Phenotypic evolutionary rates were measured for 27 craniometric characters in 12 extant OTUs from the bat genus Myotis (Chiroptera, Vespertilionidae). Squared Mahalanobis distance was used as a multivariate measure for amount of divergence, and squared Mahalanobis distance weighted by time was used as a measure for the rate of divergence. Estimates for the rates of divergence were found to be consistent with random walk hypothesis. Thus, the divergence in Myotis could be guided by random drift and mutations. The high dispersion in rate estimates suggests also a possible input of randomly fluctuating selection. The highest rates were recorded for divergence between M. myotisM. blythii species group and the other OTUs. Rates of divergence between the subspecies of M. blythii occur to be lesser than rates of divergence between the earlier diverged species, their divergence could probably be slowed down by stabilizing selection. Size-adjusted data appeared to be lesser then the initial data, and it can be concluded that both size and shape were involved in divergence of Myotis species. The skeletal characters in bats are known to be extremely conservative during long-term evolution, however, the possibility for random walk at short time interval implies that bat evolution is constrained rather ecologically and biomechanically than genetically or developmentally.  相似文献

A new isolate designated as strain EB172 was isolated from a digester treating palm oil mill effluent and was investigated by polyphasic taxonomic approach. The cells were rod-shaped, Gram-negative, non-pigmented, non-spore-forming and non-fermentative. Phylogenetic analysis using the 16S rRNA gene sequence showed that the strain clustered with the genus Comamonas. Its closest neighbours were the type strains Comamonas terrigena (96.8%), Comamonas koreensis (93.4%), Comamonas composti (92.9%), and Comamonas kerstersii (91.1%). The ability of the strain EB172 to produce polyhydroxyalkanoates (PHA) when supplied with organic acids made this bacterium unique among Comamonas species. The bacterial strain was clearly distinguished from all of the existing strains by phylogenetic analysis, fatty acid composition and a range of physiological and biochemical characteristics. The G+C content of the genomic DNA was 59.1 mol%. The strain showed good growth in acetic, propionic and n-butyric acids. Comamonas sp. EB172 produced 9.8 g/l of cell dry weight and accumulated 59 (wt%) of PHAs when supplemented with mixed organic acids from anaerobically treated palm oil mill effluent. It is evident from the genotypic, phenotypic data and ability to produce PHAs that strain EB172 represents a new strain in the genus Comamonas (GeneBank accession no. EU847238).  相似文献

This study aimed to investigate the effects of aqueous cinnamon extract (ACE) on 7, 12-Dimethylbenz[a]anthracene (DMBA)-induced oral carcinogenesis in hamster cheek pouch (HCP) mucosa. Sixty male Syrian hamsters were randomly divided into six equal groups. The hamsters of groups I, II and III received no treatment, DMBA and ACE respectively, for 16 weeks. Groups IV and V were handled as group II and concomitantly treated with ACE for the same period and additionally group V received ACE for other 16 weeks after the stoppage of DMBA application. Group VI hamsters were handled as group III and additionally received DMBA for other 16 weeks after the stoppage of ACE supplementation. Hamsters of each group were euthanized according to the experimental schedule. The buccal pouches were and prepared for H&E stain, PAS reagent, CD3 and PDGF immunohistochemical reactivity. All groups showed dysplastic changes with varying degrees except groups I and III. Deep invasive carcinomas were recorded in 90% of the samples of group II, 60% of group IV, 50% of group V and 40% of group VI. From the previous results, it can be concluded that ACE has the potentiality preventing oral cancer initiation better than inhibiting oral cancer progression.  相似文献

Basic fibroblast growth factor (bFGF; FGF-2) is one of 19 related members of a growth factor family with mitogenic and hormone-regulatory functions. In Xenopus laevis oocytes, a 1.5-kb FGF-2 antisense (GFG) RNA complementary to the third exon and 3'-untranslated region (UTR) of FGF-2 mRNA has been implicated in FGF-2 mRNA editing and stability. The human homolog has been cloned, and we localized this gene by yeast artificial chromosome (YAC), somatic cell, and radiation hybrid panels to the same chromosomal site as FGF-2 (chromosome 4, JO4513 adjacent to D4S430), confirming this as a human endogenous antisense gene. The full-length GFG antisense RNA encodes a 35-kDa protein, which is highly homologous with the MutT family of antimutator nucleosidetriphosphatases (NTPases). We show that human pituitary tumors express FGF-2 and its endogenous antisense partner GFG. While normal pituitary expresses GFG but not FGF-2, pituitary adenomas express FGF-2 and have reduced levels of GFG; aggressive and recurrent adenomas expressed more FGF than GFG mRNA. To examine the effects of this antisense gene in the pituitary, we transfected the pituitary-derived GH4 mammosomatotroph cell line with constructs encoding the full-length human GFG cDNA. Transiently and stably transfected cells expressed the 35-kDa GFG protein that was localized to the cytoplasm. These cells exhibited enhanced PRL expression as documented by transiently transfected PRL-luciferase reporter assay and by endogenous PRL protein. GFG expression in these cells did not alter endogenous FGF-2 expression but increased the proportion of the higher molecular mass 22-kDa form of GH. Moreover, GFG expression inhibited cell proliferation as shown by [(3)H]thymidine incorporation, proliferating cell nuclear antigen (PCNA) nuclear staining, and cell cycle analysis. We conclude that the GFG-encoded protein has divergent hormone-regulatory and antiproliferative actions in the pituitary that are independent of FGF-2 expression. GFG represents a novel mechanism involved in restraining pituitary tumor cell growth while promoting hormonal activity.  相似文献

Two chitinases, A and B, were purified from the culture supernatant of Streptomyces albovinaceus S-22 by ammonium sulphate fraction (80%) and Sephadex G-200 gel filtration. Both enzymes had molecular weights estimated to be 43 and 45kDa by SDS polyacrylamide gel electrophoresis. The enzymes were active at 40°C and pH 5.6 after 120min, and stable at temperatures below 40°C in the absence of chitin. The purified enzyme had potential for cell wall lysis of fungal pathogenesis tested.  相似文献

Flavonoids are frequently found in fruits and vegetables, and are currently under investigation as potential chemopreventive agents. The present study reports the inhibitory effects of six flavonoids, quercetin, genistein, daidzein, equol, (+)-catechin and flavone, on sulphation of p-nitrophenol, a model substrate for the P-form of PST (thermostable, TS) and dopamine, a model substrate for the M-form of PST (thermolabile, TL). Kinetic studies of the PST activity and the inhibitory effects of flavonoids on the P-form of PST activity (using Hanes-Wolf and Dixon plots) show low Km and Ki values. Quercetin was found to be the most potent inhibitor and flavone was the least active inhibitor among the flavonoids. Kinetic analysis showed that the inhibition was non-competitive and Ki values were determined for each flavonoid. These observations suggest the potential for clinically important pharmacological and toxicological interactions by flavonoids.  相似文献

Propolis ethanolic extract (PEE) at 3 and 4 g/L and ultragriseofulvin (UG) at 0.75 and 1 g/L reduced the percentage of conidia germination in twoAspergillus flavus isolates. PEE at 1–4 g/L decreased the mycelial dry mass ofA. flavus isolates by 11–80%, and aflatoxin B1 production by 34–100%. UG concentrations of 0.25–1 g/L reduced the growth and aflatoxin B1 production of the isolates by 16–88 and 48–98%, respectively. Any increase in PEE and UG concentration was accompanied by a clear decrease in the per cent conidia germination, growth and aflatoxin B1 production. At equal concentration, UG was about 4-times more effective than PEE.  相似文献

Acidic dehydration of D-mannitol (1) gave a mixture of anhydrides (2) that was isopropylidenated and subsequently tritylated. A single component crystallized from the resulting mixture and was shown to be the novel 2,5-anhydro-1,3-O-isopropylidene-6-O-trityl-D-glucitol (4) by chemical and physical analysis and by comparison of its deprotected, dibenzoylated derivative (10) with authentic 2,5-anhydro-1,6-di-O-benzoyl-D-glucitol. Acid hydrolysis of 4 afforded pure 2,5-anhydro-D-glucitol (9) in better yield than by the previously reported route. The 4-O-acetyl (5), 4-O-chloro-acetyl (6), 4-O-methyl (7), and 4-O-(methylsulfonyl) (8) derivatives of 4, the tetra-O-acetyl (11) derivative of 9, and the 3,4-di-O-acetyl (12) derivative of 10, have been prepared and spectrally characterized. Complete proton-n.m.r. analysis yields first-order coupling constants that indicate the E1 (D) conformation for the tetrahydrofuran ring and the chair conformation for the 1,3-dioxane ring of 4-2-8. Obtainable coupling constants suggest that 11 and 12 exist in the oE and/or oT1, conformations.  相似文献
