prey, predators efficiency of prey detection, and the properties of predators associative learning, including generalization and memory decay. This average level of associative memory is of central importance in determining preys fitness and, thus, the evolution of warning signals (i.e. aposematism). The derived formula also shows that another species with similar signal enhances the fitness of an aposematic species of concern as long as their signal is similar enough for generalization to occur. The model developed here can be extended to more complicated cases and the basic idea can be applied to modeling other interactions involving associative learning with generalization.  相似文献

Many murine tumor cells express not only individual haplotype-matched class I MHC molecules, but also species-specific CD1d molecules. The former class I MHC molecules generally present internally synthesized tumor-derived peptide antigens to highly specific CD8+ cytotoxic T lymphocytes (CTLs) in acquired immunity. In contrast, the latter CD1d molecules may present tumor-associated glycolipid antigens to broadly crossreactive natural killer T (NKT) cells, which might correlate with controlling tumor metastasis. Here, we showed that murine hepatoma cell line Hepa1-6-derived acid-eluted substances might contain both Db class I MHC-restricted antigens and CD1d-restriced substances, which could sensitize not only syngeneic bone marrow-derived DCs (BM-DCs), but also allogeneic BM-DCs expressing haplotype-mismatched class I MHC and species-specific CD1d molecules. To our surprise, intravenous (i.v.) immunization of C57BL/6 mice with the former syngeneic BM-DCs carrying acid-eluted materials primed both CD4CD8 and CD8+ NKT cells in the spleen, whereas immunization with the latter allogeneic BM-DCs loaded the tumor-derived substances primed CD4CD8, but not CD8+ NKT cells. A mixture of steamed soybean and boiled rice with seeded Aspergillus oryzae was naturally fermented without addition of yeasts or Lactobacilli, and kept matured for 12 months at room temperature. Chemical analysis of this rice-koji miso sample for lipid changes during maturation showed that triacylglycerol was gradually decomposed into free fatty acid, with distinct formation of fatty acid ethyl ester which, six months after the start of fermentation, came to account for 35.0% of total lipid. The ester was constituted primarily with linoleic acid (ca. 50%) and oleic acid (ca. 20%), no appreciable change in this proportion being observed during maturation. Also, the proportion was unique in that this did not reflect the fatty acid composition in a mixture of the two materials. It is possible to monitor the maturation of the rice-koji miso by following up the increase with time in fatty acid ethyl ester.  相似文献
