Ultra-violet induced mutations to growth factor requirement and penicillin resistance in a blue-green alga

Summary After exposures to UV, two mutant strains of Phormidium mucicola were isolated which are stable and inheritable:1. Strain 5/5 mL-1 is a photoheterotrophic mutant requiring addition of an organic growth factor to the basal medium which appears fulfilled by either casein hydrolysate, glucose, acetate or vitamin mixture and fails to grow heterotrophically.2. mr/p. This strain shows resistance upto 10.24 g/ml of penicillin whereas the parent does not survive beyond a penicillin dose of 0.1 g/ml.     

Electron microscopic studies on liver 5-aminolaevulinate synthase

The structure of chick embryo liver 5-aminolaevulinate synthase has been examined by electron microscopic studies using negative staining. From the different projections of the enzyme particles observed in electron micrographs, a model for the enzyme molecule has been proposed. In this model, an enzyme molecule consists of two curved and identical subunits associated in opposite polarities. From the dimensions of an enzyme molecule subunit measured from electron micrographs, the relative molecular mass of each subunit is estimated to be 70 000.     

Isolation and characterization of a unique protease from sporulating cells of Bacillus subtilis

Two proteases, designated I and II, have been isolated from sporulating cells of Bacillus subtilis. They were partially purified by ammonium sulfate fractionation, Sephadex chromatography and affinity columns. Protease I was found to be similar to an already characterized B. subtilis protease. Protease II is trypsin-like in its substrate specificity and is distinct from protease I in its pH optimum, pH stability, molecular weight, substrate specificity, heat stability and sensitivity to various inhibitors. While both enzymes were produced primarily during sporulation, they attained maximum levels of activity at different times. Distinct functions for these proteases in post exponential B. subtilis are likely.     

Extractive lactic acid fermentation using ion-exchange resin

Lactic acid fermentation is an end-product-inhibited reaction. The restriction imposed by lactic acid on its fermentation can be avoided by extractive fermentation techniques. Studies were performed by attaching an ion-exchange resin packed column with a 2-L fermentor for separation of lactic acid. The fermentation, in a conventional batch mode, resulted in a lactic acid yield of 0.828 g . g(-1) and a lactic acid productivity of 0.313 g . L(-1) . h(-1). However, these could be further enhanced to 0.929 g . g(-1) and 1.665 g . L(-1) . h(-1) by extractive fermentation techniques. The effect of temperature on extractive fermentation was remarkable and has been included in this work.     

The fine structure of psychrophilic and mesophilic Rhodotorula rubra

Transmission electron microscopy of thin sections of two mesophilic and a psychrophilic Rhodotorula rubra revealed structures normally associated with yeast. The capsule of all three strains was thicker in the cells grown in glucose than those grown in glucose-free media. The cell wall of all three strains showed more lamellae in the cells grown in a medium containing glycerol. Budding was of the Rhodoturula type. Endoplasmic reticula running parallel to the nucleus were commonly observed in the psychrophiles but not in the mesophiles. In the psychrophile, in association with the plasmalemma, lomasomes and plasmalemmasomes were observed. Giant mitochondria were commonly seen in the cells grown in a fermentable carbohydrate-free medium. Vacuoles were mostly empty.
