Chronic inhibitory effects of two aminophenyl pyrrolidinediones on aromatase [Poster 11]

The aromatase-inhibiting effects of aminoglutethimide and of two aminophenyl-2, 5-pyrrolidinediones were studied in rats treated with pregnant mares' serum gonadotropin. All three compounds significantly inhibited ovarian aromatase activity and reduced plasma estradiol concentrations after 5 days of treatment. One of the aminophenyl pyrrolidinediones was less potent than the other two compounds, the IC₅₀ values for inhibition of the aromatization of testosterone being 19.6 μM for aminoglutethimide and 21 and 63.1 μM for the aminophenyl pyrrolidinediones.     

Infective stages of Leishmania in the sandfly vector and some observations on the mechanism of transmission

Infective stages of Leishmania (Leishmania) amazonensis, capable of producing amastigote infections in hamster skin, were shown to be present in the experimentally infected sandfly vector Lutzomyia flaviscutellata 15, 25, 40, 49, 70, 96 and 120 hours after the flies had received their infective blood-meal. Similarly, infective stages of Leishmania (L.) chagasi were demonstrated in the experimentally infected vector Lu. longipalpis examined 38, 50, 63, 87, 110, 135, 171 and 221 hours following the infective blood-meal, by the intraperitoneal inoculation of the flagellates into hamsters. The question of whether or not transmission by the bite of the sandfly is dependent on the presence of "metacyclic" promastigotes in the mouthparts of the vector is discussed.     

Host zinc metabolism and the Ehrlich ascites tumour. Zinc redistribution during tumour-related stress.

Zinc redistribution between plasma and liver has been examined in mice injected with Ehrlich-ascites-tumour cells. Within 24 h of injection plasma Zn levels decrease and Zn appears in newly synthesized liver metallothionein. This response is dependent upon the number of tumour cells injected into the host. Uptake of Zn into liver and its specific accumulation in a Zn-binding protein, identified as metallothionein, continues for a number of days and reaches a plateau as tumour growth ceases. Over this time period, plasma copper rises. This redistribution also occurs in mice pretreated with cadmium in their drinking water for 1 month at levels of 20, 50, and 100 micrograms/ml. However, in each case there is a lag of 3 days before Zn increases in the livers of these animals which already contain substantial amounts of Cd/Zn-metallothionein. When Ehrlich cells are injected into mice previously placed on a Zn-deficient diet for several days, plasma Zn is already low and no net uptake of Zn into liver metallothionein is apparent. Finally, it is shown that ascites fluid can itself stimulate a transient shift of host of Zn into liver. Heat-inactivated fluid loses this property. It is suggested that, in the peritoneum, tumour cells initiate a stress response mediated by an ascites-fluid factor.     

Associative recall properties of the trion model of cortical organization

We developed a cooperative model of the cortical column incorporating an idealized subunit, the trion (which represents a localized group of neurons), and a discrete time step for firing. We found that networks composed of a small number of trions (with symmetric interactions) supported up to thousands of quasi-stable, periodic firing patterns (MPs) which could be selected out with only small changes in interaction strengths using a Hebb-type algorithm. Here we report a study of the associative recall properties showing striking features: By considering all possible initial firing patterns (for a given set of network connections), we find 1) It takes on the average only 2–5 time steps to recall an MP. 2) Many of the MPs can be individually accessed by thousands of different initial patterns. The variety of examples presented illustrate the rich, general nature of the model.     

Ecological life histories of the three aquatic nuisance plants,Myriophyllum spicatum,Potamogeton crispus and Elodea canadensis

The life histories of Myriophyllum spicatum L., Elodea canadensis Michx., and Potamogeton crispus L., serious aquatic nuisances in many regions of the world, are reviewed to provide insights into the life style of successful aquatic nuisance plants. Specifically, their distribution and spread in North America; their life cycle, productive and reproductive potential; and their ecosystem relationships are reviewed. Hopefully this review will improve a manager's ability to deal with aquatic nuisance problems. It also provides suggestions for basic research needed to develop more effective management practices.It was found that all three species possess a number of adaptations, including an ability to rapidly propagate vegetatively, an opportunistic nature for obtaining nutrients, a life cycle that favors cool weather, and a number of mechanisms which enhance photosynthetic efficiency, which allow them to proliferate.These three species do provide benefits to the ecosystem through their roles in materials cycling and energy flow. Therefore, management of these species should take an integrated approach which recognizes these benefits.The life history information available about the three species varies tremendously; however, a better understanding of resource gain and allocation is needed to manage all three species. Specifically, more research is needed to provide a better understanding of: 1) the role bicarbonate plays in photosynthesis, 2) the role roots play in supplying CO₂ to the plabts, 3) resource accumulation and allocation under different temperature and light regimes, 4) resource allocation on a seasonal basis, and 5) nutrient cycling under different management regimes.     

An improved procedure for the preparation of alkyl sulphate esters suitable for the study of secondary alkylsulphohydrolase enzymes.

A simple, rapid and convenient method for the synthesis of secondary alkyl sulphate esters is described in which the sodium alkoxide of the parent alcohol is sulphated by using triethylamine-SO3 complex. The procedure gives relatively good yields, even for the sulphation of long-chain alcohols and those in which the hydroxy group is remote from the terminal carbon atoms. Positional isomerization, arising from the migration of the hydroxy group along the carbon chain, is absent, and resolved enantiomers of alcohols react with complete retention of configuration.     

R Factors in Hospital Infection

In order to assess the clinical importance of R factors 524 “coliform” infections were studied in a general hospital. Of these, 95 were caused by Pseudomonas aeruginosa. Among the remaining 429, 43% were caused by drug-resistant enterobacteria; resistance was frequently multiple and determined by transmissible R factors. Choice of therapy was thus limited but in every case at least one antibiotic, effective in vitro, was still available.
