In endothelial cells, H2O2 induces the rapid formation of focal adhesion complexes at the ventral face of the cells and a major reorganization of the actin cytoskeleton into dense transcytoplasmic stress fibers. This change in actin dynamics results from the activation of the mitogen-activated protein (MAP) kinase stress-activated protein kinase-2/p38 (SAPK2/p38), which, via MAP kinase-activated protein (MAPKAP) kinase-2/3, leads to the phosphorylation of the actin polymerization modulator heat shock protein of 27 kD (HSP27). Here we show that the concomitant activation of the extracellular signal-regulated kinase (ERK) MAP kinase pathway by H2O2 accomplishes an essential survival function during this process. When the activation of ERK was blocked with PD098059, the focal adhesion complexes formed under the plasma membrane, and the actin polymerization activity led to a rapid and intense membrane blebbing. The blebs were delimited by a thin F-actin ring and contained enhanced levels of HSP27. Later, the cells displayed hallmarks of apoptosis, such as DEVD protease activities and internucleosomal DNA fragmentation. Bleb formation but not apoptosis was blocked by extremely low concentrations of the actin polymerization inhibitor cytochalasin D or by the SAPK2 inhibitor SB203580, indicating that the two processes are not in the same linear cascade. The role of HSP27 in mediating membrane blebbing was assessed in fibroblastic cells. In control fibroblasts expressing a low level of endogenous HSP27 or in fibroblasts expressing a high level of a nonphosphorylatable HSP27, H2O2 did not induce F-actin accumulation, nor did it generate membrane blebbing activity in the presence or absence of PD098059. In contrast, in fibroblasts that expressed wild-type HSP27 to a level similar to that found in endothelial cells, H2O2 induced accumulation of F-actin and caused bleb formation when the ERK pathway was inhibited. Cis-platinum, which activated SAPK2 but induced little ERK activity, also induced membrane blebbing that was dependent on the expression of HSP27. In these cells, membrane blebbing was not followed by caspase activation or DNA fragmentation. We conclude that the HSP27-dependent actin polymerization–generating activity of SAPK2 associated with a misassembly of the focal adhesions is responsible for induction of membrane blebbing by stressing agents. 相似文献

We introduce here a simple approach for rapidly determining restriction
maps for a number of regions of a genome; this involves "anchoring" a map
with a rare restriction site (in this case the seldom-cutting EagI)
followed by partial digestion of a frequent-cutting enzyme (e.g., Sau 3A).
We applied this technology to five species of the Anopheles gambiae
complex. In a single Southern blot we obtained about a 15-kb restriction
map each for the mtDNA, rRNA gene, and a scnDNA region for each of five
species. Phylogenetic analyses of these regions yield trees at odds with
the more traditional chromosome inversion-based trees. The value of the
approach for systematic purposes is the ease with which several large,
independent regions of the genome can be quickly assayed for molecular
variation.

Major sulfur pools are quantified in soils and aboveground biomass of a coniferous boreal forest. Total ecosystem S averages
1395 kg·ha−1 of which 98% is found in the soil, with 89% being in the mineral horizons. Organic S dominates in soil, tree parts (trunks,
branches + foliage, roots) and litterfall, ranging from 77 to 99% of total S concentration. Carbon-bonded S, ester sulfate
and SO4-S in soil profiles range respectively from 51–68%, 29–37% and 1–14% of total S concentrations and account respectively for
57, 33 and 10% of total S on an areal basis. Adsorbed SO4 accounts for 82% of total SO4, and can be predicted from Al bound to organic matter, amorphous Al and pH (r2 = 0.81). There is a strong relationship between % carbon and carbon-bonded S in 4 of the 5 soil horizons studied which represent
over 95% of the total soil organic matter, whereas ester sulfate is related to % carbon in 3 soil horizons representing only
37% of the soil organic matter. An analysis of atmospheric S loading and S data for 10 forested sites in Europe and North

There are two distinct reasons for making comparisons of genetic variation for quantitative characters. The first is to compare evolvabilities, or ability to respond to selection, and the second is to make inferences about the forces that maintain genetic variability. Measures of variation that are standardized by the trait mean, such as the additive genetic coefficient of variation, are appropriate for both purposes. Variation has usually been compared as narrow sense heritabilities, but this is almost always an inappropriate comparative measure of evolvability and variability. Coefficients of variation were calculated from 842 estimates of trait means, variances and heritabilities in the literature. Traits closely related to fitness have higher additive genetic and nongenetic variability by the coefficient of variation criterion than characters under weak selection. This is the reverse of the accepted conclusion based on comparisons of heritability. The low heritability of fitness components is best explained by their high residual variation. The high additive genetic and residual variability of fitness traits might be explained by the great number of genetic and environmental events they are affected by, or by a lack of stabilizing selection to reduce their phenotypic variance. Over one-third of the quantitative genetics papers reviewed did not report trait means or variances. Researchers should always report these statistics, so that measures of variation appropriate to a variety of situations may be calculated. 相似文献

We have reviewed the available data on V(M), the amount of genetic variation in phenotypic traits produced each generation by mutation. We use these data to make several qualitative tests of the mutation-selection balance hypothesis for the maintenance of genetic variance (MSB). To compare V(M) values, we use three dimensionless quantities: mutational heritability, V(M)/V(E); the mutational coefficient of variation, CV(M); and the ratio of the standing genetic variance to V(M), V(G)/V(M). Since genetic coefficients of variation for life history traits are larger than those for morphological traits, we predict that under MSB, life history traits should also have larger CV(M). This is confirmed; life history traits have a median CV(M) value more than six times higher than that for morphological traits. V(G)/V(M) approximates the persistence time of mutations under MSB in an infinite population. In order for MSB to hold, V(G)/V(M) must be small, substantially less than 1000, and life history traits should have smaller values than morphological traits. V(G)/V(M) averages about 50 generations for life history traits and 100 generations for morphological traits. Plant responses to herbivory include tolerance (i.e. compensatory growth) and defense. Several factors influence the tolerance of a plant following herbivory, including plant genetic identity, site nutrient availability, and previous and/or concurrent herbivory. We studied the effects of these factors on the compensatory response of Salix planifolia ssp. planifolia, a shrub species common in the boreal and subarctic regions of North America. We cloned several genets of S. planifolia and submitted them to simulated root and/or leaf herbivory while varying the nutrient availability. Simulated leaf herbivory was more detrimental to the plant than simulated root herbivory, reducing both above- and below-ground tissue production. Leaf demography was unaffected by either simulated herbivory treatment. There was some compensatory growth following simulated leaf and root herbivory, but only the root compartment responded to increased nutrient availability. Simulated leaf herbivory increased leaf transpiration and reduced stomatal resistance, suggesting increased carbon fixation. The unexpected finding of the experiment was the absence of interactions among factors (genotype, nutrient availability and type of tissue damage) on the compensatory response of S. planifolia. In canine coronary artery preparations, the proteinase-activated receptor-2 (PAR(2)) activating peptides (PAR(2)-APs) SLIGRL-NH(2) and 2-furoyl-LIGRLO-NH(2) caused both an endothelium-dependent relaxation and an endothelium-independent contraction. Relaxation was caused at peptide concentrations 10-fold lower than those causing a contractile response. Although trans-cinnamoyl-LIGRLO-NH(2), like other PAR(2)-APs, caused relaxation, it was inactive as a contractile agonist and instead antagonized the contractile response to SLIGRL-NH(2). RT-PCR-based sequencing of canine PAR(2) revealed a cleavage/activation (indicated by underlines) sequence (SKGR/SLIGKTDSSLQITGKG) that is very similar to the human PAR(2) sequence (R/SLIGKV). As a synthetic peptide, the canine PAR-AP (SLIGKT-NH(2)) was a much less potent agonist than either SLIGRL-NH(2) or 2-furoyl-LIGRLO-NH(2), either in the coronary contractile assay or in a Madin-Darby canine kidney (MDCK) cell PAR(2) calcium signaling assay. In the MDCK signaling assay, the order of potencies was as follows: 2-furoyl-LIGRLO-NH(2) > SLIGRL-NH(2) = trans-cinnamoyl-LIGRLO-NH(2) > SLIGKT-NH(2), as expected for PAR(2) responses. In the coronary contractile assay, however, the order of potencies was very different: SLIGRL-NH(2) > 2-furoyl-LIGRLO-NH(2) > SLIGKT-NH(2), trans-cinnamoyl-LIGRLO-NH(2) = antagonist. Because of 1) the distinct agonist (relaxant) and antagonist (contractile) activity of trans-cinnamoyl-LIGRLO-NH(2) in the canine coronary contractile assays, 2) the different concentration ranges over which the peptides caused either relaxation or contraction in the same coronary preparation, and 3) the markedly distinct structure-activity profiles for the PAR-APs in the coronary contractile assay, compared with those for PAR(2)-mediated MDCK cell calcium signaling, we suggest that the canine coronary tissue possesses a receptor system for the PAR-APs that is distinct from PAR(2) itself. 相似文献

The specific shape of the relationship between plant diversity and productivity and the causal mechanism(s) behind the observed pattern(s) are still highly debated. Recent advances suggest that the relationship depends on several environmental variables and may change with the observational scale. In this study, a multivariate, multiscale approach was used to identify the variables that determine the relationship between species richness and annual production along a forest/old field edge in southern Québec (Canada). Various relationships between richness and production were found at different distances to the edge. In the forest, most relationships were positive and linear, while in the old field the relationship shifted from positive linear to non-significant with increasing distance from the edge. In the forest or in the old field, the shape of the relationship (all distances from the edge combined) was unimodal. Path analyses showed that species richness was determined mostly by production, which was influenced by different limiting resources, depending on the community (forest or old field). An increasing range in production created by pooling across community types can confound the resources and/or conditions determining the diversity-productivity relationship. 相似文献
