Conformational studies of Escherichia coli pyruvate oxidase

In this study the effects of experimental modifications of plasma membrane lipid lateral mobility on the electrical membrane properties and cation transport of mouse neuroblastoma cells, clone Neuro-2A, have been studied. Short-term supplementation of a chemically defined growth medium with oleic acid or linoleic acid resulted in an increase in the lateral mobility of lipids as inferred from fluorescence recovery after photobleaching of the lipid probe 3,3'-dioctadecylindocarbocyanide iodide. These changes were accompanied by a marked depolarization of the membrane potential from -51 mV to -36 mV, 1.5 h after addition, followed by a slow repolarization. Tracer flux studies, using 86Rb+ as a radioactive tracer for K+, demonstrated that the depolarization was not caused by changes in (Na+ + K+)-ATPase-mediated K+ influx or in the transmembrane K+ gradient. The permeability ratio (PNa/PK), determined from electrophysiological measurements, however, increased from 0.10 to 0.27 upon supplementation with oleic acid or linoleic acid. This transient rise of PNa/PK was shown by 24Na+ and 86Rb+ flux measurements to be due to both an increase of the Na+ permeability and a decrease of the K+ permeability. None of these effects occurred upon supplementation of the growth medium with stearic acid.     

The complete amino acid sequence of bovine liver catalase and the partial sequence of bovine erythrocyte catalase

The data upon which the sequence of the 506 residues in the subunit of bovine liver catalase (BLC) is based are presented in detail. A partial sequence of bovine erythrocyte catalase (BEC) which accounts for 493 residues shows complete concordance with the BLC data. On the other hand, BEC has at least 517 residues, that is, an extension beyond the C terminus of the BLC data. Although normally BLC has only 506 residues, there is evidence that, at some point in its history, it also had the C-terminal extension. It is speculated that this extension is lost in BLC either through a different processing of the molecule in liver than in erythrocytes or by partial degradation in the first stages of catabolism.     

The production of mixed-species Bos taurus-Bos indicus twin calves

Bos taurus-Bos indicus twin calves were produced by transferring Day 6–8 Bos indicus or Bos taurus embryos to previously inseminated Bos taurus or Bos indicus cows. Embryos were transferred to 112 recipients of which 64 (57.1%) were diagnosed as pregnant by rectal palpation at 7–8 weeks gestation. Twin foetuses were diagnosed in 38 (65.5%) of 58 cows in which the location and number of foetuses was recorded. Substantial foetal losses occurred after 7–8 weeks and only 44 cows calved giving 21 sets of twins and 23 singletons. Six of the single calves were from transferred embryos. The duration of gestation of the twin pregnancies (282 days) was similar to that of single Bos taurus pregnancies (282 days for Jerseys and 286 days for Friesians) and 14 days shorter than single Bos indicus pregnancies (296 days). Consequently Bos indicus members of mixed-species twins were premature at birth and required intensive care to ensure their survival.
