Benjamin Franklin had at least two accidents that resulted in electricity passing through his brain. In addition, he witnessed a patient's similar accident and performed an experiment that showed how humans could endure shocks to the head without serious ill effects, other than amnesia. Jan Ingenhousz, Franklin's Dutch-born medical correspondent better known for his discovery of photosynthesis, also had a serious accident that sent electricity though his head and, in a letter to Franklin, he described how he felt unusually elated the next day. During the 1780s, Franklin and Ingenhousz encouraged leading French and English electrical "operators" to try shocking the heads of melancholic and other deranged patients in their wards. Although they did not state that they were responding to Ingenhousz's and Franklin's suggestions, Birch, Aldini, and Gale soon did precisely what Ingenhousz and Franklin had suggested. Over the last one hundred years, compartmentalization and water management activities have reduced water flow to the ridge
and slough landscape of the Everglades. As a result, the once corrugated landscape has become topographically and vegetationally
uniform. The focus of this study was to quantify variation in surface flow in the ridge and slough landscape and to relate
flow conditions to particulate transport and deposition. Over the 2002–2003 and 2003–2004 wet seasons, surface velocities
and particulate accumulation were measured in upper Shark River Slough in Everglades National Park. Landscape characteristics
such as elevation, plant density and biomass also were examined to determine their impact on flow characteristics and material
transport. The results of this study demonstrate that the release of water during the wet season not only increases water
levels, but also increased flow speeds and particulate transport and availability. Further, flow speeds were positively and
significantly correlated with water level thereby enhancing particulate transport in sloughs relative to ridges especially
during peak flow periods. Our results also indicate that the distribution of biomass in the water column, including floating
plants and periphyton, affects velocity magnitude and shape of vertical profiles, especially in the sloughs where Utricularia spp. and periphyton mats are more abundant. Plot clearing experiments suggest that the presence of surface periphyton and
Utricularia exert greater control over flow characteristics than the identity (i.e., sawgrass or spike rush) or density of emergent macrophytes,
two parameters frequently incorporated into models describing flow through vegetated canopies. Based on these results, we
suggest that future modeling efforts must take the presence of floating biomass, such as Utricularia, and presence of periphyton into consideration when describing particulate transport. 相似文献

Activation of protein 4.1R exon 16 (E16) inclusion during erythropoiesis represents a physiologically important splicing switch that increases 4.1R affinity for spectrin and actin. Previous studies showed that negative regulation of E16 splicing is mediated by the binding of heterogeneous nuclear ribonucleoprotein (hnRNP) A/B proteins to silencer elements in the exon and that down-regulation of hnRNP A/B proteins in erythroblasts leads to activation of E16 inclusion. This article demonstrates that positive regulation of E16 splicing can be mediated by Fox-2 or Fox-1, two closely related splicing factors that possess identical RNA recognition motifs. SELEX experiments with human Fox-1 revealed highly selective binding to the hexamer UGCAUG. Both Fox-1 and Fox-2 were able to bind the conserved UGCAUG elements in the proximal intron downstream of E16, and both could activate E16 splicing in HeLa cell co-transfection assays in a UGCAUG-dependent manner. Conversely, knockdown of Fox-2 expression, achieved with two different siRNA sequences resulted in decreased E16 splicing. Moreover, immunoblot experiments demonstrate mouse erythroblasts express Fox-2. These findings suggest that Fox-2 is a physiological activator of E16 splicing in differentiating erythroid cells in vivo. Recent experiments show that UGCAUG is present in the proximal intron sequence of many tissue-specific alternative exons, and we propose that the Fox family of splicing enhancers plays an important role in alternative splicing switches during differentiation in metazoan organisms. 相似文献

Extensive portions of the southern Everglades are characterized by series of elongated, raised peat ridges and tree islands
oriented parallel to the predominant flow direction, separated by intervening sloughs. Tall herbs or woody species are associated
with higher elevations and shorter emergent or floating species are associated with lower elevations. The organic soils in
this “Ridge-and-Slough” landscape have been stable over millennia in many locations, but degrade over decades under altered
hydrologic conditions. We examined soil, pore water, and leaf phosphorus (P) and nitrogen (N) distributions in six Ridge and
Slough communities in Shark Slough, Everglades National Park. We found P enrichment to increase and N to decrease monotonically
along a gradient from the most persistently flooded sloughs to rarely flooded ridge environments, with the most dramatic change
associated with the transition from marsh to forest. Leaf N:P ratios indicated that the marsh communities were strongly P-limited,
while data from several forest types suggested either N-limitation or co-limitation by N and P. Ground water stage in forests
exhibited a daytime decrease and partial nighttime recovery during periods of surface exposure. The recovery phase suggested
re-supply from adjacent flooded marshes or the underlying aquifer, and a strong hydrologic connection between ridge and slough.
We therefore developed a simple steady-state model to explore a mechanism by which a phosphorus conveyor belt driven by both
evapotranspiration and the regional flow gradient can contribute to the characteristic Ridge and Slough pattern. The model
demonstrated that evapotranspiration sinks at higher elevations can draw in low concentration marsh waters, raising local
soil and water P concentrations. Focusing of flow and nutrients at the evapotranspiration zone is not strong enough to overcome
the regional gradient entirely, allowing the nutrient to spread downstream and creating an elongated concentration plume in
the direction of flow. Our analyses suggest that autogenic processes involving the effects of initially small differences
in topography, via their interactions with hydrology and nutrient availability, can produce persistent physiographic patterns
in the organic sediments of the Everglades. 相似文献

Active lymph transport relies on smooth muscle cell (SMC) contractions around collecting lymphatic vessels, yet regulation of lymphatic vessel wall assembly and lymphatic pumping are poorly understood. Here, we identify Reelin, an extracellular matrix glycoprotein previously implicated in central nervous system development, as an important regulator of lymphatic vascular development. Reelin-deficient mice showed abnormal collecting lymphatic vessels, characterized by a reduced number of SMCs, abnormal expression of lymphatic capillary marker lymphatic vessel endothelial hyaluronan receptor 1 (LYVE-1), and impaired function. Furthermore, we show that SMC recruitment to lymphatic vessels stimulated release and proteolytic processing of endothelium-derived Reelin. Lymphatic endothelial cells in turn responded to Reelin by up-regulating monocyte chemotactic protein 1 (MCP1) expression, which suggests an autocrine mechanism for Reelin-mediated control of endothelial factor expression upstream of SMC recruitment. These results uncover a mechanism by which Reelin signaling is activated by communication between the two cell types of the collecting lymphatic vessels--smooth muscle and endothelial cells--and highlight a hitherto unrecognized and important function for SMCs in lymphatic vessel morphogenesis and function. 相似文献

The lateral transmembrane protein-protein interaction has been regarded as "undruggable" despite its importance in many biological processes. The homo-trimerization of transmembrane domain 5 (TMD-5) of latent membrane protein 1 (LMP-1) is critical for the constitutive oncogenic activation of the Epstein-Barr virus (EBV). Herein, we report a small molecule agent, NSC 259242 (compound 1), to be a TMD-5 self-association disruptor. Both the positively charged acetimidamide functional groups and the stilbene backbone of compound 1 are essential for its inhibitory activity. Furthermore, cell-based assays revealed that compound 1 inhibits full-length LMP-1 signaling in EBV infected B cells. These studies demonstrated a new strategy for identifying small molecule disruptors for investigating transmembrane protein-protein interactions. 相似文献

Alzheimer's disease, the most common cause of dementia in the elderly and characterized by the deposition and accumulation of plaques, is composed in part of β-amyloid (Aβ) peptides, loss of neurons, and the accumulation of neurofibrillary tangles. Here, we describe ponezumab, a humanized monoclonal antibody, and show how it binds specifically to the carboxyl (C)-terminus of Aβ40. Ponezumab can label Aβ that is deposited in brain parenchyma found in sections from Alzheimer's disease casualties and in transgenic mouse models that overexpress Aβ. Importantly, ponezumab does not label full-length, non-cleaved amyloid precursor protein on the cell surface. The C-terminal epitope of the soluble Aβ present in the circulation appears to be available for ponezumab binding because systemic administration of ponezumab greatly elevates plasma Aβ40 levels in a dose-dependent fashion after administration to a mouse model that overexpress human Aβ. Administration of ponezumab to transgenic mice also led to a dose-dependent reduction in hippocampal amyloid load. To further explore the nature of ponezumab binding to Aβ40, we determined the X-ray crystal structure of ponezumab in complex with Aβ40 and found that the Aβ40 carboxyl moiety makes extensive contacts with ponezumab. Furthermore, the structure-function analysis supported this critical requirement for carboxy group of AβV40 in the Aβ-ponezumab interaction. These findings provide novel structural insights into the in vivo conformation of the C-terminus of Aβ40 and the brain Aβ-lowering efficacy that we observed following administration of ponezumab in transgenic mouse models. 相似文献

The maize (Zea mays) kernel plays a critical role in feeding humans and livestock around the world and in a wide array of industrial applications. An understanding of the regulation of kernel starch, protein, and oil is needed in order to manipulate composition to meet future needs. We conducted joint-linkage quantitative trait locus mapping and genome-wide association studies (GWAS) for kernel starch, protein, and oil in the maize nested association mapping population, composed of 25 recombinant inbred line families derived from diverse inbred lines. Joint-linkage mapping revealed that the genetic architecture of kernel composition traits is controlled by 21-26 quantitative trait loci. Numerous GWAS associations were detected, including several oil and starch associations in acyl-CoA:diacylglycerol acyltransferase1-2, a gene that regulates oil composition and quantity. Results from nested association mapping were verified in a 282 inbred association panel using both GWAS and candidate gene association approaches. We identified many beneficial alleles that will be useful for improving kernel starch, protein, and oil content. 相似文献

Recent studies suggest a possible link between post-traumatic stress disorder (PTSD) and obesity risk, which would have implications for the development of obesity-related diseases in this population. The present study examined the association between PTSD and obesity and whether this association differed by sex in a representative sample of the US population. A secondary objective was to determine whether the association between PTSD and obesity was mediated by binge eating disorder (BED). Data were from the Collaborative Psychiatric Epidemiology Surveys (CPES), which comprises three nationally representative cross-sectional surveys that were conducted between 2001 and 2003. Logistic regression analyses weighted to represent the general US adult population were performed. In the total sample of 20,013 participants, rates of obesity were 24.1% for persons without a lifetime history of PTSD and 32.6% among persons with PTSD in the past year. Adjusting for socio-demographic characteristics, depression, substance and alcohol abuse/dependence, and psychotropic medication status, past year PTSD was associated with greater likelihood of obesity (odds ratio (OR) = 1.51; 95% confidence interval (CI) = 1.18, 1.95), with no differences by gender. BED did not statistically mediate the relationship between PTSD and obesity. The present study provides support for a link between PTSD and obesity. Findings further existing literature by indicating that the association is consistent across sexes and is not statistically mediated by BED. 相似文献
