Peri-urban water bodies are at risk from excessive pollution as they are direct sinks for urban effluents. The occurrence of oestrogenic and androgenic endocrine disrupting chemicals (EDCs) in effluents and water bodies around the city of Bulawayo, Zimbabwe, and their effects on wild fish was investigated in 2013. Effluent and water were sampled from sewage treatment plants, an urban stream, and effluent-polluted dams, and were compared with a ‘pristine’ dam upstream of Bulawayo. Organic pollutants were extracted by solid-phase extraction and tested for EDCs using a yeast oestrogen/androgen screen. Oestrogenic and androgenic potencies were expressed as 17β-oestradiol equivalent (EEq) or dihydrotestosterone equivalent (TEq). Tilapia and catfish from the dams were analysed for gonado-somatic indices and testis histopathology. Effluents from STPs, which directly flow into Umguza Dam, had EEq of 33 ng l?1 and 55 ng l?1, respectively. Umguza Dam, Khami Dam and Matsheumhlope Stream had EEqs of 237 ng l–1, 9 ng l?1 and 2 ng l?1, respectively. Androgenic activity was detected in only one STP (TEq = 93 ng l?1). Tilapia sampled from effluent-polluted dams had high incidences of testis-ova, but catfish had no signs of reproductive dysfunction. The major viral phosphoproteins (p12) of the Rauscher murine leukemia virus (R-MuLV) and the simian sarcoma-associated virus (SSAV) bind in vitro to their homologous 70S and 35S viral RNAs. Using purified 32P-labeled RNA and 125I-labeled p12 protein, complexes that are stabilized by formaldehyde-cross-linking can be readily detected after velocity gradient centrifugation. The in vitro reconstructed ribonucleoprotein complexes are seen only with p12 proteins incubated with viral RNAs isolated from the same type C viruses; no such complexes form with heterologous protein-RNA mixtures. Homologous but not heterologous p12 molecules compete with radiolabeled p12 protein for the specific viral RNA binding sites. The competition assay permits the detection of 10 ng of viral p12 protein. The major internal protein of type C viruses (p30) does not bind to viral RNA using identical assay conditions. From the specific activities of the radiolabeled components and also by equilibrium sedimentation analysis, we estimate that fewer than 15 molecules of p12 protein bind to each molecule of viral RNA. Both the specificity and stoichiometry of the p12-RNA interactions suggest that these RNA tumor virus proteins have a regulatory role in cells. 相似文献

Four new type C viruses were isolated from putatively virus-negative baboon lung, kidney, and testicular cells by cocultivation with several permissive host cell lines. The baboon type C viruses are infectious for cells from various mammalian species, but do not replicate in any baboon cell lines so far tested. These viruses can be distinguished from other major classes of mammalian type C viruses, including previous isolates from primates, but are most closely related to endogenous feline viruses of the RD-114/CCC group. By immunologic criteria, viral host range, and nucleic acid hybridization studies, the baboon type C viruses are highly related to one another and represent a distinct new class of endogenous primate type C viruses. 相似文献

A nylon wool-adherent, B cell-enriched population is required during the in vitro induction of third order effector suppressor T cells (Ts3). This B cell population expresses IgM and IgD and is devoid of conventional T cell markers such as Thy-1, L3T4, and Lyt-1. Treatment of the B cell population with anti-NP antibodies expressing the NPb idiotype and complement specifically eliminated the ability to generate Ts cell activity, suggesting that the critical B cells expressed anti-idiotypic receptors. To independently verify the role of anti-idiotypic B cells in the generation of Ts cells, B cells were panned on antibody-coated plates. The results demonstrated that only NPb idiotype-binding B cells could induce effector suppressor cells from naive T cell populations.
