Among glycoside hydrolases, beta-glucosidase plays a unique role in many physiological and biocatalytical processes that involve the beta-linked O-glycosyl bond of various oligomeric saccharides or glycosides. Structurally, the enzyme can be grouped into glycoside hydrolase family 1 and 3. Although the basic ("retaining, double-displacement") mechanism for the catalysis of family 3 beta-glucosidase has been established, in-depth understanding of its structure-function relationship, particularly the substrate specificity that is of great interest for developing the enzyme as a versatile biocatalyst, remains limited. To further probe the active site, we carried out a comparative study on a family 3 beta-glucosidase from Aspergillus oryzae with substrates and competitive inhibitors of different structures, in attempt to evaluate the site-specific spatial and chemical interactions between a pyranosyl substrate and the enzyme. Our results showed the enzyme having a strict stereochemical requirement (to accommodate beta-d-glucopyranose) for its "-1" active subsite, in contrast to its family 1 counterpart. 相似文献

Tissue microenvironments can regulate cell behavior by imposing physical restrictions on their geometry and size. An example
of these phenomena is cardiac morphogenesis, where morphometric changes in the heart are concurrent with changes in the size,
shape, and cytoskeleton of ventricular myocytes. In this study, we asked how myocytes adapt their size, shape, and intracellular
architecture when spatially confined in vitro. To answer this question, we used microcontact printing to physically constrain
neonatal rat ventricular myocytes on fibronectin islands in culture. The myocytes spread and assumed the shape of the islands
and reorganized their cytoskeleton in response to the geometric cues in the extracellular matrix. Cytoskeletal architecture
is variable, where myocytes cultured on rectangular islands of lower aspect ratios (length to width ratio) were observed to
assemble a multiaxial myofibrillar arrangement; myocytes cultured on rectangles of aspect ratios approaching those observed
in vivo had a uniaxial orientation of their myofibrils. Using confocal and atomic force microscopy, we made precise measurements
of myocyte volume over a range of cell shapes with approximately equal surface areas. When myocytes are cultured on islands
of variable shape but the same surface area, their size is conserved despite the changes in cytoskeletal architecture. Our
data suggest that the internal cytoskeletal architecture of the cell is dependent on extracellular boundary conditions while

The COVID-19 pandemic has placed unprecedented pressure on biopharmaceutical companies to develop efficacious preventative and therapeutic treatments, which is unlikely to abate in the coming years. The importance of fast progress to clinical evaluation for treatments, which tackle unmet medical needs puts strain on traditional product development timelines, which can take years from start to finish. Although previous work has been successful in reducing phase 1 timelines for recombinant antibodies, through utilization of the latest technological advances and acceptance of greater business risk or costs, substantially faster development is likely achievable without increased risk to patients during initial clinical evaluation. To optimize lessons learned from the pandemic and maximize multi-stakeholder (i.e., patients, clinicians, companies, regulatory agencies) benefit, we conducted an industry wide benchmarking survey in September/October 2021. The aims of this survey were to: (i) benchmark current technical practices of key process and product development activities related to manufacturing of therapeutic proteins, (ii) understand the impact of changes implemented in COVID-19 accelerated Ab programs, and whether any such changes can be retained as part of sustainable long-term business practices and (iii) understand whether any accelerative action(s) taken have (negatively) impacted the wider development process. This article provides an in-depth analysis of this data, ultimately highlighting an industry perspective of how biopharmaceutical companies can sustainably adopt new approaches to therapeutic protein development and production. 相似文献

Dental plaque is a natural biofilm which has been a focus of attention for many years because of its known roles in caries and periodontal diseases. Acid production by plaque bacteria leads to the erosion of tooth mineral in caries, and the cariogenicity of plaque is related to population levels of acid-tolerant organisms such as mutants streptococci. However, the biofilm character of plaque allows for survival of a diverse flora, including less acid-tolerant organisms, some of which can produce ammonia from arginine or urea to counter acidification. Plaque is often considered to be relatively anaerobic. However, evidence is presented here that both supragingival and subgingival plaque have active oxygen metabolism and that plaque bacteria, including anaerobes, have developed defenses against oxidative stress. Even in subgingival plaque associated with periodontitis, measured residual oxygen levels are sufficient to allow for oxygen metabolism by organisms considered to be extremely anaerobic such asTreponema denticola, which metabolizes oxygen by means of NADH oxidases and produces the protective enzymes superoxide dismutase and NADH peroxidase. The finding that plaque bacteria produce a variety of protective enzymes is a good indicator that oxidative stress is a part of their everyday life. The biofilm character of plaque allows for population diversity and coexistence of aerobes, anaerobes and microaerophiles. Overall, agents that affect oxidative metabolism offer possibilities for reducing the pathogenic activities of plaque. 相似文献

The genotypic heterogeneity of Streptococcus oralis isolated from the oral cavity was investigated using repetitive extragenic palindromic PCR. Unrelated subjects harbored unique genotypes, with numerous genotypes being isolated from an individual. S. oralis is the predominant aciduric bacterium isolated from noncarious tooth sites. Genotypic comparison of the aciduric populations isolated at pH 5.2 with those isolated from mitis-salivarius agar (MSA) (pH 7.0) indicated that the aciduric populations were genotypically distinct in the majority of subjects (χ2 = 13.09; P = 0.0031). Neither the aciduric nor the MSA-isolated strains were stable, with no strains isolated at baseline being isolated 4 or 12 weeks later in the majority of subjects. The basis of this instability is unknown but is similar to that reported for Streptococcus mitis. Examination of S. oralis strains isolated from cohabiting couples demonstrated that in three of five couples, genotypically identical strains were isolated from both partners and this was confirmed by using Salmonella enteritidis repetitive element PCR and enterobacterial PCR typing. These data provide further evidence of the physiological and genotypic heterogeneity of non-mutans streptococci. The demonstration of distinct aciduric populations of S. oralis implies that the role of these and other non-mutans streptococci in the caries process requires reevaluation. 相似文献

Background
Inhibition of parasite growth is a major objective of blood-stage malaria vaccines. The in vitro assay of parasite growth inhibitory activity (GIA) is widely used as a surrogate marker for malaria vaccine efficacy in the down-selection of candidate blood-stage vaccines. Here we report the first study to examine the relationship between in vivo Plasmodium falciparum growth rates and in vitro GIA in humans experimentally infected with blood-stage malaria.Methods
In this phase I/IIa open-label clinical trial five healthy malaria-naive volunteers were immunised with AMA1/C1-Alhydrogel+CPG 7909, and together with three unvaccinated controls were challenged by intravenous inoculation of P. falciparum infected erythrocytes.Results
A significant correlation was observed between parasite multiplication rate in 48 hours (PMR) and both vaccine-induced growth-inhibitory activity (Pearson r = −0.93 [95% CI: −1.0, −0.27] P = 0.02) and AMA1 antibody titres in the vaccine group (Pearson r = −0.93 [95% CI: −0.99, −0.25] P = 0.02). However immunisation failed to reduce overall mean PMR in the vaccine group in comparison to the controls (vaccinee 16 fold [95% CI: 12, 22], control 17 fold [CI: 0, 65] P = 0.70). Therefore no impact on pre-patent period was observed (vaccine group median 8.5 days [range 7.5–9], control group median 9 days [range 7–9]).Conclusions
Despite the first observation in human experimental malaria infection of a significant association between vaccine-induced in vitro growth inhibitory activity and in vivo parasite multiplication rate, this did not translate into any observable clinically relevant vaccine effect in this small group of volunteers.Trial Registration

Monthly surveys of Bewick's Swans Cygnus columbianus bewickii , Whooper Swans Cygnus cygnus and Mute Swans Cygnus olor in Britain and Ireland were made during the 1990–1991 winter to determine factors affecting the swans' selection of feeding sites. Geographic location and habitat both influenced site selection. Whooper Swans occurred in greatest numbers at sites in Scotland, northeastern England and Northern Ireland, whereas Bewick's Swans had a more southerly distribution, reflecting differences in the migratory routes used by these two species. The resident Mute Swans were more widespread, with large flocks occurring in southeastern England and in parts of Scotland. Whooper and Mute Swans were found mainly on permanent inland waters (68% and 61%, respectively), but the majority of Bewick's Swans (60%) were on arable land. The percentage of Bewick's Swan flocks found on permanent inland waters (42%) was higher than that found on arable fields (23%), indicating that the large number recorded on arable land was a result of the birds congregating at a comparatively small number of sites. Overall, less than 15% of Whooper Swans and 3% of Mute Swans were on arable crops during the winter, but the largest flocks were associated with arable land for all three species. Thus, although the occurrence of large flocks at particular arable sites may give an impression that swans feed mainly on farmland, the swans are in fact more widely dispersed. Regional variation in the percentage of juveniles present was recorded for all three species. Changes during the winter in the distribution of juveniles, and of the swans as a whole, are considered in relation to food supply and to migratory routes for the Bewick's and Whooper Swans. 相似文献

Premature yeast flocculation (PYF) is a sporadic fermentation problem in the brewing industry that results in incomplete yeast utilization of fermentable sugars in wort. Culture-independent, PCR-based fingerprinting techniques were applied in this study to identify the associations between the occurrence of the PYF problem during brewery fermentation with barley malt-associated microbial communities (both bacteria and fungi). Striking differences in the microbial DNA fingerprint patterns for fungi between PYF positive (PYF +ve) and negative (PYF ?ve) barley malts were observed using the terminal restriction fragment length polymorphism (TRFLP) technique. The presence of terminal restriction fragments (TRFs) of 360–460 bp size range, for fungal HaeIII restriction enzyme-derived TRFLP profiles appeared to vary substantially between PYF +ve and PYF ?ve samples. The source of the barley malt did not influence the fungal taxa implicated in PYF. TRFLP analysis indicates bacterial taxa are unlikely to be important in causing PYF. Virtual digestion of fungal sequences tentatively linked HaeIII TRFs in the 360–460 bp size range to a diverse range of yeast/yeast-like species. Findings from this study suggest that direct monitoring of barley malt samples using molecular methods could potentially be an efficient and viable alternative for monitoring PYF during brewery fermentations. 相似文献

In the first leaf of wheat (Triticum aestivum L. cv. Capelle) the content of soluble protein diminished to about 50% of the initial value between the 7th and the 19th day after sowing. In order to understand proteolysis in the leaves, the activities of several peptidases were measured in extracts from leaves at four different ages. The carboxypeptidase activities increased during the growth of the leaves, and then began to decrease. The activities of the alkaline peptidases increased, while that of the benzoyl-DL-arginine-p-nitroanilide (BAPA) hydrolysing enzyme decreased during the whole period studied. The “naphthylamidase” activities showed first a slow rise, but then leveled off. Two bands with naphthylamidase activity could be detected after disc electrophoresis. All the peptidases studied were present in the leaves at rather high concentrations. This indicates that they all may participate in the hydrolysis of leaf proteins into free amino acids. 相似文献
