The microbial surface and flocculability were qualitatively characterized through the combination of the surface thermodynamic and the extended DLVO approaches, with Ralstonia eutropha, a polyhydroxybutyrate-producing bacterium, as an example. The negativity of the ζ potential of R. eutropha decreased from the initial −19.5 to −11 mV in its cultivation with the consumption of glucose. The total interfacial free energy (ΔG adh) was changed from −80 to 28.5 mJ m−2 in its entire growth process. This suggests that the bacterial surface changed from hydrophobic into hydrophilic, resulting in an alteration of its surface characteristics and flocculability in its different growth phases. As a result, the stability ratio of suspensions increased with the increasing cultivation time, indicating that the cell particles became more repulsive with each other and led to a more stable suspension of R. eutropha in its cultivation. The obtained information in this work might be useful for better understanding the surface characteristics and the flocculability and even manipulating its flocculability in the microbial growth process.  相似文献

The S-layer protein CTC surface display system of Bacillus thuringiensis was used to test the possibility of displaying avian influenza virus nucleoprotein (NP) on the cell surface of B. thuringiensis. By fusing np with the anchoring motif of ctc, four recombinant plasmids were constructed. They harbored fusion gene ctc-np, csa-ctc-np (csa representing csaAB operon, very important in anchoring S-layer protein on cell surface), ctc-npp (npp representing the part fragment of np), and csa-ctc-npp, respectively. Five recombinant strains were obtained by transferring recombinant plasmids to B. thuringiensis plasmid-free derivative strain BMB171. The vegetative cells of five strains were used as agglutinogens for slide agglutination assays. The assays showed recombinant NP proteins successfully displayed on the cell surface of five strains. After immunization of chickens with spores by oral route, all five strains elicited a humoral response to NP and exhibited immunogenicity as indicated by enzyme-linked immunosorbent assay (ELISA). ELISA also showed that one of five strains, CN (bearing csa-ctc-npp), exhibited the highest immunogenicity among five strains, which suggested that the best way of constructing ctc fusion gene was the csa-ctc-npp. The strategy developed in this study suggests the possibility of generating a heat-stable and oral veterinary vaccine with B. thuringiensis surface display system.  相似文献

To prevent in vivo degradation, small peptides are usually expressed in fusion proteins from which target peptides can be released by proteolytic or chemical reagents. In this report, small ubiquitin-related modifier (SUMO) linked with a hexa-histidine tag was used as a fusion partner for the production of recombinant human urodilatin, a hormone for the treatment of acute decompensated heart failure. The fusion protein, which was overexpressed mainly as inclusion bodies in Escherichia coli, constituted about 25% of the total cell proteins. After purification by Ni-sepharose affinity chromatography and renaturation in refolding buffer, the fusion protein was cleaved with SUMO protease 1. Urodilatin was separated from the fusion partner by the subtractive chromatography using Ni-sepharose once again, and then further purified with reverse-phase high performance liquid chromatography. In vitro activity assay demonstrated that the recombinant urodilatin had a potent vasodilatory effect on rabbit aortic strips with an EC50 of 1.77 ± 0.53 μg/ml, which was similar to that of the synthetic urodilatin standard. The expression strategy presented in this study allows convenient high yield and easy purification of small recombinant peptides with native sequences. Z. Sun and Z. Diet composition and prey selection of pike (Esox lucius) were studied in Çivril Lake, Turkey. The lake is eutrophic in character with a surface area of ca. 64 km?2 and mean depth of 3 m. Stomach contents of 409 specimens were collected between October 2003 and April 2005. Pike diet included 10 prey fish species, two Crustaceae, four Insecta, one Hirudinae and one Amphibia. Feeding was homogeneous, with most pike specializing in prey fish and a few pike specializing on miscellaneous items. Feeding activity varied by season and ontogeny. Stomach fullness and the percentage of fish with prey were highest in spring and in small pike, while feeding intensity was lowest in winter and in large sized pike. Diet composition was dominated by fish, including Carasius gibelio, Chondrostoma meandrense, Esox lucius, Gambusia affinis, Gobio gobio, Hemigrammocapoeta kemali, Leuciscus cephalus, and Tinca tinca. Crustacea were also a significant component in spring and in small sized pike. The most important prey items were C. meandrense, Gammarus sp., H. kemali, and L. cephalus. Pike feeding in winter and summer was homogeneous, specializing mainly on fish as prey, while the diet in spring and autumn was heterogeneous with some pike specializing on Gammarus sp. Cannibalism at 8.7% was observed only in the large sized pike (>40 cm). Pike strongly preferred C. meandrense (Selectivity index V = 0.372; χ2 = 27.739; P < 0.01), G. gobio (V = 0.192; χ2 = 7.350; P < 0.01) and T. tinca (V = 0.146; χ2 = 4.257; P < 0.05) despite their low abundance in the lake. Hemigrammocapoeta kemali was the most abundant prey fish in the environment; however, it was a negatively selected food item (V = ?0.358; χ2 = 25.642; P < 0.01). Cyprinus carpio also inhabits the lake, but was not preferred by pike (V = ?0.056; χ2 = 0.625; P > 0.05).  相似文献

Chromium picolinate is a popular nutritional supplement whose safety has been questioned because of the potential risk of oxidative DNA damage. To investigate this possibility, a dose-dependent study was performed in piglet hepatocyte cultures in which low (8 microM), medium (200 microM), and high (400 microM) doses of chromium picolinate were tested and compared to untreated controls. After 48 h incubation, there were no significant differences in the levels of intracellular reactive oxygen species, medium lactate dehydrogenase activity, and comet indicators between the three experimental groups and controls (p > 0.05). In the 8 microM-treated group, the intracellular malondialdehyde content was significantly decreased relative to controls (p < 0.05). All of the studied parameters showed a dose-dependent increase that was statistically significant between the low and high doses (p < 0.05). These results suggest that: (1) chromium picolinate may affect the oxidative status of piglet hepatocytes; (2) the appropriate dose (approximately physiological concentration) of chromium picolinate can inhibit lipid peroxidation, and (3) high doses of chromium picolinate have no significant effects on oxidative damage in piglet hepatocytes, but the existing evidence also imply that exposure to a higher dose appears to be unwarranted.  相似文献

The protective effects of single dose of garlic oil (GO) on acute ethanol-induced fatty liver were investigated. Mice were treated with ethanol (4.8 g/kg bw) to induce acute fatty liver. The liver index, the serum and hepatic triglyceride (TG) levels and the histological changes were examined to evaluate the protective effects. Hepatic malondialdehyde (MDA), glutathione (GSH) levels and superoxide dismutase (SOD), glutathione reductase (GR), glutathione peroxidase (GSH-Px), glutathione-S-transferase (GST) activities were determined for the antioxidant capacity assay. Acute ethanol exposure resulted in the enlargement of the liver index and the increase of the serum and hepatic TG levels (P<0.01), which were dramatically attenuated by GO pretreatment in a dose-dependent manner (P<0.01). GO treatment (simultaneously with ethanol exposure) exhibited similar effects to those of pretreatment, while no obviously protective effects were displayed when it was used at 2h after ethanol intake. Histological changes were paralleled to these indices. Beside this, GO dramatically prolonged the drunken time and shortened the waking time, and these effects were superior to those of silymarin and tea polyphenol. In addition, GO dose-dependently suppressed the elevation of MDA levels, restored the GSH levels and enhanced the SOD, GR and GST activities. Compared with the ethanol group, the MDA levels decreased by 14.2% (P<0.05), 29.9% and 32.8% (P<0.01) in GO groups 50, 100 and 200 mg/kg, respectively. The GST activity increased by 9.97%, 19.94% (P<0.05) and 42.12% (P<0.01) of the ethanol group in GO groups 50, 100 and 200 mg/kg, respectively, while the GR activity increased by 28.57% (P<0.05), 37.97% (P<0.01), 50.45% (P<0.01) of the ethanol group in GO groups 50, 100 and 200 mg/kg, respectively. These data indicated that single dose of GO possessed ability to prevent acute ethanol-induced fatty liver, but may lose its capacity when used after ethanol exposure. The protective effects should be associated with its antioxidative activities.  相似文献

Ethanol‐dichloromethane crude extract from peel of pear (Pyrus bretschneideri Rehd. cv. Pingguoli) was separated by thin layer chromatographic plates and bioassayed with conidia of Alternaria alternata. The inhibition zones differed significantly in retention factor (Rf) at expanding stage, harvest time and after 100 days of cold storage. The compounds in the inhibition zones were isolated and identified with gas chromatography and mass spectroscopy. Palmitate methyl, oleic acid methyl, linolenic acid methyl and squalene were present at all stages. The concentration of these chemicals was the highest in expanding stage fruit peel and decreased rapidly with fruit development. It is suggested that these compounds may be the main antifungal compounds in the growing fruit. The phthalate alkyl esters occurred at relatively higher concentrations in pear peel at harvest and after 100 days of cold storage. Six phthalate alkyl esters were identified from peel of pear fruit after 100 days of cold storage. It is also supposed that these esters may be the antifungal compounds in postharvest pear.  相似文献
