The Oryza sativa (rice) resistance gene Pia confers resistance to the blast fungus Magnaporthe oryzae carrying the AVR-Pia avirulence gene. To clone Pia, we employed a multifaceted genomics approach. First, we selected 12 R-gene analog (RGA) genes encoding nucleotide binding site-leucine rich repeats (NBS-LRRs) proteins from a region on chromosome 11 that shows linkage to Pia. By using seven rice accessions, we examined the association between Pia phenotypes and DNA polymorphisms in the 10 genes, which revealed three genes (Os11gRGA3-Os11gRGA5) exhibiting a perfect association with the Pia phenotypes. We also screened ethyl methane sulfonate (EMS)-treated mutant lines of the rice cultivar 'Sasanishiki' harboring Pia, and isolated two mutants that lost the Pia phenotype. DNA sequencing of Os11gRGA3-Os11gRGA5 from the two mutant lines identified independent mutations of major effects in Os11gRGA4. The wild-type 'Sasanishiki' allele of Os11gRGA4 (SasRGA4) complemented Pia function in both mutants, suggesting that SasRGA4 is necessary for Pia function. However, when the rice cultivar 'Himenomochi' lacking Pia was transfected with SasRGA4, the Pia phenotype was not recovered. An additional complementation study revealed that the two NBS-LRR-type R genes, SasRGA4 and SasRGA5, that are located next to each other and oriented in the opposite direction are necessary for Pia function. A population genetics analysis of SasRGA4 and SasRGA5 suggests that the two genes are under long-term balancing selection. 相似文献

Condensation of Z-PPh2CH2C(But)=NNH2 with 4-nitroacetophenone gave the azine phosphine Z,E-PPh2CH2C(But)=N-N=CMe(C6H4NO2-4) (I). The corresponding phsophine oxide II was prepared by treatment of I with H2O2. The phosphine I with [Mo(CO)4(nbd)] (nbd=norbornadiene) gave [Mo(CO)4{PPh2CH2C(But)=N-N=CMe(C6H4NO2-4)}] (1a); the corresponding tungsten 1b and chromium 1c complexes were made similarly. The crystal structure of 1a was determined by X-ray diffraction and showed the presence of a six-membered chelate ring with the bulky 4-nitrophenyl group held close to the metal. Oxidation of 1a with bromine gave the seven-coordinate molybdenum (II) complex 2. Treatment of [PtMe2(cod)] (cod=cycloocta-1,5-diene) with I at 20°C gave the dimethyl-platinum (II) complex [PtMe2{PPh2CH2C(But)=N-N=CMe(C6H4NO2-4)}] (3a) which with MeI gave the iodotrimethylplatinum(IV) complex 4. Treatment of 3a with C≡O opened the chelate ring to give the dimethyl(carbonyl)platinum(II) complex 5 containing a monodentate phosphine ligand. When 3a was heated in toluene solution at 110°C it gave the cyclometallated methylplatinum(II) complex [PtMe{PPh2CH2C(But)=N-N=CMe(C6H3NO2-4)}] (6). Treatment of 6 with MeI gave the platinum(IV) complex 7. The dichloropalladium(II) complex [PdCl2{PPh2CH2C(But)=N-N=CMe(C6H4NO2-4)}] (3b) was prepared by treatment of [PdCl2(NCPh)2] with I in CH2Cl2. Treatment of [PtCl2(NCMe)2] with 2 equiv. of I gave the trans-bis(phosphine) complex 8. When 2 equiv. of I were treated with [PtCl2(cod)] followed by NH4PF6 this gave the salt 9a containing two six-membered chelate rings; the analogous palladium(II) 9b) salt was also prepared. Treatment of 2 equiv. of I with [PtCl2(cod)] followed by NH4PF6 gave the PF6 salt 10 containing a six-membered chelate ring and a monodentate ligand. When 10 was treated with AgNO3 followed by NH4PF6 this gave the bis-chelate complex 11 containing five- and six-membered chelate rings. Treatment of [IrCl(CO)2(p-toluidine)] with I gave the cyclometallated iridium(III) hydride complex [IrHClCO{PPh2CH2C(But)=N-N=CMe(C6H3NO2-4)}] (12). [RuCl2(PPh3)3] with the phosphine I resulted in the Ru(II) complex 13 in which the ortho hydrogens of the 4-nitrophenyl group are agostically interacting with ruthenium. Proton, Phosphorus-31, some carbon-13 NMR and IR data have been obtained. Traumatic brain injury (TBI) can occur anywhere along the cortical mantel. While the cortical contusions may be random and disparate in their locations, the clinical outcomes are often similar and difficult to explain. Thus a question that arises is, do concussions at different sites on the cortex affect similar subcortical brain regions? To address this question we used a fluid percussion model to concuss the right caudal or rostral cortices in rats. Five days later, diffusion tensor MRI data were acquired for indices of anisotropy (IA) for use in a novel method of analysis to detect changes in gray matter microarchitecture. IA values from over 20,000 voxels were registered into a 3D segmented, annotated rat atlas covering 150 brain areas. Comparisons between left and right hemispheres revealed a small population of subcortical sites with altered IA values. Rostral and caudal concussions were of striking similarity in the impacted subcortical locations, particularly the central nucleus of the amygdala, laterodorsal thalamus, and hippocampal complex. Subsequent immunohistochemical analysis of these sites showed significant neuroinflammation. This study presents three significant findings that advance our understanding and evaluation of TBI: 1) the introduction of a new method to identify highly localized disturbances in discrete gray matter, subcortical brain nuclei without postmortem histology, 2) the use of this method to demonstrate that separate injuries to the rostral and caudal cortex produce the same subcortical, disturbances, and 3) the central nucleus of the amygdala, critical in the regulation of emotion, is vulnerable to concussion. 相似文献

Epidemiological studies suggest that precursor steroids are implicated in the aetiology of breast cancer. However, our understanding of the role of precursor steroids in breast cancer is complicated by fact that there are many precursor steroids, which are metabolically inter-related and have divergent proliferative activities on the growth of breast cancer cell lines. In this study the proliferative affects of 5 alpha-dihydrotestosterone and 5-androstene-3 beta,17 beta-diol, which may be considered true metabolites acting at a tissue level, on MCF7, T47D and MDAMB231 breast cancer cell lines have been examined by a flow cytometric technique. DNA cell cycle analysis demonstrates that 5-androstene-3 beta,17 beta-diol stimulates the proliferation of hormone-dependent cell lines at physiological levels by an oestrogen receptor mediated mechanism whereas 5 alpha-dihydrotestosterone does not affect the proliferation of MCF7 and T47D cell lines at physiological levels over short (48 h) incubations. Both 5 alpha-dihydrotestosterone and 5-androstene-3 beta,17 beta-diol stimulate proliferation of hormone-dependent cell lines at pharmacological levels via and interaction with the oestrogen receptor. In long (6-9 days) incubations both 5 alpha-dihydrotestosterone and 5-androstene-3 beta,17 beta-diol inhibit the 17 beta-oestradiol induced proliferation of MCF7 and T47D cell lines, however, 5 alpha-dihydrotestosterone inhibits while 5-androstene-3 beta,17 beta-diol stimulates basal proliferation. Many viruses alter different stages of apoptosis of infected cells as a strategy for successful infection. Few studies have addressed mechanisms of equine herpesvirus 1 (EHV-1) strain-induced cell death. We investigated the effect of an abortigenic strain (AR8 strain) on heterologous Madin–Darby bovine kidney cells and homologous equine dermis (ED) cells cell lines. We compared morphologic and biochemical features of early and late apoptosis at different postinfection times. We investigated translocation of phosphatidylserine to the cell surface, nuclear fragmentation and changes in the cytoskeleton using flow cytometry and annexin V/propidium iodide staining, DNA laddering, terminal deoxynucleotidyl transferase UTP nick-end labeling assay and immunofluorescence staining of cytokeratin 18 cleavage. AR8 EVH-1 strain interfered with apoptosis in both cell lines, particularly during the middle stage of the replication cycle; this was more evident in ED cells. Although this antiapoptotic effect has been reported for other alpha herpesviruses, our findings may help elucidate how EHV-1 improves its infectivity during its cycle. 相似文献
