The physicochemical conditions and microbiological characteristics of the formation waters of the Kongdian bed of the Dagang oil field (China) were studied. It was demonstrated that this bed is a high-temperature ecosystem with formation waters characterized by low mineralization. The concentrations of nitrogen and phosphorus compounds, as well as of electron acceptors, are low. Oil and oil gas are the main organic matter sources. The bed is exploited with water-flooding. The oil stratum was inhabited mostly by anaerobic thermophilic microorganisms, including fermentative (10(2)-10(5) cells/ml), sulfate-reducing (0-10(2) cells/ml), and methanogenic (0-10(3) cells/ml) microorganisms. Aerobic bacteria were detected mainly in the near-bottom zone of injection wells. The rate of sulfate reduction varied from 0.002 to 18.940 microg S(2-) l(-1) day(-1) and the rate of methanogenesis from 0.012 to 16.235 microg CH4 l(-1) day(-1). Microorganisms with great biotechnological potential inhabited the bed. Aerobic thermophilic bacteria were capable of oxidizing oil with the formation of biomass, the products of partial oxidation of oil (volatile acids), and surfactants. During growth on the culture liquid of oiloxidizing bacteria, methanogenic communities produced methane and carbon dioxide, which also had oil-releasing capabilities. Using various labeled tracers, the primary filtration flows of injected solutions at the testing site were studied. Our comprehensive investigations allowed us to conclude that the tested method for microbial enhancement of oil recovery based on the activation of the stratal microflora can be applied in the Kongdian bed horizons. 相似文献

Spontaneous mutations in Drosophila melanogaster are related mainly to transposable elements (TEs). They are caused by both migration of TEs over the genome (transpositions) and the ability of TEs to induce chromosomal mutations. Migration of DNA transposons is accompanied by formation of double-strand DNA breaks (DSBs), which are repaired by host repair systems encoded by genes for recombination repair. We relied on this notion to develop a combined approach to the investigation of the type of DNA breaks accompanying transpositions; investigation of systems involved in DSB repair; and detection of repair genes, whose products were involved in repair of DNA breaks induced by TE transposition. The approach is based on the combination of experimental insertional mutagenesis systems and genetic environment deficient for enzymes of the repair system in a single genome. Selected 7-alkylidene substituted cephems were synthesized and subjected to antitumor assay. The effect of substituents was examined to establish structure-activity relationships. Interaction of insectoacaricide Me (EtO)P(S)SCH2SCH2COOMe (I), its activation metabolites (P = O (II), S = O, and P = O, S = O (III) analogues), and a detoxication product (-COOH analoque (IV) with rat liver carboxylesterase, acetylcholinesterase and butyrylcholinesterase of warm-blooded animals, as well as with cholinesterase and carboxylesterase of American cockroach has been studied. Low toxicity of (I) towards warm-blooded animals and American cockroach is shown to result from its rapid hydrolysis with corresponding carboxylesterases to form (IV). Monothiophosphonates (II) and (III) are not hydrolyzed by carboxylesterases but inhibit them irreversibly. Derivatization of the natural flavonoid dihydroquercetin with p-aminobenzoic acid was carried out in an ethyl acetate/citric buffer biphasic system using laccase from the fungus Trametes hirsuta. The main reaction product yield was ~68 mol %. The product was characterized by 1H NMR, 13C NMR, and liquid chromatography-mass spectroscopy, and its structure was elucidated. The reaction product affected viability of cultured human rhabdomyosarcoma cells (RD cell line) in a dose-dependent manner and, therefore, can be of interest to pharmaceutical industry. 相似文献

In leukocytes of exudate from diabetic rabbits, the activities of hexokinase, phosphoglucomutase and glucose-6-phosphate dehydrogenase are increased, and a tendency of adenylate kinase activity to decline is observable. The activities of UDP-pyrophosphatase, UDP-glycogentransferase, 6-phosphogluconate dehydrogenase and glutahione reductase in the exudate erythrocytes in diabetes are not essentially altered. The decrease of the key enzymes of glycolysis and pentose phosphate cycle, providing the leukocytes with energy and metabolites, reduces the functional activity of leukocytes from exudate in diabetes. 相似文献
