ATR (ATM and Rad3-related) initiates a DNA damage signaling pathway in human cells upon DNA damage induced by UV and UV-mimetic agents and in response to inhibition of DNA replication. Genetic data with human cells and in vitro data with Xenopus egg extracts have led to the conclusion that the kinase activity of ATR toward the signal-transducing kinase Chk1 depends on the mediator protein Claspin. Here we have reconstituted a Claspin-mediated checkpoint system with purified human proteins. We find that the ATR-dependent phosphorylation of Chk1, but not p53, is strongly stimulated by Claspin. Similarly, DNA containing bulky base adducts stimulates ATR kinase activity, and Claspin acts synergistically with damaged DNA to increase phosphorylation of Chk1 by ATR. Mutations in putative phosphorylation sites in the Chk1-binding domain of Claspin abolish its ability to mediate ATR phosphorylation of Chk1. We also find that a fragment of Claspin containing the Chk1-binding domain together with a domain conserved in the yeast Mrc1 orthologs of Claspin is sufficient for its mediator activity. A highly stable phosphonate‐functionalized anthraquinone is introduced as the redox‐active material in a negative potential electrolyte (negolyte) for aqueous redox flow batteries operating at nearly neutral pH. The design and synthesis of 2,6‐DPPEAQ, (((9,10‐dioxo‐9,10‐dihydroanthracene‐2,6‐diyl)bis(oxy))bis(propane‐3,1‐diyl))bis(phosphonic acid), which has a high solubility at pH 9 and above, is described. Chemical stability studies demonstrate high stability at both pH 9 and 12. By pairing 2,6‐DPPEAQ with a potassium ferri/ferrocyanide positive electrolyte across an inexpensive, nonfluorinated permselective polymer membrane, this near‐neutral quinone flow battery exhibits an open‐circuit voltage of 1.0 V and a capacity fade rate of 0.00036% per cycle and 0.014% per day, which is the lowest ever reported for any flow battery in the absence of rebalancing processes. It is further demonstrated that the negolyte pH drifts upward upon atmospheric oxygen penetration but, when oxygen is excluded, oscillates reversibly between 9 and 12 during cycling. These results enhance the suitability of aqueous‐soluble redox‐active organics for use in large‐scale energy storage, potentially enabling massive penetration of intermittent renewable electricity. 相似文献

Summary
Drosophila melanogaster males were irradiated at various stages of pre-imaginal development, and mutation rates were compared between first broods of different age groups, and between successive broods in the same age group. The findings were correlated with observations on germ cell development in the larval and pupal testis. Sensitivity to the mutagenic effect of X-rays is low in all premeiotic stages. It increases suddenly and dramatically with the onset of meiosis, remains stationary through meiosis, and subsequently increases again up to a peak in the late spermatid stage when transformation into morphologically mature spermatozoa is taking place. This stage provides sperm for the first brood from males treated at about the middle of the pupal period, and mutation rate for a given dose of X-rays is here about 4 times as high as in mature sperm and about 1 1/2 times as high as during meiosis. Subsequently, mutation rate drops again gradually to the level characteristic for mature sperm. Germinal selection cannot account for any of these changes; but it may be the main cause for the slight increase in response which occurs during spermatogonial development.With 10 figures in the text. 相似文献

Total Petroleum Hydrocarbons (TPH) are one of the most common groups of persistent environmental toxic organic contaminants to many organisms as well as to humans. In the present work, oil-polluted soil samples were phyto-remediated and analyzed. The investigated soil samples were collected from a location close to the oil petroleum production site in Ras-Gharib, Red Sea, Egypt. The phytoremediation process through TPH reduction and/or removal was carried out using Helianthus annuus (sunflower plant) based on its efficiency as a phytoremediator for organic pollutants. A preliminary four-week scheme of Helianthus annuus remediation, supported by twice quantized fertilization, provided a result of ~56% clean soil. Contaminated and phyto-remediated samples were diagnosed and analyzed through particle size distribution, Carbon-Hydrogen-Sulfur-Nitrogen elemental analysis (CHSN), Organic Matter content (OM%), Total Petroleum Hydrocarbons determination (%) and spectroscopically through Laser Induced Breakdown Spectroscopy (LIBS) and Laser Induced Fluorescence (LIF). Promising results have been achieved indicating the feasibility of planting sunflower for effective TPH remediation of the polluted soil and also the possibility of in situ monitoring of the remediation with easy, cost effective and fast spectrochemical analytical techniques, namely LIBS and LIF. 相似文献

Multi-stage cascade impactors (CIs) are the preferred measurement technique for characterizing the aerodynamic particle size
distribution of an inhalable aerosol. Stage mensuration is the recommended pharmacopeial method for monitoring CI “fitness
for purpose” within a GxP environment. The Impactor Sub-Team of the European Pharmaceutical Aerosol Group has undertaken an
inter-laboratory study to assess both the precision and accuracy of a range of makes and models of instruments currently used
for optical inspection of impactor stages. Measurement of two Andersen 8-stage ‘non-viable’ cascade impactor “reference” stages
that were representative of jet sizes for this instrument type (stages 2 and 7) confirmed that all instruments evaluated were
capable of reproducible jet measurement, with the overall capability being within the current pharmacopeial stage specifications
for both stages. In the assessment of absolute accuracy, small, but consistent differences (ca. 0.6% of the certified value)
observed between ‘dots’ and ‘spots’ of a calibrated chromium-plated reticule were observed, most likely the result of treatment
of partially lit pixels along the circumference of this calibration standard. Measurements of three certified ring gauges,
the smallest having a nominal diameter of 1.0 mm, were consistent with the observation where treatment of partially illuminated
pixels at the periphery of the projected image can result in undersizing. However, the bias was less than 1% of the certified
diameter. The optical inspection instruments evaluated are fully capable of confirming cascade impactor suitability in accordance

The effects of Depoprovera and Deladroxone were studied in humans, on certain milk components as well as on the growth of the nursed infants. Both drugs caused reduction in milk yield. Both drugs caused an increase in the concentration of milk total proteins, however. Depoprovera caused an increase while Deladroxone caused a decrease in the total amount of milk proteins per feed. Depoprovera showed no effect while Deladroxone caused an increase in the concentration of milk lipids; however, both drugs caused reduction in the total amount of milk lipids per feed. Both drugs showed no effect on the concentration of milk lactose, but caused reduction in the total amount of milk lactose per feed. The percentage increase in weight of nursed infants was decreased by Depoprovera, but not affected by Deladroxone. 相似文献
