In monocarpic plants, reproduction is closely associated with senescence, which is itself often correlated to specific environmental signals. Floerkea proserpinacoides (Limnanthaceae) is a spring ephemeral annual of the deciduous forests of eastern North America. The phenology of its growth and reproduction is considered to be a specific adaptation to the short period during which there is a high availability of resources (mostly light). Indeed, flowering starts 2-3 wk following seedling emergence soon after snowmelt and continues until tree canopy closure. However, fruit maturation is postponed for several weeks and is followed by the plant's death. The objective of this study is to determine if senescence in F. proserpinacoides is primarily cued by fruit maturation or deteriorating light conditions associated with tree canopy closure. Plants for which reproductive investment was manipulated by removing their carpels were grown either in full light or in the shade. Carpel-removal plants reached a higher biomass than control plants (46.0- 57.5% higher), especially in full light. However, longevity was greater in carpel-removal plants, particularly in the shade (25.3-37.8% greater). These results thus suggest that fruit maturation, not deteriorating light conditions associated with canopy closure, is the primary cue of plant senescence in F. proserpinacoides.  相似文献

The sensory bias model of sexual selection posits that female mating preferences are by-products of natural selection on sensory systems. Although sensory bias was proposed 20 years ago, its critical assumptions remain untested. This paradox arises because sensory bias has been used to explain two different phenomena. First, it has been used as a hypothesis about signal design, that is, that males evolve traits that stimulate female sensory systems. Second, sensory bias has been used as a hypothesis for the evolution of female preference itself, that is, to explain why females exhibit particular preferences. We focus on this second facet. First, we clarify the unique features of sensory bias relative to the alternative models by considering each in the same quantitative genetic framework. The key assumptions of sensory bias are that natural selection is the predominant evolutionary mechanism that affects preference and that sexual selection on preferences is quantitatively negligible. We describe four studies that would test these assumptions and review what we can and cannot infer about sensory bias from existing studies. We suggest that the importance of sensory bias as an explanation for the evolution of female preferences remains to be determined.  相似文献

The RD1 genomic region is present in virulent strains of Mycobacterium tuberculosis (MTB), missing from the vaccine strain M. bovis BCG, and its importance to virulence has been established experimentally. Based on in silico analysis, it has been suggested that RD1 may encode a novel secretion system, but the mechanism by which this region affects virulence is unknown. Here we examined mutants disrupted in five individual RD1 genes. Both in vitro and in vivo, each mutant displayed an attenuated phenotype very similar to a mutant missing the entire RD1 region. Genetic complementation of individual genes restored virulence. Attenuated mutants could multiply within THP-1 cells, but they were unable to spread to uninfected macrophages. We also examined export of two immunodominant RD1 proteins, CFP-10 and ESAT-6. Export of these proteins was greatly reduced or abolished in each attenuated mutant. Again, genetic complementation restored a wild-type phenotype. Our results indicate that RD1 genes work together to form a single virulence determinant, and argue that RD1 encodes a novel specialized secretion system that is required for pathogenesis of MTB.  相似文献

In endothelial cells, vascular endothelial growth factor (VEGF) induces an accumulation of stress fibers associated with new actin polymerization and rapid formation of focal adhesions at the ventral surface of the cells. This cytoskeletal reorganization results in an intense motogenic activity. Using porcine endothelial cells expressing one or the other type of the VEGF receptors, VEGFR1 or VEGFR2, or human umbilical vein endothelial cells pretreated with a VEGFR2 neutralizing antibody, we show that VEGFR2 is responsible for VEGF-induced activation of the stress-activated protein kinase-2/p38 (SAPK2/p38), phosphorylation of focal adhesion kinase (FAK), and enhanced migratory activity. Activation of SAPK2/p38 triggered actin polymerization whereas FAK, which was phosphorylated independently of SAPK2/p38, initiated assembly of focal adhesions. Both processes contributed to the formation of stress fibers. Geldanamycin, an inhibitor of HSP90 blocked tyrosine phosphorylation of FAK, assembly of focal adhesions, actin reorganization, and cell migration, all of which were reversed by overexpressing HSP90. We conclude that VEGFR2 mediates the physiological effect of VEGF on cell migration and that two independent pathways downstream of VEGFR2 regulate actin-based motility. One pathway involves SAPK2/p38 and leads to enhanced actin polymerization activity. The other involves HSP90 as a permissive signal transduction factor implicated in FAK phosphorylation and assembly of focal adhesions.  相似文献

Researchers have suggested fluctuating asymmetry (FA) as an indicator of environmental stress and have usually tested this assertion by examining relations between FA of single traits and stress. Fluctuating asymmetry stress relations are real but are typically weak and difficult to detect. Researchers would like to maximize the probability of detecting FA-stress relations when they exist. We assert that analyses based on the FA of multiple traits may provide better methods for detecting stress. In this article, we used computer simulations to compare the ability of six analyses to detect differences in FA between stressed and unstressed populations. We show that the optimal analysis depends upon the underlying form of the FA distributions. We also show that two of the analyses had inflated Type I errors in some situations. Finally, we quantify the advantage of our preferred analysis over those of single-trait FA in detecting stress.  相似文献

The growth rates of four saline-lake diatom taxa were measuredunder varying conditions of salinity (5, 8 and 11), brine type(sulfate- versus bicarbonate-dominated) and nitrogen form (NH4+versus NO3), using a full factorial design. With NO3as the nitrogen source, Cyclotella quillensis, Cymbella pusillaand Anomoeoneis costata exhibited lower growth rates in thesulfate versus bicarbonate media. The strain of Chaetoceroselmorei used in these experiments, isolated from a sulfate-dominatedlake, was unable to grow on NO3 alone. In the NH4+ treatments,neither salinity nor brine type affected the growth rates ofC.quillensis or C.elmorei. When supplied with NH4+, C.pusillaand A.costata had higher growth rates in the bicarbonate versussulfate media, although for C.pusilla the difference on NH4+was not as great as on NO3. The impact of brine typeon NO3 use is consistent with the theory that sulfateinhibits molybdate uptake, as molybdenum is required for NO3use but not NH4+. Cymbella pusilla was the only taxon affectedby changes in salinity. The four taxa used in these experimentsare frequently found in saline lakes and saline-lake sediments,hence they are used in paleoclimate reconstructions; the resultspresented here provide additional information that may enhancethese diatom-based reconstructions.  相似文献

A detailed diatom record from Lake K?lksj?n, west-central Sweden, reveals two periods of abrupt ecological change correlative with the 8.2?ka cooling event. Using a combination of abrupt step changes and piece-wise linear regressions, the diatom data were analyzed for change points over time, and two sudden and large events that are described as regime shifts were detected. During the first event at c. 8040?cal. y BP, a doubling in diatom biomass took place over 5?C10?years. This increase in primary productivity can be connected to an erosion event in the catchment that resulted in an abrupt increase in nutrient supply to the lake. The second event was characterized by a substantial shift within the planktonic diatom community from taxa indicative of colder conditions to those indicating warm over 5?C10?years at c. 7850?cal. y BP. This event was superimposed on a successive change from periphytic to planktonic diatom dominance over a 250-year period and a gradual diversification of the periphytic community that spanned c. 150?years. Rapid climate warming following the 8.2?ka event likely caused these changes and both regime shifts are examples of externally driven abrupt ecological change. This study demonstrates that it is possible to detect, quantify and test for regime shifts in paleoecological data, and it highlights the need for high sampling resolution and precise chronological control. High-resolution paleoecological reconstructions of ecological regime shifts in response to climate change can provide useful analogues of future changes in ecosystem structure and functioning.  相似文献

Pyrodinium bahamense is a dinoflagellate of concern in subtropical and tropical coastal environments. To date, there is only a single published study on its fatty acids, but no published data on its sterol composition. Sterols, which are membrane‐reinforcing lipids in eukaryotes, display a great diversity of structures in dinoflagellates, with some serving as chemotaxonomic markers. We have examined the sterol compositions of two isolates of P. bahamense from Indian River Lagoon and Tampa Bay, Florida, and have found both to produce three sterols: cholesterol, dinosterol, and 4α‐methylgorgostanol. All three sterols are found in closely related, armored taxa.  相似文献

Bacterial endosymbionts of sap-sucking insects provide their host with a num- ber of beneficial qualities,including the supply of nutrition,defense against parasitoids, and protection from heat stress.Damage to these bacterial associates can therefore have a negative impact on the fitness of their insect host.We evaluated observational and experi- mental factors regarding the normative hemlock woolly adelgid (Adelges tsugae Annand) (Hemiptera:Adelgidae)to help understand the roles of its three recently identified symbionts,including under heat stress conditions.The prevalence of A.tsugae's facultafive symbiont (Serratia symbiotica)was examined at different spatial scales to determine how variable infection rates are for this symbiont.There was no significant difference found in infection rates between adelgids on a tree,within a plot,or within a state.However, significantly more adelgids in Georgia (95%)had S.symbiotica compared to those in New York (68%).Microsatellite genotyping of the adelgids found that this difference was most likely not the result of a second introduction ofA.tsugae into eastern North America.Comparison orS.symbiotica proportions between first and fourth instars showed that symbiont absence did not affect the ability ofA.tsugae to survive aestivation.Evaluations of sym- biont densities within each adelgid found that when S.symbiotica was absent,the density of obligate symbionts was significantly higher.Exposure to heat stress (32.5℃)was not consistently correlated with changes in symbiont densities over a 4-d period.Overall,we have shown that symbiont prevalence and densities vary within the broad population of A.tsugae in eastern North America,with potentially significant effects upon the ecology of this important pest.  相似文献
