Frequent occurrences of coral bleaching and the ensuing damage to coral reefs have generated interest in documenting stress
responses that precede bleaching. The objective of this study was to assess and compare physiological changes in healthy,
semi-bleached and totally bleached colonies of two coral species, Porites lutea and Acropora formosa, during a natural bleaching event in the Lakshadweep Archipelago in the Arabian Sea to determine the traits that will be
useful in the diagnosis of coral health. In April 2002, three “health conditions” were observed as “appearing healthy,” “semi-bleached”
and “bleached” specimens for two dominant and co-occurring coral species in these islands. Changes in the pigment composition,
zooxanthellae density (ZD), mitotic index (MI) of zooxanthellae, RNA/DNA ratios and protein profile in the two coral species
showing different levels of bleaching in the field were compared to address the hypothesis of no difference in health condition
between species and bleaching status. The loss in chlorophyll (chl) a, chl c and ZD in the transitional stage of semi-bleaching in the branched coral A. formosa was 80, 75 and 80%, respectively. The losses were much less in the massive coral P. lutea, being 20, 50 and 25%, respectively. The decrease in zooxanthellar density and chl a was accompanied by an increased MI of zooxanthellae and RNA/DNA ratios in both the species. There was an increase in accumulation
of lipofuscin granules in partially bleached P. lutea tissue, which is an indication of cellular senescence. Multivariate statistical analyses showed that colonies of P. lutea ranked in different health conditions differed significantly in chl a, chl c, ZD, RNA/DNA ratios, and protein concentrations, whereas in A. formosa chl a, chl c, chl a/c, phaeopigments and MI contributed to the variance between health conditions. 相似文献
The purpose of present study was to evaluate commercial preparations of carbamazepine tablets with respect to drug release
through a defined sequence of experiments using Minitab software. The compliance of products with respect to United States
Pharmacopeia (USP) dissolution test and comparison of the products with respect to drug release in different dissolution conditions
is reported in the present paper. The different dissolution conditions studied include dissolution medium (1% SLS in purified
water, 0.1 N HCl), volume (900 and 1,000 ml), rpm (50 rpm, 75 rpm). Studies indicated that all six products complied with
USP dissolution criteria. However, the extent of influence of dissolution conditions on drug release was varied among the
products. Distinct dissolution profiles were observed and there was no correlation with disintegration time in certain products.
The in vitro dissolution experimentation helped in identifying the discriminatory dissolution conditions and also the formulations that
were unaffected with change of dissolution variables. In summary, commercial preparations of carbamazepine vary widely in
their dissolution behavior in multi dissolution run experimentation. Identifying this behavior of the products was essential
as an in vitro tool for screening a good and a bad formulation. 相似文献
Endophytes have proven to be an invaluable resource of chemically diverse secondary metabolites that act as excellent lead compounds for anticancer drug discovery. Here we report the promising cytotoxic effects of Cladosporol A (HPLC purified >98%) isolated from endophytic fungus Cladosporium cladosporioides collected from Datura innoxia. Cladosporol A was subjected to in vitro cytotoxicity assay against NCI60 panel of human cancer cells using MTT assay. We further investigated the molecular mechanism(s) of Cladosporol A induced cell death in human breast (MCF-7) cancer cells. Mechanistically early events of cell death were studied using DAPI, Annexin V-FITC staining assay. Furthermore, immunofluorescence studies were carried to see the involvement of intrinsic pathway leading to mitochondrial dysfunction, cytochrome c release, Bax/Bcl-2 regulation and flowcytometrically measured membrane potential loss of mitochondria in human breast (MCF-7) cancer cells after Cladosporol A treatment. The interplay between apoptosis and autophagy was studied by microtubule dynamics, expression of pro-apoptotic protein p21 and autophagic markers monodansylcadaverine staining and LC3b expression.
Results
Among NCI60 human cancer cell line panel Cladosporol A showed least IC50 value against human breast (MCF-7) cancer cells. The early events of apoptosis were characterized by phosphatidylserine exposure. It disrupts microtubule dynamics and also induces expression of pro-apoptotic protein p21. Moreover treatment of Cladosporol A significantly induced MMP loss, release of cytochrome c, Bcl-2 down regulation, Bax upregulation as well as increased monodansylcadaverine (MDC) staining and leads to LC3-I to LC3-II conversion.
Conclusion
Our experimental data suggests that Cladosporol A depolymerize microtubules, sensitize programmed cell death via ROS mediated autophagic flux leading to mitophagic cell death.

The majority of the flavours and fragrances used worldwide are produced by chemical synthesis at low price. However, consumers prefer natural compounds because of increasing health and nutrition awareness in routine life. Hence, biotransformation is an alternative process to produce natural aroma compounds. Microorganisms have been gradually used more to produce natural aroma compounds with various applications in food, agriculture and pharmaceutical industries. This paper reviews the role of microorganisms in the transformation of ferulic acid to 4-vinyl guaiacol. The microbial processes based on biocatalytic method are discussed in terms of their advantages over chemical synthesis, plant cell cultures and enzyme catalyzed reactions. Thus, the transformation of ferulic acid by microorganisms could have possible use in food, pharmaceutical industry and become an increasingly important platform for the production of natural aroma compounds. 相似文献
Neuroendocrine (NE) differentiation has gained increased attention as a prostate cancer (PC) prognostic marker. The aim of this study is to determine whether host germline genetic variation influences tumor progression and metastasis in C57BL/6-Tg(TRAMP)8247Ng/J (TRAMP) mouse model of aggressive NEPC. TRAMP mice were crossed to the eight progenitor strains of the Collaborative Cross recombinant inbred panel to address this. Tumor growth and metastasis burden were quantified in heterozygous transgene positive F1 male mice at 30 weeks of age. Compared to wild-type C57BL/6J-Tg(TRAMP)824Ng/J males, TRAMP x CAST/EiJ, TRAMP x NOD/ShiLtJ and TRAMP x NZO/HlLtJ F1 males displayed significant increases in tumor growth. Conversely, TRAMP x WSB/EiJ and TRAMP x PWK/PhJ F1 males displayed significant reductions in tumor growth. Interestingly, despite reduced tumor burden, TRAMP x WSB/EiJ males had an increased nodal metastasis burden. Patterns of distant pulmonary metastasis tended to follow the same patterns as that of local dissemination in each of the strains. All tumors and metastases displayed positive staining for NE markers, synaptophysin, and FOXA2. These experiments conclusively demonstrate that the introduction of germline variation by breeding modulates tumor growth, local metastasis burden, and distant metastasis frequency in this model of NEPC. These strains will be useful as model systems to facilitate the identification of germline modifier genes that promote the development of aggressive forms of PC. 相似文献

Cryptosporidium parvum is the second leading cause of persistent diarrhea among children in low-resource settings. This study examined the effect of oregano essential oil (OEO) and carvacrol (CV) on inhibition of C. parvum infectivity in vitro. HCT-8 cells were seeded (1 × 106) in 96-well microtiter plates until confluency. Cell viability and infectivity were assessed by seeding HCT-8 cell monolayers with C. parvum oocysts (1 × 104) in two modalities: 1) 4 h co-culture with bioactive (0–250 μg/mL) followed by washing and incubation (48 h, 37 °C, 5% CO2) in bioactive-free media; and 2) 4 h co-culture of C. parvum oocysts followed by washing and treatment with bioactive (0–250 μg/mL) during 48-h incubation. Cell viability was tested using Live/Dead? assay whereas infectivity was measured using C. parvum-specific antibody staining via immunofluorescence detection. Loss of cell viability was observed starting at 125 μg/mL and 60 μg/mL for OEO and CV, respectively. Neither OEO nor CV modulated the invasion of C. parvum sporozoites in HCT-8 cells. Treatment with bioactive after invasion reduced relative C. parvum infectivity in a dose-dependent manner to 55.6 ± 10.4% and 45.8 ± 4.1% at 60 and 30 μg/mL of OEO and CV, respectively. OEO and CV are potential bioactives to counteract C. parvum infection in children. 相似文献
