Cancer cells differ from normal cells in many aspects, including loss of differentiation and uninhibited cell proliferation. Recent studies have suggested that gut-enriched Krüppel-like factor (GKLF) played an important role in the regulation of cell growth in the colon. Studies from this laboratory have shown that GKLF protein predominantly expressed in the cytoplasm but not the nucleus of colon cancer cells, suggesting that impaired nuclear translocation of GKLF might contribute to cancer formation. In this report, a region containing putative nuclear localization signal (NLS) of GKLF (PKRGRR; amino acids 385-390) was investigated. Mutation of KR to WT had no effect on the inhibitory properties of GKLF on cyclin D1 promoter activity and [(3)H]thymidine uptake in HT-29 cells, whereas mutation of RR to GL abolished GKLF function completely. Additional mutation analyses demonstrated that Arg(390) is the most critical moiety within this region that mediated GKLF function and its nucleus localization. Cotransfection of Arg(390) mutant (RR/RS) completely inhibited wild-type GKLF function, and GFP-RR/RS GKLF fusion proteins failed to translocate to the nucleus. The results from this study demonstrate that Arg(390) confers the NLS of GKLF and that the nucleus-localization-deficient mutant serves as dominant-negative inhibitor of GKLF function. 相似文献

Background
Surveying survivors from a large fire provides an opportunity to explore the impact of emotional trauma on psychological outcomes.Methods
This is a cross-sectional survey of survivors of The Station Fire. Primary outcomes were post-traumatic stress (Impact of Event Scale – Revised) and depressive (Beck Depression Inventory) symptoms. Linear regression was used to examine differences in symptom profiles between those with and without physical injuries. The free-response section of the survey was analyzed qualitatively to compare psychological sequelae of survivors with and without physical injuries.Results
104 participants completed the study survey; 47% experienced a burn injury. There was a 42% to 72% response rate range. The mean age of respondents was 32 years, 62% were male, and 47% experienced a physical injury. No significant relationships were found between physical injury and depressive or post-traumatic stress symptom profiles. In the qualitative analysis, the emotional trauma that survivors experienced was a major, common theme regardless of physical injury. Survivors without physical injuries were more likely to experience survivor guilt, helplessness, self-blame, and bitterness. Despite the post-fire challenges described, most survivors wrote about themes of recovery and renewal.Conclusions
All survivors of this large fire experienced significant psychological sequelae. These findings reinforce the importance of mental health care for all survivors and suggest a need to understand factors influencing positive outcomes. 相似文献74.
The effects of selective ibotenate lesions of the complete hippocampus (CHip), the hippocampal ventral pole (VP), or the medial prefrontal cortex (mPFC) in male rats were assessed on several measures related to energy regulation (i.e., body weight gain, food intake, body adiposity, metabolic activity, general behavioral activity, conditioned appetitive responding). The testing conditions were designed to minimize the nonspecific debilitating effects of these surgeries on intake and body weight. Rats with CHip and VP lesions exhibited significantly greater weight gain and food intake compared with controls. Furthermore, CHip-lesioned rats, but not rats with VP lesions, showed elevated metabolic activity, general activity in the dark phase of the light-dark cycle, and greater conditioned appetitive behavior, compared with control rats without these brain lesions. In contrast, rats with mPFC lesions were not different from controls on any of these measures. These results indicate that hippocampal damage interferes with energy and body weight regulation, perhaps by disrupting higher-order learning and memory processes that contribute to the control of appetitive and consummatory behavior. 相似文献

Heat shock may enhance the thermotolerance of, or cause detrimental effects on, a variety of cell types or organisms, depending on the duration and intensity of the thermal challenge. Experiments were designed to investigate the effect of heat shock on the developmental competence and cytoskeletal structures of bovine oocytes following IVF. In Experiment 1, bovine cumulus-oocyte complexes (COCs) were subjected to standard IVM culture conditions for 20 h and were then randomly allocated to groups for heat shock at 42 degrees C for 0 (control), 1, 2, or 4h. The oocytes were fertilized after heat shock and followed by culture in KSOM for 8d. There were no significant differences in cleavage rates, but blastocyst formation (27% versus 44%) and total cell number per blastocyst (82+/-21 versus 108+/-36; mean+/-S.D.) were lower in the 4-h heat shock group compared to the control (P<0.05). Trophectoderm, but not ICM, cell numbers were decreased (P<0.05) in the 4-h heat shock group compared to the control. Alterations in the meiotic spindle of IVM oocytes (n=120-126) were examined after 1 to 4-h of heat shock in Experiments 2 and 3. The metaphase spindle became elongated or aberrant and smaller following heat shock, compared to the non-heat shock oocytes (P<0.05). The basis for changes in spindle configuration and the differential decrease in trophectoderm cell numbers after heat shock are not clear, but may lead to reduced embryonic development and perhaps the low pregnancy rate of domestic animals during hot seasons. 相似文献

It has been proposed that memory for odors does not have a short-term (or
working) memory system. The distinction between short- and long- term
memory in other sensory modalities has been generally supported by three
main lines of evidence: capacity differences between the proposed systems,
evidence of differential coding, and differential memory losses in
neuropsychological patients. The present paper examines these issues in an
effort to establish a similar distinction for the memory of olfactory
stimuli. Each of these lines of evidence is examined in relation to the
literature on olfactory memory. Based on this examination, it seems that
there is at least preliminary support from each of these lines of evidence
to advocate a distinction between a long- and short-term memory for
olfactory stimuli. Emphasis is placed upon the qualitative similarity of
olfactory memory to other memory systems. This similarity is further
highlighted through an examination of the literature pertinent to serial
position effects in memory for olfactory stimuli.
