正确的系统发生重建对于理解进化事件至关重要.尽管分子系统学对于解决此类问题取得了极大的成功,由于一些诸如密码子使用偏性等的内在约束,来源于DNA的信息可能仍然存在着局限.因为发生在祖先的替代性转换,果蝇Drosophila saltans 5个种亚组由不同基因构建的分子系统树之间存在着冲突(在以往发表的分子系统学研究中,这些种组的每一个种亚组全少有一个代表).本文用40个形态学特征重新分析了这些种组.不同于以前发表的大多数假说,本研究支序分类学的结果表明,果蝇sturtevanti种亚组是一个较早的分支,而剩下的4个亚组形成一个支持度较高的类群;后者又可以再分为两个姐妹群:一个包含cordata和elliptica亚组,另一个包含parasaltans和saltans亚来组.本研究结果修正了果蝇saltans种组的分子进化(密码子使用偏性),并强调形态学对于系统发生重建和理解分子进化现象的重要作用.  相似文献

Unbiased methods to assess the firing activity of individual neurons in the neocortex have revealed that a large proportion of cells fire at extremely low rates (<0.1 Hz), both in their spontaneous and evoked activity. Thus, firing in neocortical networks appears to be dominated by a small population of highly active neurons. Here, we use a fosGFP transgenic mouse to examine the properties of cells with a recent history of elevated activity. FosGFP-expressing layer 2/3 pyramidal cells fired at higher rates compared to fosGFP(-) neurons, both in vivo and in vitro. Elevated activity could be attributed to increased excitatory and decreased inhibitory drive to fosGFP(+) neurons. Paired-cell recordings indicated that fosGFP(+) neurons had a greater likelihood of being connected to each other. These findings indicate that highly active, interconnected neuronal ensembles are present in the neocortex and suggest these cells may play a role in the encoding of sensory information. The number of neurosensory bristles on abdominal sternites of Drosophila is a most investigated trait for quantitative genetic studies. However, the developmental pattern expressed on successive segments in both sexes has remained so far a neglected field. We explored three aspects of this general problem with an isofemale line design: comparing two distantly related species, Drosophila melanogaster and Zaprionus indianus, investigating bristle number variation along the antero-posterior axis, and analysing the sexual dimorphism. Antero-posterior variations could be analysed from segment A2 to A7 in females, and A2-A5 in males. In D. melanogaster, males and females showed parallel changes with a consistently lower number in males. In Z. indianus females the number was quite stable along the abdomen, while in males an important antero-posterior increase was found. The sexual dimorphism was further analysed by considering the female-male correlation and the female/male ratio. The results suggest that sternite bristle number is determined by several developmental genetic systems. One is acting along the antero-posterior axis and may be associated to a gradient, since the genetic correlation decreases when more distant segments are compared. Another is acting in the same way on most segments of both sexes, since the female-male genetic correlation is similar between homologous and non-homologous segments. Finally, genes with specific sex effects are acting on A7 in females of both species, and on A5 in Z. indianus males. The overall architecture of female and male abdomen seems to be constrained by the development of reproductive organs. A large difference between species suggests, however, that the sexual dimorphism of abdominal bristle number is not evolutionarily constrained.  相似文献

We analyzed the vegetation of Lake Burullus at the deltaic Mediterranean coast of Egypt, the sand bar between its northern shore and Mediterranean Sea, the water courses that drain into the lake and the wetland around it. Our ultimate aim was to identify threatened species and communities and the environmental factors that affect their distribution in order to formulate a plan for their conservation. The total number of the recorded species was 197 (100 annuals and 97 perennials), including 12 floating and submerged hydrophytes. Three species are endemic to Egypt: two annuals (Sinapis arvensis subsp. allionii and Sonchus macrocarpus) and one perennial (Zygophyllum album var. album). Thirty-four species are rare allover Egypt (15 annuals and 19 perennials). The lake area included 10 types of habitat (sand formations, salt marshes, lake cuts, terraces, slopes, water edges and open water of the drains, islets, shores and lake). The vegetation was classified into 13 groups (i.e., plant communities). Six groups were dominated or co-dominated by the common reed (Phragmites australis); these groups occupied a wide environmental gradient from xeric to hydric habitats. Five other groups were dominated by halophytic species (Arthrocnemum macrostachyum, Suaeda vera, Sarcocornia fruticosa, Halocnemum strobilaceum and Salsola Kali). The remaining two groups were dominated by the emergent Typha domingensis and the submerged Potamogeton pectinatus. Moisture, salinity and sedimentation were the main factors that governed the plant succession in this wetland.  相似文献

mazEF is a stress-induced toxin-antitoxin module, located on the chromosome of Escherichia coli, that we have previously described to be responsible for programmed cell death in E. coli. mazF specifies a stable toxin, and mazE specifies a labile antitoxin. Recently, it was reported that inhibition of translation and cell growth by ectopic overexpression of the toxin MazF can be reversed by the action of the antitoxin MazE ectopically overexpressed at a later time. Based on these results, it was suggested that rather than inducing cell death, mazF induces a state of reversible bacteriostasis (K. Pederson, S. K. Christensen, and K. Gerdes, Mol. Microbiol. 45:501-510, 2002). Using a similar ectopic overexpression system, we show here that overexpression of MazE could reverse MazF lethality only over a short window of time. The size of that window depended on the nature of the medium in which MazF was overexpressed. Thus, we found "a point of no return," which occurred sooner in minimal M9 medium than it did in the rich Luria-Bertani medium. We also describe a state in which the effect of MazF on translation could be separated from its effect on cell death: MazE overproduction could completely reverse the inhibitory effect of MazF on translation, while not affecting the bacteriocidic effect of MazF at all. Our results reported here support our view that the mazEF module mediates cell death and is part of a programmed cell death network.  相似文献

Detergent-solubilized cell wall extracts of the gram-positive, strictly aerobic bacterium Nocardia asteroides contain channel-forming activity as judged from reconstitution experiments using lipid bilayer membranes. The cell wall porin was identified as a protein with an apparent molecular mass of about 84 kDa based on SDS-PAGE. The porin was purified to homogeneity using preparative SDS-PAGE. The 84-kDa protein was no longer observed after heating in SDS buffer. The presumed dissociation products were not observed on SDS-polyacrylamide gels. The cell wall porin increased the specific conductance of artificial lipid bilayer membranes from phosphatidylcholine/phosphatidylserine mixtures by the formation of cation-selective channels, which had an average single-channel conductance of 3.0 nS in 1 M KCl. The single-channel conductance was only moderately dependent on the bulk aqueous KCl concentration, which indicated negative point charge effects on the channel properties. The analysis of the concentration dependence of the single-channel conductance using the effect of negative charges on channel conductance suggested that the diameter of the cell wall channel is about 1.4 nm. Asymmetric addition of the cell wall porin to lipid bilayer membranes resulted in an asymmetric voltage dependence. The cell wall channel switched into substates, when the cis side of the membrane, the side of the addition of the protein, had negative polarity. Sequences of the internal transcribed spacer region 1 (ITS1) of the ribosomal DNA were used to determine the phylogenetic relationships of species ofTrichodermasect.Pachybasium.To this end, 85 strains—including all the availableex-type strains—were analyzed. Parsimony analysis demonstrated that the section is nonmonophyletic, distributing the 85 strains among three main groups that were supported by bootstrap values. Group A comprises two clades (A1 and A2), with A1 includingT. polysporum, T. piluliferum,andT. minutisporum,while A2 includedT. hamatum, T. pubescens,andT. strigosumin addition to species previously included in sect.Trichoderma(i.e.,T. viride, T. atroviride,andT. koningii). Theex-type strain ofT. fasciculatumformed a separate branch basal to clade A. Clade B contained the sect.PachybasiummembersT. harzianum, T. fertile, T. croceum, T. longipile, T. strictipile, T. tomentosum, T. oblongisporum, T. flavofuscum, T. spirale,and the anamorphs ofHypocrea semiorbisandH. cf. gelatinosa.Sequence differences among clades A1, A2, and B were in the same order of magnitude as between each of them andT. longibrachiatum,which was used as an outgroup in these analyses. Sequence differences within clades A1, A2, and B were considerably smaller: in some cases (i.e.,T. virensandT. flavofuscum; T. strictipileandH. cf. gelatinosa), the ITS1-sequences were identical, suggesting conspecifity. In other cases (e.g.,T. crassumandT. longipile; T. harzianum, T. inhamatum, T. croceum, T. fertile,andH. semiorbis; T. hamatumandT. pubescens;andT. viride, T. atroviride,andT. koningii) differences were in the range of 1–3 nt only, suggesting a very close phylogenetic relationship. The sequence of a previously described aggressive mushroom competitor group ofT. harzianumstrains (Th2) was strikingly different from that of theex-type strain ofT. harzianumand closely related species and is likely to be a separate species.  相似文献

Light‐regulated skin colour change is an important physiological process in invertebrates and lower vertebrates, and includes daily circadian variation and camouflage (i.e. background adaptation). The photoactivation of melanopsin‐expressing retinal ganglion cells (mRGCs) in the eye initiates an uncharacterized neuroendocrine circuit that regulates melanin dispersion/aggregation through the secretion of alpha‐melanocyte‐stimulating hormone (α‐MSH). We developed experimental models of normal or enucleated Xenopus embryos, as well as in situ cultures of skin of isolated dorsal head and tails, to analyse pharmacological induction of skin pigmentation and α‐MSH synthesis. Both processes are triggered by a melanopsin inhibitor, AA92593, as well as chloride channel modulators. The AA9253 effect is eye‐dependent, while functional data in vivo point to GABAA receptors expressed on pituitary melanotrope cells as the chloride channel blocker target. Based on the pharmacological data, we suggest a neuroendocrine circuit linking mRGCs with α‐MSH secretion, which is used normally during background adaptation.  相似文献

A channel-forming protein was identified in cell wall extracts of the Gram-positive, strictly aerobic bacterium Nocardia farcinica . The cell wall porin was purified to homogeneity and had an apparent molecular mass of about 87 kDa on tricine-containing SDS–PAGE. When the 87 kDa protein was boiled for a longer time in sodium dodecylsulphate (SDS) it dissociated into two subunits with molecular masses of about 19 and 23 kDa. The 87 kDa form of the protein was able to increase the specific conductance of artificial lipid bilayer membranes from phosphatidylcholine (PC) phosphatidylserine (PS) mixtures by the formation of ion-permeable channels. The channels had on average a single-channel conductance of 3.0 nS in 1 M KCl, 10 mM Tris-HCl, pH 8, and were found to be cation selective. Asymmetric addition of the cell wall porin to lipid bilayer membranes resulted in an asymmetric voltage dependence. The single-channel conductance was only moderately dependent on the bulk aqueous KCl concentration, which indicated point charge effects on the channel properties. The analysis of the single-channel conductance data in different salt solutions using the Renkin correction factor, and the effect of negative charges on channel conductance suggested that the diameter of the cell wall porin is about 1.4–1.6 nm. Channel-forming properties of the cell wall porin of N. farcinica were compared with those of mycobacteria and corynebacteria. The cell wall porins of these members of the order Actinomycetales share common features because they form large and water-filled channels that contain negative point charges.  相似文献
