A human skull with mandible from the Ngada District on the island of Flores, Indonesia, is described in order to contribute to the knowledge of variation in cranial architecture, which is important in interpretations of evolutionary cerebralisation. The skull was excavated in 1924 and sent to the National Museum in Copenhagen. The “Copenhagen Flores” (CF) male skull is radiocarbon-dated and of modern age. The cranium is small, but larger than e.g. Liang Bua skull (LB1) in every measurement. The (CT-scan based) cranial capacity of 1258 ml is normal for modern humans, but somewhat lower than values from the middle or upper Palaeolithics. The metric cranial data analysed in FORDISC, characterize the skull as a male Vietnamese rather than a Chinese or White individual. Tooth morphology shows the sundadont pattern and tooth size corresponds to that of teeth from Bali, Java and Malayan Orang Asli. Remarkable are the marked asymmetries in the dentition with rotation of an upper premolar and congenital absence of a third molar. In these respects the CF skull is similar to dentitions belonging to the pygmoid villagers of Rampasasa, a village not far from the Liang Bua cave, and to LB1. 相似文献

Elevated levels of inorganic nitrogen (N) deposition and earthworm invasion have the potential to alter N dynamics in eastern North American temperate forests. A regional comparison was conducted across 21 sugar maple (Acer saccharum Marsh) stands in southern Ontario, where forest floor C:N ratios ranged from 17 to 38 showed that, similar to many other studies, rates of potential net mineralization and nitrification increased below a forest floor C:N ratio threshold of approximately 25 and that nitrification rates are positively correlated with foliar N concentration. However, detailed measurements at four representative stands, receiving between 9.8 and 19 kg N ha?1 y?1 in throughfall, showed that foliar N levels were highest at the site with the lowest N deposition. The primary difference amongst these sites was the presence of invasive earthworms. Specifically, sites without earthworms had significantly higher forest floor N with a lower C:N ratio than the sites with earthworms. There was no significant difference in the rate of sugar maple litter decomposition or chemistry amongst the sites assessed after 540 days using fine (2-mm mesh) litter bags, suggesting that differences in forest floor N levels were most likely due to consumption of litter by large earthworm species and that the lower C:N ratio of the forest floor in sites without earthworms is brought about primarily by a much longer residence time. This work supports the conclusions that forest floor N concentration (or C:N ratio) has a very strong control on N dynamics in forests, but shows that the presence of earthworms can have an impact on forest floor C:N ratio and hence N dynamics that is greater than current levels of atmospheric inorganic N deposition in temperate forests of Ontario. 相似文献

In his considerations of “organs of extreme perfection,” Charles Darwin described the evidence that would be necessary to
support the evolutionary origin of the eye, namely, demonstration of the existence of “numerous gradations” from the most
primitive eye to the most perfect one, where each such tiny change had provided a survival advantage (however slight) to the
organism possessing the subtly altered form. In this paper, we discuss evidence indicating that the vertebrate eye did indeed
evolve through numerous subtle changes. The great majority of the gradual transitions that did occur have not been preserved
to the present time, either in the fossil record or in extant species; yet clear evidence of their occurrence remains. We
discuss the remarkable “eye” of the hagfish, which has features intermediate between a simple light detector and an image-forming
camera-like eye and which may represent a step in the evolution of our eye that can now be studied by modern methods. We also
describe the important clues to the evolutionary origin of the vertebrate eye that can be found by studying the embryological
development of our own eye, by examining the molecular genetic record preserved in our own genes and in the genes of other
vertebrates, and through consideration of the imperfections (or evolutionary “scars”) in the construction of our eye. Taking
these findings together, it is possible to discuss in some detail how the vertebrate eye evolved. 相似文献

Summary The Carboniferous, particularly during the Serpukhovian and Bashkirian time, was a period of scarce shallow-water calcimicrobial-microbialite
reef growth. Organic frameworks developed on high-rising platforms are, however, recorded in the Precaspian Basin subsurface,
Kazakhstan, Russia, Japan and Spain and represent uncommon occurrences within the general trend of low accumulation rates
and scarcity of shallow-water reefs. Sierra del Cuera (Cantabrian Mountains, N Spain) is a well-exposed high-rising carbonate
platform of Late Carboniferous (Bashkirian-Moscovian) age with a microbial boundstone-dominated slope dipping from 20° up
to 45°. Kilometer-scale continuous exposures allow the detailed documentation of slope geometry and lithofacies spatial distribution.
This study aims to develop a depositional model of steep-margined Late Paleozoic platforms built by microbial carbonates and
to contribute to the understanding of the controlling factors on lithofacies characteristics, stacking patterns, accumulation
rates and evolution of the depositional architecture of systems, which differ from light-dependent coralgal platform margins.
From the platform break to depths of nearly 300 m, the slope is dominated by massive cement-rich boundstone, which accumulated
through the biologically induced precipitation of micrite. Boundstone facies (type A) with peloidal carbonate mud, fenestellid
and fistuliporid bryozoans, sponge-like molds and primary cavities filled by radiaxial fibrous cement occurs all over the
slope but dominates the deeper settings. Type B boundstone consists of globose centimeter-scale laminated accretionary structures,
which commonly host botryoidal cement in growth cavities. The laminae nucleate around fenestellid bryozoans, sponges, Renalcis and Girvanella-like filaments. Type B boundstone typically occurs at depths between 20–150 m to locally more than 300 m and forms the bulk
of the Bashkirian prograding slope. The uppermost slope boundstone (type C; between 0 and 20–100 m depth) includes peloidal
micrite, radiaxial fibrous cement, bryozoans, sponge molds, Donezella, Renalcis, Girvanella, Ortonella, calcareous algae and calcitornellid foraminifers.
From depths of 80–200 m to 450 m, 1–30 m thick lenses of crinoidal packstone, spiculitic wackestone, and bryozoan biocementstone
with red-stained micrite matrix are episodically intercalated with boundstone and breccias. These layers increase in number
from the uppermost Bashkirian to the Moscovian in parallel with the change from a rapidly prograding to an aggrading architecture.
The red-stained strata share comparable features with Lower Carboniferous deeper-water mud-mound facies and were deposited
during relative rises of sea level and pauses in boundstone production. Rapid relative sea-level rises might have been associated
with changes in oceanographic conditions not favourable for thecalcimicrobial boundstone growth, such as upwelling of colder,
nutrient-rich waters lifting the thermocline to depths of 80–200 m.
Downslope of 150–300 m, boundstones interfinger with layers of matrix-free breccias, lenses of matrix-rich breccias, platform-
and slope-derived grainstone and crinoidal packstone. Clast-supported breccias bound by radiaxial cement are produced by rock
falls and avalanches coeval to boundstone growth. Matrix-rich breccias are debris flow deposits triggered by the accumulation
of red-stained layers. Debris flows develop following the relative sea-level rises, which favour the deposition of micrite-rich
lithofacies on the slope rather than being related to relative sea-level falls and subaerial exposures. The steep slope angles
are the result of in situ growth and rapid stabilization by marine cement in the uppermost part, passing into a detrital talus, which rests at the
angle of repose of noncohesive material. In the Moscovian, the aggradational architecture and steeper clinoforms are the result
of increased accommodation space due to tectonic subsidence and due to a reduction of slope accumulation rates (from 240±45−605±35
m/My to 130±5 m/My). The increasing number of red-stained layers and the decrease of boundstone productivity are attributed
to environmental changes in the adjacent basin, in particular during relative rises of sea level and to possible cooling due
to icehouse conditions. The geometry of the depositional system appears to be controlled by boundstone growth rates. During
the Bashkirian, the boundstone growth potential is at least 10 times greater than average values for ancient carbonate systems.
The slope progradation rates (nearly 400–1000 m/My) are similar to the highest values deduced for the Holocene Bahamian prograding
platform margin. The fundamental differences with modern systems are that progradation of the microbial-boundstone dominated
steep slope is primarily controlled by boundstone growth rates rather than by highstand shedding from the platform top and
that boundstone growth is largely independent from light and controlled by the physicochemical characteristics of seawater. 相似文献

We generated and characterized novel antibody-cytokine fusion proteins (“immunocytokines”) based on murine interleukin-7 (IL7), an immunomodulatory protein which has previously shown anti-cancer activity in preclinical models and whose human counterpart is currently being investigated in clinical trials. The sequential fusion of the clinical-stage antibody fragment scFv(F8), specific to a tumor-associated splice isoform of fibronectin, yielded an immunocytokine (termed “F8-mIL7”) of insufficient pharmaceutical quality and in vivo tumor targeting performance, with a striking dose dependence on tumor targeting selectivity. By contrast, a novel immunocytokine design (termed “F8-mIL7-F8”), in which two scFv moieties were fused at the N- and C-terminus of murine IL7, yielded a protein of excellent pharmaceutical quality and with improved tumor-targeting performance [tumor: blood ratio = 16:1, 24 h after injection]. Both F8-mIL7 and F8-mIL7-F8 could induce tumor growth retardation in immunocompetent mice, but were not able to eradicate F9 tumors. The combination of F8-mIL7-F8 with paclitaxel led to improved therapeutic results, which were significantly better compared to those obtained with saline treatment. The study indicates how the engineering of novel immunocytokine formats may help generate fusion proteins of acceptable pharmaceutical quality, for those immunomodulatory proteins which do not lend themselves to a direct fusion with antibody fragments. 相似文献

The previously described Bacillus subtilis reporter strain BAU-102 is capable of detecting cell wall synthesis inhibitors that act at all stages of the cell wall synthesis pathway. In addition, this strain is capable of detecting compounds with hydrophobic/surfactant activity and alternative mechanisms of cell wall disruption. BAU-102 sequesters preformed beta-gal in the periplasm, suggesting leakage of beta-gal as the means by which this assay detects compound activities. A model is proposed according to which beta-gal release by BAU-102 reflects activation of pathways leading to autolysis. The authors also report a simplified high-throughput assay using BAU-102 combined with the fluorogenic substrate N-methylumbelliferyl-beta-D-galactoside as a single reagent. Cell wall inhibitors release beta-gal consistently only after 60 min of incubation, whereas compounds with surfactant activity show an almost immediate release. A high-throughput screen of a 480-compound library of known bioactives yielded 8 compounds that cause beta-gal release. These results validate the BAU-102 assay as an effective tool in antimicrobial drug discovery. 相似文献
