The role of cis-abscisic acid (ABA) and gibberellins (GAs) in the induction of cell-cycle activities has been studied during imbibition and subsequent germination of tomato seeds. Using flow cytometry, nuclear replication activity was investigated in embryo root tips isolated from seeds of the ABA-deficient mutant sit w , the GA-deficient mutant gib-1, and the wild-type (MM) tomato (Lycopersicon esculentum Mill. cv. Moneymaker) upon imbibition in water, 10 μM GA4+7, 5 μM ABA or 5 μM ABA+10 μM GA4+7. The nuclei of fully matured dry MM, sit w and gib-1 seeds predominantly showed 2C DNA signals, indicating that the cell-cycle activity of most root-tip cells had been arrested at the G1 phase of nuclear division. However, ABA-deficient sit w seeds contained a significantly higher amount of G2 cells (4C DNA) compared with the other genotypes, suggesting that, during maturation, cell-cycle activity in sit w seeds is less efficiently arrested in G1. Upon imbibition in water, an induction of the 4C signal, indicating nuclear replication, was observed in the root tip cells of both MM and sit w embroys. The augmentation in the 4C signal occurred before visible germination. Gib-1 seeds did not show cell-cycle activity and did not germinate in water. Upon imbibition in GA4+7, both cell-cycle activity and subsequent germination were enhanced in MM and sit w seeds, and were induced in gib-1. In ABA, the germination of MM and sit w seeds was inhibited while nuclear replication of these seeds was not affected. It is concluded that GA influences germination by acting upon processes that precede cell-cycle activation, while ABA affects growth by acting upon processes that follow cell-cycle activation. 相似文献

A comparative genetic analysis of esterase (E.C.3.1.1.1) isozymes of wheat cultivar Chinese Spring in endosperm, embryo, coleoptile, leaf and root tissues revealed eight sets of isozymes characterised by different tissue specificities, pI ranges and the chromosomal locations of their controlling genes. This data was considered together with previously published work, resulting in a proposed rationalization of nine sets of wheat esterase isozymes. Although this classification included two sets of isozymes controlled by genes on the short arms of homoeologous group 3 chromosomes and three sets on the long arms of the same chromosomes, for which no recombination evidence of genetic distinctness has been obtained among either group, it is argued that the different characteristics of the various sets warrant retention of separate set nomenclatures. The capsid protein of duck hepatitis B virus (DHBV) is phosphorylated at multiple sites during viral infection. A cluster of sites is located near the C terminus of the 262-amino-acid protein. We have used site-directed mutagenesis to show that three serines and one threonine serve as phosphate acceptor amino acids in the C terminus. An additional six potential phosphate acceptor sites in this region were apparently not utilized. Each serine or threonine that served as a phosphate acceptor was adjacent to a downstream proline, while all six serines that were not acceptors for phosphate residues lacked adjacent downstream prolines. Mutation of the downstream proline to glycine at each site had the same effect as mutating the serine itself, suggesting an SP or TP motif as an essential feature for capsid protein phosphorylation. Phosphorylation at these four sites resulted in complex shifts in electrophoretic mobility in sodium dodecyl sulfate gels of the capsid protein or of a C-terminal peptide containing the phosphorylated sites, suggesting that specific conformations of the C terminus are associated with different combinations of phosphorylated serines. We speculate that distinct functions of the C terminus may be associated with different phosphorylated domains on the intact capsid. 相似文献

Abstract: In a previous study, protein kinase FA /glycogen synthase kinase-3 ( FA /GSK-3 ) was identified as a myelin basic protein (MBP) kinase associated with intact brain myelin. In this report, the phosphorylation sites of MBP by kinase FA /GSk-3 were further determined by two-dimensional electrophoresis/TLC, phosphoamino acid analysis, tryptic peptide mapping, Edman degradation, and direct sequencing. Kinase FA /GSK-3 phosphorylates MBP on both threonine and serine residues. Three tryptic phosphopeptide peaks were resolved by C18 reverse-phase HPLC. Sequential manual Edman degradation together with direct sequence analysis revealed that T(p)PPPSQGK is the phosphorylation site sequence for the first major phosphopeptide peak. When mapping with the bovine brain MBP sequence, we finally demonstrate Thr97 -Pro, one of the in vivo phosphorylation sites in MBP, as the major site phosphorylated by kinase FA /GSK-3, implicating a physiologically relevant role of FA /GSK-3 in the regulation of brain myelin function. By using the same approach, we also identified NIVT94 (p)PR as the phosphorylation site sequence in the second major tryptic phosphopeptide derived from [32 P]MBP phosphorylated by kinase FA /GSK-3, further indicating that kinase FA /GSK-3 represents a Thr-Pro motif-directed MBP kinase involved in the phosphorylation of brain myelin. 相似文献

In rice (Oryza sativa L.), cytosolic triosephosphate isomerase (TPI) is encoded by a single gene. TPI catalyzes a vital step in glycolysis, and RNA blots showed that the tpi gene is expressed in all vegetative tissues (root, culm, and leaves) and in rice suspension cells. No effect of light on expression was detected, but submergence of rice seedlings resulted in elevated levels of TPI mRNA in roots and culms. The 2767-bp 5[prime] upstream sequence of the tpi gene was fused translationally with the [beta]-glucuronidase (gusA) gene, and the resulting construct, TPI-GUS, was found to express constitutive, high levels of GUS activity in transgenic tobacco (Nicotiana tabacum) plants. However, the same construct yielded no GUS activity in stably transformed rice plants, and RNA blots showed that no GUS mRNA could be detected even though stable integration of functional copies of the construct was confirmed by Southern blot and genomic polymerase chain reaction analyses. Transient assays using particle bombardment yielded high levels of GUS expression from the TPI-GUS construct in tobacco leaves, but essentially no expression in rice, barley, or maize leaves. When the first intron of the tpi gene was included in the construct (TPI-int1-GUS), transient GUS activity was routinely obtained in rice leaves, revealing that the first intron of the rice tpi gene is crucial for its expression in rice. TPI-int1-GUS also directed transient GUS expression in maize and barley leaves, but little or no activity was obtained from this construct in tobacco, tomato, or soybean leaves. Summary Free ammonia inhibition to Nitrobacter population in a nitrifying biofilm was investigated. It was found that when the tree ammonia concentration is greater than 0.1 mgN/l the oxidative activity of Nitrobacter was significantly inhibited, and resulting in a transient accumulation of nitrite ions. The results show that Nitrobacter population is capable of rapidly recovering its lost metabolic activity once the free ammonia concentration becomes less than 0.1 mgN/l, and a complete oxidation of nitrite to nitrate was achieved. 相似文献

The effects of shock loading on the performance of a nitrifying biofilm reactor were investigated over a wide range of conditions by varying the feed concentration of ammonium (SO) and dilution rate (D) respectively. It was found that variation in the effluent ammonium concentration as SO or D changes was subject to a semi-U shaped curve. The response patterns of the nitrifying biofilm reactor to one-step change in SO at a constant dilution rate, and to one-step change in D with keeping constant SO have no significant difference. 相似文献
